Approximately 950 individual sequences of nonribosomally biosynthesised peptides are produced by the genus Trichoderma/Hypocreathat belong to a perpetually growing class of mostly linear antibiotic oligopeptides,which are rich in the non-proteinogenic α-aminoisobutyric acid(Aib).Thus,they are comprehensively named ***,peptaibiotics represent ca.80% of the total inventory of secondarymetabolites currently known from Trichoderma/*** unique membrane-modifying bioactivity results from amphipathicity and helicity,thus making them ideal candidates in assisting both colonisation and defence of the natural habitats by their fungal *** this,reports on the in vivo-detection of peptaibiotics have scarcely been published in the *** order to evaluate the significance of peptaibiotic production for a broader range of potential producers,we screened nine specimens belonging to seven hitherto uninvestigated fungicolous or saprotrophic Trichoderma/Hypocrea species by liquid chromatography coupled to electrospray high resolution mass *** of peptaibiotics found were independently confirmed by analysing the peptaibiome of pure agar cultures obtained by single-ascospore isolation from the *** the nine species examined,five were screened positive for peptaibiotics.A total of 78 peptaibiotics were sequenced,56(=72%)of which are ***,dihydroxyphenylalaninol and O-prenylated tyrosinol,two C-terminal residues,which have not been reported for peptaibiotics before,were found as well as new and recurrent sequences carrying the recently described tyrosinol residue at their *** majority of peptaibiotics sequenced are 18-or 19-residue *** homologies with‘classical representatives’of subfamily 1(SF1)-peptaibiotics argue for the formation of transmembrane ion channels,which are prone to facilitate the producer capture and defence of its substratum.

目的　了解Kaposi肉瘤与人疱疹病毒 8(HHV8)或Kaposi肉瘤相关病毒的关系 ;建立适合于临床病理诊断的原位杂交方法。方法　采用地高辛标记的单链HHV8DNA探针 ,辅以增强探测敏感性的信号扩增系统 (tyramidesignalamplification)对 14例甲醛固定石蜡包埋的Kaposi肉瘤组织标本进行了HHV8原位检测 ,为了比较 ,同时也采用了聚合酶链反应 (PCR)检测技术。结果　用原位杂交和PCR两种方法共发现 10例 (71% )Kaposi肉瘤组织中存在HHV8感染。其中 8例在两种技术检测中均呈阳性。HHV8阳性信号见于肿瘤梭形细胞和内皮细胞 ,也见于各期病变 (早中期的斑片和斑块病变 ,晚期的结节病变 ) ,以及原发 ,复发和 (或 )转移病变。所有阴性对照在两种方法中均呈阴性。结论　Kaposi肉瘤与HHV8存在密切关系 ;这种地高辛标记单链DNA探针。

Long-term rice-rice-green manure rotation changing the microbial communities in typical red paddy soil in South China

On the basis of a long-term（30 years） field experiment that involved four rotation systems, rice-rice-winter fallow（RRF）, rice-rice-ryegrass（RRG）, rice-rice-rape（RRP）, and rice-rice-milk vetch（RRV）, this study described the effects of green manure on the microbial communities in the red paddy soils using 454 pyrosequencing for the 16 S r RNA gene. The Chao1 richness and non-parametric Shannon＇s index increased in all soil samples that received green manure treatments. The communities＇ structures with the green manure applications were significantly dissimilar from that under the winter fallow. Using Metastats tests, many genera in the RRG, RRP and RRV soils were significantly different from those in the RRF soil, including a number of genera that functioned in the nitrogen and sulfur cycles. Analyses of the genera with these functions revealed the shifts in microbial ecosystem functions after long-term green manuring. Changes in the microbial communities increased the ammonium supply and decreased the soil acidification in green-manure-amended soils. Together, these data suggested powerful effects of green manure on both the microbial communities and the biogeochemical cycle driven by the shifts in bacterial functional groups.

Is there a gender difference in antidiuretic response to desmopressin in children?

Women with nocturia are more sensitive to desmopressin, a synthetic arginine vasopressin (AVP) analogue, with significant antidiuretic responses to desmopressin orally disintegrating tablet (ODT) 25 μg, compared with men who require 58 μg to achieve similar responses. In children the current desmopressin dose recommendation to treat primary nocturnal enuresis (PNE) is the same for boys and girls. This post hoc analysis of data from a randomised, doubleblind single-dose study of 84 children with PNE aged 6 - 12 years explored gender differences in sensitivity to desmopressin in children. Following water loading to suppress endogenous AVP, placebo or desmopres-sin 30, 60, 120, 240, 360 or 480 μg was administered when urinary production reached >0.13 mL/min/kg. The endpoints of urinary osmolality and duration of urinary-concentrating action (DOA) (above three thresholds: 125, 200 and 400 mOsm/kg) were analysed to compare efficacy in boys and girls, in each treatment group. The DOA and urinary osmolality were similar in both genders in the desmopressin 120 - 480 μg groups. Boys receiving desmopressin ODT 30 - 60 μg tended to increased urinary osmolality and experienced 1 - 2 hours longer DOA than girls. The same pattern of higher values in boys compared with girls was seen for all measures of urinary osmolality. Conclusion: In a limited sample of pre-pubertal children the antidiuretic response to desmopressin was largely similar between genders, in contrast to findings in adults.

Fermentative Bioethanol Production Using Enzymatically Hydrolysed <i>Saccharina latissima</i>

The increased demand for machinery and transport has led to an overwhelming increase in the use of fossil fuels in the last century. Concerning the economic and environmental concern, macroalgae with high fermentable polysaccharide content (mainly mannitol, cellulose and laminarin), can serve as an excellent alternative to food crops for bioethanol production, a renewable liquid fuel. In this study, Saccharina latissima, a brown macroalgae readily available on the Norwegian coast was used as the carbohydrate source for the fermentative production of bioethanol. The macroalgae harvested was found to contain 31.31 ± 1.73 g of reducing sugars per 100 g of dry Saccharina latissima upon enzymatic hydrolysis. The subsequent fermentation with Saccharomyces cerevisiae produced an ethanol yield of 0.42 g of ethanol per g of reducing sugar, resulting in a fermentation efficiency of 84% as compared to the theoretical maximum. Using these results, an evaluation of the fermentation process has demonstrated that the brown macroalgae Saccharina latissima could become a viable bioethanol source in the future.

Modeling Preparative Chromatographic Separation of Heavy Rare Earth Elements and Optimization of Thulium Purification

Rare Earth Elements are in growing demand globally. This paper presents a case study of applied mathematical modeling and multi objective optimization to optimize the separation of heavy Rare Earth Elements, Terbium-Lutetium, by means of preparative solid phase extraction chromatography, which means that an extraction ligand, HDEHP, is immobilized on a C18 silica phase, and nitric acid is used as an eluent. An ICP-MS was used for online detection of the Rare Earths. A methodology for calibration and optimization is presented, and applied to an industrially relevant mixture. Results show that Thulium is produced at 99% purity, with a productivity of 0.2 - 0.5 kg Tm per m3 stationary phase and second, with Yields from 74% to 99%.
