Effects of docosahexaenoic acid or arachidonic acid supplementation on the behavior of cardiomyocytes derived from human pluripotent stem cells

Background:Human heart changes its energetic substrates from lactate and glucose to fatty acids during the neonatal *** the lack of fatty acids in media for the culture of cardiomyocytes derived from human pluripotent stem cells(hiPS-CM),researchers have supplemented mixtures of fatty acids to hiPS-CM and reported the enhancement in the maturation of *** our previous studies,we separately supplemented two polyunsaturated fatty acids(PUFAs),docosahexaenoic acid(DHA)or arachidonic acid(AA),to rat fetal cardiomyocytes and found that the supplementations upregulated the expressions of mRNAs for cardiomyocyte differentiation,fatty acid metabolism,and cellular *** enhancement in cellular contractility was attributed to the improvement in intercellular connection rather than a direct enhancement of the contractile ***:This study reports the successive results of the effects of DHA or AA supplementation on *** addition to the contractile force and mRNA measurements used in the previous study,we further investigated the effect of different cellular aggregations on the contractile force output by means of finite element analysis,measured glucose and fatty acids metabolites,and assessed cTNT and MLC2v expressions through immunofluorecsence ***:It showed that the sole supplementation of albumin-conjugated DHA or AA can be taken up by hiPS-CM without other uptake-enhancing factors,and the supplementations may activate the CD36_ERRγmetabolic *** or AA supplementation increased the cellular contractile ratio on collagen gels and AA supplementation stimulated hiPS-CM aggregation to form cellular *** enhancement effect on the hiPS-CM contractile force was modest since the increase in contractile force was not *** supplementation was more effective than DHA supplementation because it significantly upregulated mRNA expressions of P300 and ***,finite element analysis showed that the formati

An experimental apparatus was developed to study the three dimensional separated flow with spiral-foci. The internal decelerating flow was generated by the air suction from a side wall to produce the separation on an opposite-side wall. The relation between the upstream boundary layer and the generation of spiral-foci in the separation region was observed by a tuft method. As a result, it was clarified that the spiral-focus type separation could be produced on the side wall and its behavior was closely related to the vortices supplied into the separation region from the boundary layer developing along top wall or bottom one.

Disaster Preparedness:Looking through the Lens of Hospitals in Japan

Critical facilities, such as hospitals, play a crucial role in the socioeconomic and psychological recovery of the population after a disaster. Hospitals are considered important due to their roles in saving lives in the affected population and must be able to withstand hazards and remain functioning during and after a disaster. This article assesses earthquake preparedness of hospitals in eight Japanese cities using a questionnaire survey. The questionnaire consists of six parameters and 21 indicators from the “four pillars of hospital preparedness” including structural, nonstructural, functional, and human resources. The results show that the majority of the respondent hospitals fulfill the functional preparedness, which is useful during the emergency period of a disaster, while the other three pillars—structural, nonstructural, and human resources—need to be strengthened. This study helps to assess the status of disaster preparedness as well as the gaps for these hospitals in the Tohoku and Nankai Trough regions, drawing lessons from the Great East Japan Earthquake and Tsunami of the Tohoku area. This status and the gaps are used as a departure point to indicate how to enhance preparedness and resilience to future disaster risks.

Modifying oxygen tension affects bone marrow stromal cell osteogenesis for regenerative medicine

AIM To establish a hypoxic environment for promoting osteogenesis in rat marrow stromal cells(MSCs) using osteogenic matrix cell sheets(OMCSs).METHODS Rat MSCs were cultured in osteogenic media under one of four varying oxygen conditions: Normoxia(21% O_2) for 14 d(NN), normoxia for 7 d followed by hypoxia(5% O_2) for 7 d(NH), hypoxia for 7 d followed by normoxia for 7 d(HN), or hypoxia for 14 d(HH). Osteogenesis was evaluated by observing changes in cell morphology and calcium deposition, and by measuring osteocalcin secretion(ELISA) and calcium content. In vivo syngeneic transplantation using OMCSs and β-tricalcium phosphate discs, preconditioned under NN or HN conditions, was also evaluated by histology, calcium content measurements,and real-time quantitative *** In the NN and HN groups, differentiated, cuboidal-shaped cells were readily observed, along with calcium deposits. In the HN group, the levels of secreted osteocalcin increased rapidly from day 10 as compared with the other groups, and plateaued at day 12(P < 0.05). At day 14, the HN group showed the highest amount of calcium deposition. In vivo, the HN group showed histologically prominent new bone formation, increased calcium deposition, and higher collagen type Ⅰ?messenger RNA expression as compared with the NN *** The results of this study indicate that modifying oxygen tension is an effective method to enhance the osteogenic ability of MSCs used for OMCSs.

