Differential bone metabolism and protein expression in mice fed a high-fat diet versus Daurian ground squirrels following natural pre-hibernation fattening

This study compared the effects on bone metabolism and morphology of pathological obesity induced by excessive fat intake in a non-hibernator(mice)versus healthy obesity due to pre-hibernation fattening in a hibernator(ground squirrels).Kunming mice were fed a high-fat diet to provide a model of pathological obesity(Ob group).Daurian ground squirrels fattened naturally in their pre-hibernation season(PRE group)were used as a healthy obesity ***-computed tomography(micro-CT)and three-point bending tests were used to determine the microstructure and mechanical properties of *** blots were used to analyze protein expression levels related to bone metabolism(Runt-related transcription factor 2(RunX2),osteocalcin(OCN),alkaline phosphatase(ALP),osteoprotegerin(OPG),receptor activator of nuclear factor-κb ligand(RANKL),cathepsin K,matrix metallopeptidase 9(MMP9),patched protein homolog 1(Ptch1),phosphorylatedβ-catenin(P-β-catenin),and glycogen synthase kinase-3β(GSK-3β)).Compared with controls,there was no obvious bone loss in the Ob mice,and the stiffness of the femur was increased *** with summer active squirrels,bone formation was enhanced but the mechanical properties did not change in the PRE group *** Ob mice,western blots showed significantly increased expression levels of all proteins except RunX2,OPG,and *** ground squirrels showed significantly increased expression of most proteins except OCN and Ptch1,which decreased significantly,and P-β-catenin and OPG,which did not *** conclusion,for non-hibernating mice,moderate obesity had a certain protective effect on bones,demonstrating two-way regulation,increasing both bone loss and bone *** pre-hibernating ground squirrels,the healthy obesity acquired before hibernation had a positive effect on the microstructure of bones,and also enhanced the expression levels of proteins related to bone formation,bone resorption,and Wnt signaling.

To the Editor： Hepatocellular carcinoma （HCC） remains one of the commonest cancers worldwide especially in hepatitis b endemic regions. Its aggressive behavior is characterised by the natural history of increasing size, a tendency for vascular invasion into the hepatic veins and portal veins. Further growth into the inferior vena cava （IVC） and right atrium （RA） is an infrequent finding but signifies a pre-terminal event with a dismal prognosis.

Antibiotic bone Cement and Ultraviolet Light Use in Total Knee Arthroplasty

Objective: The purpose of this study was to determine usefulness of antibiotic bone cement and UV light in primary Total Knee Arthroplasty (TKA). Patients and Methods: between 1986 and 2008, 3105 TKAs were performed using 750 mg of cefuroxime per bag of cement (Group 1), 7537 using cefuroxime and UV light (Group 2), and 4573 using UV light and gentamicin premixed (Group 3). Results: The primary infection rate was 0.57% (87 acute postoperative infections). Of these, 27 (0.9%) were in Group 1, 44 (0.6%) were in Group 2, and 16 (0.35%) were in Group 3 (p = 0.0013). Kaplan-Meier survival in Groups 1 - 3 for reoperation for aseptic loosening of any component at 5 years was 0.9908, 0.9927, and 0.9959, respectively (p = 0.1315). Conclusion: Despite potential concerns mentioned in the literature, these results continue to support the use of antibiotic bone cement and UV lights during TKA.

Proton-pump inhibitor use does not affect semen luality in subfertile men

Proton-pump inhibitors （PPIs） are among the most widely used drugs worldwide. PPI use has recently been linked to adverse changes in semen quality in healthy men; however, the effects of PPI use on semen parameters remain largely unknown specifically in cases with male factor infertility. We examined whether PPI use was associated with detrimental effects on semen parameters in a large population of subfertile men. We retrospectively reviewed data from 12 257 subfertile men who had visited our fertility clinic from 2003 to 2013. Patients who reported using any PPIs for 〉3 months before semen sample collection were included; 7698 subfertile men taking no medication served as controls. Data were gathered on patient age, medication use, and conventional semen parameters; patients taking any known spermatotoxic medication were excluded. Linear mixed-effect regression models were used to test the effect of PPI use on semen parameters adjusting for age. A total of 248 patients （258 samples） used PPIs for at least 3 months before semen collection. In regression models, PPI use （either as the only medication or when used in combination with other nonspermatotoxic medications） was not associated with statistically significant changes in semen parameters. To our knowledge, this is the largest study to compare PPI use with semen parameters in subfertile men. Using PPIs was not associated with detrimental effects on semen quality in this retrospective study.

Cysteine-rich secretory protein-1 （CRISP-1） is a glycoprotein secreted by the epididymal epithelium. It is a member of a large family of proteins characterized by two conserved domains and a set of 16 conserved cysteine residues. In mammals, CRISP-1 inhibits sperm-egg fusion and can suppress sperm capacitation. The molecular mechanism of action of the mammalian CRISP proteins remains unknown, but certain non-mammalian CRISP proteins can block ion channels. In the rat, CRISP-1 comprises two forms referred to as Proteins D and E. Recent work in our laboratory demonstrates that the D form of CRISP-1 associates transiently with the sperm surface, whereas the E form binds tightly. When the spermatozoa are washed, the E form of CRISP-1 persists on the sperm surface after all D form has dissociated. Cross-linking studies demonstrate different protein-protein interaction patterns for D and E, although no binding partners for either protein have yet been identified. Mass spectrometric analyses revealed a potential post-translational modification on the E form that is not present on the D form. This is the only discernable difference between Proteins D and E, and presumably is responsible for the difference in behavior of these two forms of rat CRISP- 1. These studies demonstrate that the more abundant D form interacts with spermatozoa transiently, possibly with a specific receptor on the sperm surface, consistent with a capacitation-suppressing function during sperm transit and storage in the epididymis, and also confirm a tightly bound population of the E form that could act in the female reproductive tract. （Asian J Androl 2007 July; 9： 508-514）

bACKGROUND:Controversy remains over whether the middle hepatic vein should be included in the liver graft in right liver living donor liver *** in the anterior sector of a right liver graft can cause graft malfunction,which is especially devastating in the case of a graft with marginal size in relation to recipient body size on top of poor pre-transplant recipient *** case we report here highlighted the importance of the middle hepatic vein in right liver living donor liver ***:We illustrated the rectification of outflow obstruction of the middle hepatic vein in the anterior sector of right liver graft caused by technical error during *** rectification was performed with emergency re-routing using an artificial ***:Congestion in the anterior sector of the graft improved immediately and the patient’s postoperative liver function test results improved ***:The middle hepatic vein is important for effective drainage of the anterior sector of a right liver *** re-routing technique described in the report can also be applied to cases in which the middle hepatic vein is injured during hepatectomy requiring immediate reconstruction.

Since the first set X-ray area detector system of China was installed in the National Laboratory of biomacromolecules in 1990, it has been used to collect more than twenty data sets on crystals of biomacromolecules from laboratories in China. The data sets include Rsym from 2.18% for the native insulin to 8.69% for glucose isomerase. A complete data set for a medium resolution can be collected in 24-40 h. Compared with a four-cycle diffractometer, the area detector has more obvious advantages on the speed of collection, saving crystals and quality of data. The basic principles of the area detector, the analysis of data quality and the problems of processing data for R3 space group are introduced and discussed.
