Korean red ginseng decreases 1-methyl-4-phenylpyridinium-induced mitophagy in SH-SY5Y cells

Objective:Mitophagy is known to contribute towards progression of Parkinson’s *** red ginseng(KRG)is a widely used medicinal herb in East Asia,and recent studies have reported that KRG prevents 1-methyl-4-phenylpyridinium ion(MPP^(+))-induced cell *** study was undertaken to investigate whether KRG suppresses MPP^(+)-induced apoptosis and ***:SH-SY5 Y cells were incubated with KRG for 24 h,and subsequently exposed to MPP^(+).The MPP^(+)-induced cell death was confirmed with the 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay,and the terminal deoxynucleotidyl transferase-mediated d UTP nick end-labeling *** in the structure and function of mitochondria were confirmed using mitotracker,Mito SOX red mitochondrial superoxide indicator,parkin,and phosphatase and tensin homolog deleted on chromosome ten-induced putative kinase 1(PINK1)immunofluorescent *** blotting was performed to evaluate the expression of apoptosis-related factors in whole cells,including Bax,Bcl-2 and cleaved caspase-3,and mitophagy-related factors in the mitochondrial fraction,including cytochrome c,parkin,PINK1,translocase of the outer membrane 20(TOM20),p62 and Beclin ***:MPP^(+)induced cell death by cytochrome c release and caspase-3 activation;however,this effect was suppressed by KRG’s regulation of the expressions of Bcl-2 and ***,MPP^(+)exposure increased the mitochondrial expressions of parkin,PINK1,Beclin 1 and p62,and decreased TOM20,cytochrome c and Bcl-2 *** MPP^(+)-induced changes in the mitochondrial fraction were attenuated by treatment with ***:KRG effectively prevents MPP^(+)-induced SH-SY5 Y cell death by regulating cytochrome c release from mitochondria and PINK1/parkin-mediated mitophagy,through regulation of the Bcl-2 family.

The technique of region of interest-based positron emission tomography is limited by its poor reli-ability and relatively few examined brain regions. In the present study, we compared brain metabo-lism assessed using fluorine-18-fluorodeoxyglucose positron emission tomography between 14 at-tention-deficit hyperactivity disorder （ADHD） patients and 15 normal controls with scoliosis at resting state by statistical parametric mapping. Glucose metabolism was decreased in the left parahippo-campal gyrus, left hippocampus, left anterior cingulate gyrus, right anterior and posterior lobes of the cerebellum, left superior temporal gyrus, left insula, left medial and middle frontal gyri, right medial frontal gyrus, and left basal ganglia （putamen, amygdala, and caudate nucleus） in children with ADHD. These data suggest that children with ADHD exhibit hypometabolism in various brain regions compared to controls, indicating that ADHD symptoms are unlikely the result of abnormalities in specific areas.

Advanced heterolytic H_(2) adsorption of K-added Ru/MgO catalysts for accelerating hydrogen storage into aromatic benzyltoluenes

Herein,we report a highly active K-added Ru/MgO catalyst for hydrogen storage into aromatic benzyltoluenes at low temperatures to advance liquid organic hydrogen carrier *** hydrogenation activity of Ru/K/MgO catalysts exhibits a volcano-shaped dependence on the K content at the maximum with 0.02 wt%.This is in good agreement with the strength and capacity of H_(2) adsorption derived from basicity,despite a gradual decrease in the textural property and the corresponding increase in the Ru particle size with increasing the K *** functional theory calculations show that heterolytic hydrogen adsorption properties(strength and polarization)are facilitated up to a specific density of K on the Ru–MgO interface and excessive K suppresses heterolytic H_(2) adsorption by direct interaction between K and hydrogen,assuring the hydrogenation activity and H_(2) adsorption capability of Ru/K/MgO ***,the Ru/K/MgO catalyst,when K is added in an optimal amount,is highly effective to accelerate hydrogen storage kinetics at low temperatures owing to the enhanced heterolytic H_(2) adsorption.