Synthesis of P(NIPAM-co-Am)/Mesoporous Silica Composites and Their Temperature- Responsive Anion Exchange

The purpose of this study is to examine the structure and the temperature-responsive anion exchange property of amino-functionalized mesoporous silica coated with temperature-responsive copolymer, poly(N-isopropylacrylamide-co-acrylamide) (P(NIPAM-co-Am)). For this purpose, the composites which contained 0, 10, or 20 wt% of Am were synthesized. From the TG results, it was found that the amounts of copolymer immobilized on the mesoporous silica were 1.6 - 2.6 wt%. XRD patterns revealed that the structures of composites were hexagonal and almost the same as that of original mesoporous silica without polymer. At low temperature the methyl orange (MO) anions adsorbed and desorbed reversibly with changing pH of the solution, while at high temperature the MO anions did not. This temperature, at which the amount of adsorbed MO anions changed considerably, shifted to the higher temperature side with increasing the amount of added Am.

A semiconductor/dielectric interface is one of the dominant factors in device characteristics,and a variety of oxides with high dielectric constants and low interface trap densities have been used in carbon nanotube *** the crystal structure of nanotubes with no dangling bonds,there remains room to investigate unconventional dielectric ***,we fabricate carbon nanotube transistors with boron nitride nanotubes as interfacial layers between channels and gate dielectrics,where a single semiconducting nanotube is used to focus on switching behaviors at the subthreshold *** subthreshold swing of 68 mV·dec^(−1)is obtained despite a 100-nm-thick Sio_(2)dielectric,corresponding to the effective interface trap density of 5.2×10^(11)cm^(−2)·eV^(−1),one order of magnitude lower than those of carbon nanotube devices without boron nitride *** interfacial layers also result in the mild suppression of threshold voltage variation and *** achieve Ohmic contacts through the selective etching of boron nitride nanotubes with XeF2 gas,overcoming the trade-off imposed by wrapping the inner *** impacts of fluorinating carbon nanotubes on device performances are also confirmed as long as the etching is applied exclusively at source/drain *** results represent an important step toward nanoelectronics that exploit the advantage of one-dimensional van der Waals heterostructures.

Japan＇s first open sea offshore wind farm, Kamisu offshore windfarm Phase l, was stricken by an earthquake of intensity 6 on the Japanese seismic scale and a five-meter-high tsunami during the Great East Japan Earthquake on March 11,2011. The wind farm resumed operation on March 14 after checks revealed no damage to the system, even though the wind farm was temporarily forced to stop due to the grid failure caused by the earthquake. Wind turbines require a precise seismic design especially in an earthquake-prone country such as Japan. Wind power Kamisu Phase 2 was built one year after the earthquake based on the experience of Kamisu Phase 1. This paper presents the seismic design of offshore wind turbines and the situation during the earthquake and tsunami.

Effects of oral tacrolimus as a rapid induction therapy in ulcerative colitis

AIM:To determine the efficacy and safety of rapid induction therapy with oral tacrolimus without a meal in steroid-refractory ulcerative colitis(UC)***:This was a prospective,multicenter,observational *** May 2010 and August 2012,49 steroid-refractory UC patients(55 flare-ups)were consecutively *** patients were treated with oral tacrolimus without a meal at an initial dose of 0.1mg/kg per *** dose was adjusted to maintain trough whole-blood levels of 10-15 ng/m L for the first 2 *** of remission at 2 and 4 wk after tacrolimus treatment initiation was evaluated using Lichtiger’s clinical activity index(CAI).RESULTS:The mean CAI was 12.6±3.6 at *** the first 7 d,93.5%of patients maintained high trough levels(10-15 ng/m L).The CAI significantly decreased beginning 2 d after treatment *** 2wk,73.1%of patients experienced clinical *** tacrolimus initiation,31.4%and 75.6%of patients achieved clinical remission at 2 and 4 wk,*** was well ***:Rapid induction therapy with oral tacrolimus shortened the time to achievement of appropriate trough levels and demonstrated a high remission rate 28 d after treatment *** induction therapy with oral tacrolimus appears to be a useful therapy for the treatment of refractory UC.
