Stem cell therapy for the treatment of Leydig cell dysfunction in primary hypogonadism

The production of testosterone occurs within the Leydig cells of the testes. When production fails at this level from either congenital, acquired, or systemic disorders,the result is primary hypogonadism. While numerous testosterone formulations have been developed, none are yet fully capable of replicating the physiological patterns of testosterone secretion. Multiple stem cell therapies to restore androgenic function of the testes are under investigation. Leydig cells derived from bone marrow, adipose tissue, umbilical cord, and the testes have shown promise for future therapy for primary hypogonadism. In particular, the discovery and utilization of a group of progenitor stem cells within the testes, known as stem Leydig cells(SLCs), has led not only to a better understanding of testicular development, but of treatment as well. When combining this with an understanding of the mechanisms that lead to Leydig cell dysfunction, researchers and physicians will be able to develop stem cell therapies that target the specific step in the steroidogenic process that is deficient. The current preclinical studies highlight the complex nature of regenerating this steroidogenic process and the problems remain unresolved. In summary, there appears to be two current directions for stem cell therapy in male primary hypogonadism. The first method involves differentiating adult Leydig cells from stem cells of various origins from bone marrow, adipose, or embryonic sources. The second method involves isolating, identifying, and transplanting stem Leydig cells into testicular tissue. Theoretically, in-vivo re-activation of SLCs in men with primary hypogonadism due to age would be another alternative method to treat hypogonadism while eliminating the need for transplantation.

Experimenting the Feasibility of Banana Stalk for Practical Textile

Finding viable alternatives to conventional textiles and fashion has proved to be a herculean challenge for government and policy makers in Ghana. Banana stalk identified as possible eccentric material for textiles and fashion needs to be examined to ascertain its feasibility. The study intends to engage mixed approach with particular emphasis on experimental methods to ascertain feasibility of banana stalk braids for textiles. Also what to be adopted are observation, descriptive, interpretative, and narrative methods. Test results from the study are necessary for determining the extent to which thematerials could be applied for varying textiles and costumes. Adopting waste as alternative material in fashioning costumes for textile art and subsequently normal fashion for humans is a possible means to turn around the poorly performing the industrial sub-sector. It also has the potential of being converted into sacks for packaging some foodstuffs like grains, tubers, and cocoa. The success of this research shall pave way for looking inward the nation for readily available and relatively cheap materials for addressing the concern of local textiles and fashion.

Blood velocity and pressure fields in large arteries are greatly influenced by the deformability of the vessel. However, computational methods for modeling blood flow in three dimensional models of arteries have either considered a rigid wall assumption for the vessel [1] or significantly simplified geometries. Computing blood flow in deformable domains using s tandard techniques like the ALE method remains an intractable problem for realistic anatomic and physiologic models. We have developed a new method to model blood flow in three dimensional deformable models of arteries. The method couples the equations of the deformation of the vessel wall at the variational level as a boundary condition for the fluid domain, in an approach analogous to J.R. Womersley's analytical solution for axisymmetric pulsatile flow in a long elastic tube [2]. We consider a strong coupling of the degrees of freedom of the fluid and the solid domains, and a linear membrane model (enhanced with through-plane stiffness) for the vessel wall. We have used the generalized-alpha method [3] to integrate the time evolution of the resulting equations for the deformable system, in a manner similar to that described by Jansen et al. [4].

Complement receptor 2 （CR2/CD21） is predominantly expressed on the surface of mature B cells where it forms part of a coreceptor complex that functions, in part, to modulate B-cell receptor signal strength. CR2/CD21 expression is tightly regulated throughout B-cell development such that CR2/CD21 cannot be detected on pre-B or terminally differentiated plasma cells. CR2/CD21 expression is upregulated at B-cell maturation and can be induced by IL-4 and CD40 signaling pathways. We have previously characterized elements in the proximal promoter and first intron of CR2/CD21 that are involved in regulating basal and tissue-specific expression. We now extend these analyses to the CR2/CD21 core promoter. We show that in mature B cells, CR2/~D21 transcription proceeds from a focused TSS regulated by a non-consensus TATA box, an initiator element and a downstream promoter element. Furthermore, occupancy of the general transcriptional machinery in pre-B versus mature B-cell lines correlate with CR2/CD21 expression level and indicate that promoter accessibility must switch from inactive to active during the transitional B-cell window.