Brassinosteroids enhance salicylic acid-mediated immune responses by inhibiting BIN2 phosphorylation of clade I TGA transcription factors in Arabidopsis

Salicylic acid (SA) plays an important role in plant immune response, including resistance to pathogens and systemic acquired resistance. Two major components, NONEXPRESSOR OF PATHOGENESIS-RELATED GENES (NPRs) and TGACG motif-binding transcription factors (TGAs), are known to mediate SA signaling, which might also be orchestrated by other hormonal and environmental changes. Nevertheless, the molecular and functional interactions between SA signaling components and other cellular signaling pathways remain poorly understood. Here we showed that the steroid plant hormone brassinosteroid (BR) promotes SA responses by inactivating BR-INSENSITIVE 2 (BIN2), which inhibits the redox-sensitive clade I TGAs in Arabidopsis. We found that both BR and the BIN2 inhibitor bikinin synergistically increase SA-mediated physiological responses, such as resistance to Pst DC3000. Our genetic and biochemical analyses indicated that BIN2 functionally interacts with TGA1 and TGA4, but not with other TGAs. We further demonstrated that BIN2 phosphorylates Ser-202 of TGA4, resulting in the suppression of the redox-dependent interaction between TGA4 and NPR1 as well as destabilization of TGA4. Consistently, transgenic Arabidopsis overexpressing TGA4-YFP with a S202A mutation displayed enhanced SA responses compared to the wild-type TGA4-YFP plants. Taken together, these results suggest a novel crosstalk mechanism by which BR signaling coordinates the SA responses mediated by redox-sensitive clade I TGAs.

Involvement of mitochondrial Na+-Ca2+ exchange in intestinal pacemaking activity

AIM： Interstitial cells of Cajal （ICCs） are the pacemaker cells that generate slow waves in the gastrointestinal （GI） tract. We have aimed to investigate the involvement of mitochondrial Na^＋-Ca^2＋ exchange in intestinal pacemaking activity in cultured interstitial cells of Cajal. METHODS： Enzymatic digestions were used to dissociate ICCs from the small intestine of a mouse. The whole-cell patch-clamp configuration was used to record membrane currents （voltage clamp） and potentials （current clamp） from cultured ICCs. RESULTS： Clonazepam and CGP37157 inhibited the pacemaking activity of ICCs in a dose-dependent manner. Clonazepam from 20 to 60 μmol/L and CGP37157 from 10 to 30 μmol/L effectively inhibited Ca^2＋ efflux from mitochondria in pacemaking activity of ICCs. The ICsos of clonazepam and CGP37157 were 37.1 and 18.2 μmol/ L, respectively. The addition of 20 μmol/L NiCl2 to the internal solution caused a ＂wax and wane＂ phenomenon of pacemaking activity of ICCs. CONCLUSION： These results suggest that mitochondria Na^＋-Ca^2＋ exchange has an important role in intestina pacemaking activity.

Single-crystalline-level properties of ultrathin SrRuO_(3) flexible membranes with wide and clean surface

Transferring single-crystalline(SC)membranes to flexible substrates has been increasingly studied,enabling emerging functionality and enhanced performance of various devices.A commonly used support-assisted transfer process inevitably leaves dirty residue on material surfaces,limiting the further development of surface-related ***,we scale down the thickness of flexible SC SrRuO_(3)(SRO)membranes to 15 nm with a clean surface area of 2.5×2.5 mm^(2).This is accomplished by making the polyethylene terephthalate(PET)substrate surface hydrophilic via oxygen plasma treatment,thereby reducing the surface *** ultrathin,clean,wide,and flexible SC SRO membranes guarantee a high transmittance of up to 60%,a low resistivity of 10^(−4)−10^(−3)Ωcm at room temperature,and band ferromagnetism below 150 K with a high magnetic moment of~0.5μB/Ru at 10 *** SC-level properties of our SRO membranes imply their potential use in state-of-the-art platforms for next-generation electronics and energy devices.

Impact of uneven distribution of grain characteristics on yield strength and martensitic transformation of as-hot-rolled medium-entropy alloys

As-hot-rolled medium-entropy alloys(MEAs)with unevenly distributed grain sizes of face-centered cubic grains exhibit better yield strength without uniform elongation loss compared to cold-rolled and an-nealed *** operation of dynamic recrystallization(DRX)during several hot rolling passes leads to a wide range of grain sizes from submicrons to tens of micrometers due to the grain growth after nucleation:early recrystallized grains are coarser than recently recrystallized *** only the grain size but internal dislocation density of the recently recrystallized grain is *** the tensile deformation of the hot-rolled MEAs at-196 ℃,dislocation pile-ups in the relatively soft and fine DRX grains enhance yield stress and hetero-deformation-induced strain *** to the enhanced yield stress of the as-hot-rolled MEAs,stress-induced martensitic transformation easily ***,partially DRXed MEAs hot-rolled at 800 ℃ have lower yield stress than fully DRXed ones,hot-rolled at 900 and 1000 ℃.This is attributed to the softening effect of the stress-induced body-centered cubic martensitic transformation in unrecrystallized coarse grains prior to the yielding,which lowers the yield stress of the partially DRXed *** yielding,the martensitic transformation facilitates strain hard-ening and early necking is *** study presents a fresh outlook on the uneven distribution of grain sizes by hot rolling beneficial to mechanical responses of uniform elongation of~45%despite the as-rolled states with an advantage of simplified thermo-mechanical processes.

Pollen tube growth is essential for successful double fertilization, which is critical for grain yield in crop plants. Rapid alkalinization factors(RALFs) function as ligands for signal transduction during fertilization. However, functional studies on RALF in monocot plants are ***, we functionally characterized two pollen-specific RALFs in rice(Oryza sativa) using multiple clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9-induced loss-of-function mutants,peptide treatment, expression analyses, and tag reporter lines. Among the 41 RALF members in rice, OsRALF17 was specifically expressed at the highest level in pollen and pollen *** applied OsRALF17 or OsRALF19peptide inhibited pollen tube germination and elongation at high concentrations but enhanced tube elongation at low concentrations, indicating growth regulation. Double mutants of OsRALF17 and OsRALF19(ralf17/19) exhibited almost full male sterility with defects in pollen hydration, germination, and tube elongation,which was partially recovered by exogenous treatment with OsRALF17 peptide. This study revealed that two partially functionally redundant OsRALF17 and OsRALF19 bind to Oryza sativa male-gene transfer defective 2(OsMTD2) and transmit reactive oxygen species signals for pollen tube germination and integrity maintenance in rice. Transcriptomic analysis confirmed their common downstream genes, in osmtd2 and ralf17/19. This study provides new insights into the role of RALF, expanding our knowledge of the biological role of RALF in regulating rice fertilization.

Intracerebral hemorrhage （ICH） is a devastating subtype of stroke with a high mortality rate, for which there currently is no effective treatment. A perihematomal edema caused by an intense inflammatory reaction is more deleterious than the hematoma itself and can result in neurological deterioration and death. Ceria nanoparticles （CeNPs） are potent free radical scavengers with potential for biomedical applications. As oxidative stress plays a major role in post-ICH inflammation, we hypothesized that CeNPs might protect against ICH. To test this hypothesis, core CeNPs were synthesized using a modified reverse micelle method and covered with phospholipid-polyethylene glycol （PEG） to achieve biocompatibility. We investigated whether our custom-made biocompatible CeNPs have protective effects against ICH. The CeNPs reduced oxidative stress, hemin-induced cytotoxicity, and inflammation in vitro. In a rodent ICH model, intravenously administered CeNPs were mainly distributed in the hemorrhagic hemisphere, suggesting that they could diffuse through the damaged blood-brain barrier. Moreover, CeNPs attenuated microglia/macrophage recruitment around the hemorrhagic lesion and inflammatory protein expression. Finally, CeNP treatment reduced the brain edema by 68.4% as compared to the control. These results reveal the great potential of CeNPs as a novel therapeutic agent for patients with ICH.

Therapeutic DC vaccination with IL-2 as a consolidation therapy for ovarian cancer patients： a phase Ⅰ/Ⅱ trial

While ovarian cancer （OvCa） responds well to surgery and conventional chemotherapy, a high recurrence rate of advanced OvCa is observed. In this phase Ⅰ/Ⅱ study, 10 OvCa patients with minimal residual disease were treated with autologous dendritic cells （DCs） and IL-2 to evaluate the safety and feasibility of this therapeutic strategy and to characterize the antigen-specific immune alterations induced through this treatment. Approximately 4 months after initial debulking and chemotherapy, patients received two subcutaneous doses of autologous monocyte-derived DCs pulsed with autologous tumor lysate and keyhole limpet hemocyanin （KLH） at 4-week intervals. After each DC inoculation, low-dose （200 mlU） IL-2 was introduced for 14 consecutive days as an immune adjuvant. The vaccination was well tolerated. In three out of 10 patients, the inclusion status after the initial therapy showed the maintenance of complete remission （CR） after DC vaccination for 83, 80.9 and 38.2 months without disease relapse. One patient with stable disease （SD） experienced the complete disappearance of tumor after DC vaccination, and this status was maintained for 50.8 months until tumor recurrence. In two patients with partial response （PR） was not responding to DC vaccination and their disease recurred. In the three patients with disease free long-term survival, significant immune alterations were observed, including increased natural killer （NK） activity, IFN-γ-secreting T cells, immune-stimulatory cytokine secretion and reduced immune-suppressive factor secretion after DC vaccination. Thus, in patients with NED status and increased overall survival, DC vaccination induced tumor-related immunity, potentially associated with long-term clinical responses against OvCa.
