Nanostructure and mechanical properties of the osteocyte lacunar-canalicular network-associated bone matrix revealed by quantitative nanomechanical mapping

Osteocytes are the main bone cells embedded in the bone matrix where they form a large surface-area network called the lacunar-canalicular network （LCN）, interconnecting their resident spaces with the lacunae by the canaliculi. Increasing evidence points toward osteocytes playing a pivotal role in maintaining bone quality. On the one hand, osteocytes transmit mechanical strain and microenvironmental signals through the LCN to regulate the activity of osteoblasts and osteoclasts; on the other hand, osteocytes are suggested to be able to remodel the LCN-associated bone matrix. However, due to the challenges involved in the assessment and characterization of the LCN-associated bone matrix, little is known about its structure and the corresponding mechanical properties. In this work, we used quantitative nanomechanical mapping, backscattered electron imaging, and nanoindentation to characterize the LCN-associated bone matrix. The results show that the techniques can be used to probe the LCN-associated bone matrix. Nanoindentation and quantitative mechanical mapping reveal spatially inhomogeneous mechanical properties of the bone matrix associated with the osteocyte lacunae and canaliculi. The obtained nano-topography and corresponding nano-mechanical maps reveal altered mechanical properties in the immediate vicinity of the osteocyte lacunae and canaliculi, which cannot be explained solely by the topographic change.

Effect of Prometheus liver assist system on systemic hemodynamics in patients with cirrhosis: A randomized controlled study

AIM： To evaluate treatment safety and hemodynamic changes during a single 6-h treatment with the PrometheusTM liver assist system in a randomized, controlled study. METHODS： Twenty-four patients were randomized to either the study group or to one of two control groups： Fractionated Plasma Separation Adsorption and Dialysis, PrometheusTM system （Study group; n = 8）; Molecular Adsorbent Recirculation System （MARS）TM （Control group 1, n = 8）; or hemodialysis （Control group 2; n = 8）. All patients included in the study had decompensated cirrhosis at the time of the inclusion into the study. Circulatory changes were monitored with a Swan-Ganz catheter and bilirubin and creatinine were monitored as measures of protein-bound and water-soluble toxins. RESULTS： Systemic hemodynamics did not differ between treatment and control groups apart from an increase in arterial pressure in the MARS group （P = 0.008）. No adverse effects were observed in any of the groups. Creatinine levels significantly decreased in the MARS group （P = 0.03） and hemodialysis group （P = 0.04）. Platelet count deceased in the Prometheus group （P = 0.04）.CONCLUSION： Extra-corporal liver support with Prometheus is proven to be safe in patients with endstage liver disease but does not exert the beneficial effects on arterial pressure as seen in the MARS group,

Theranostic nanoagents that integrate the diagnoses and therapies within a single nanomaterial are compelling in their use for highly precise and efficient antitumor treatments. Herein, polyethylene glycol （PEG）-modified cobalt sulfide nanosheets （CoS-PEG NSs） are synthesized and unitized as a powerful theranostic nanoagent for efficient photothermal conversion and multimodal imaging for the first time. We demonstrate that the obtained CoS-PEG NSs show excellent compatibility and stability in water and various physiological solutions, and can be effectively inter- nalized by cells, but exhibit a low cytotoxicity. The CoS-PEG NSs exhibit an efficient photothermal conversion capacity, benefited from the strong near-infrared （NIR） absorption, high photothermal conversion efficiency （~ 33.0%）, and excellent photo- thermal stability. Irnportant136 the highly effective photothermal killing effect on cancer ceils after exposure to CoS-PEG NSs plus laser irradiation has been con- firmed by both the standard Cell Counting Kit-8 and live-dead cell staining assays, revealing a concentration-dependent photothermal therapeutic effect. Moreover, utilizing the strong NIR absorbance together with the T2-MR contrast ability of the CoS-PEG NSs, a high-contrast triple-modal imaging, i.e., photoacoustic （PA）, infrared thermal （IRT）, and magnetic resonance （MR） imaging, can be achieved, suggesting a great potential for multimodal imaging to provide comprehensive cancer diagnosis. Our work introduces the first bioapplication of the CoS-PEG nanomaterial as a potential theranostic nanoplatform and may promote further rational design of CoS-based nanostructures for precise/effident cancer diagnosis and therapy.

Molecular recognition between nucleobases plays a crucial role in all kinds of biological ***,real-space investigation of the recognition capability of nucleobases in the presence of interfering compounds remains ***,based on the combination of scanning tunneling microscopy imaging and density functional theory modeling,we report the impact of the presence of melamine(M)on the formation and chirality of guanine(G)-tetrads on Au(111).Although M can interact with G by double hydrogen bonding,the Hoogsteen base pairing of G is not compromised,forming identical individual G-tetrads as would have happened without the presence of M.G-tetrads coexist with M on the surface not only in separate domains,but also within the mixture network of G-tetrads and *** the adsorption orientation of G-tetrads in the mixture network diversifies into two distinct angles,all G-tetrads in the network keep the same chirality,emphasizing the high preference of homochirality in such biochemical systems.

The adsorption of 2,6-naphthalenedicarboxylic acid(NDCA)molecules on the Ag(110),Cu(110),and Ag(111)surfaces at room temperature has been studied by means of scanning tunnelling microscopy(STM).Further supporting results were obtained using X-ray photoelectron spectroscopy(XPS)and soft X-ray absorption spectroscopy(XAS).On the Ag(110)support,which had an average terrace width of only 15 nm,the NDCA molecules form extended one-dimensional(1-D)assemblies,which are oriented perpendicular to the step edges and have lengths of several hundred *** shows that the assemblies have a large tolerance to monatomic surface steps on the Ag(110)*** observed behaviour is explained in terms of strong intermolecular hydrogen bonding and a strong surface-mediated directionality,assisted by a sufficient degree of molecular backbone *** contrast,the same kind of step-edge crossing is not observed when the molecules are adsorbed on the isotropic Ag(111)or more reactive Cu(110)*** Ag(111),similar 1-D assemblies are formed to those on Ag(110),but they are oriented along the step *** Cu(110),the carboxylic groups of NDCA are deprotonated and form covalent bonds to the surface,a situation which is also achieved on Ag(110)by annealing to 200°*** results show that the formation of particular self-assembled molecular nanostructures depends significantly on a subtle balance between the adsorbate-adsorbate and adsorbate-substrate interactions and that kinetic factors play an important role.

Fabrication of 3D air-core MEMS inductors for very-highfrequency power conversions

We report a fabrication technology for 3D air-core inductors for small footprint and very-high-frequency power *** process is scalable and highly generic for fabricating inductors with a wide range of geometries and core *** demonstrate spiral,solenoid,and toroidal inductors,a toroidal transformer and inductor with advanced geometries that cannot be produced by wire winding *** inductors are embedded in a silicon substrate and consist of through-silicon vias and suspended *** inductors fabricated with 20 and 25 turns and 280-350μm heights on 4-16 mm2 footprints have an inductance from 34.2 to 44.6 nH and a quality factor from 10 to 13 at frequencies ranging from 30 to 72 *** air-core inductors show threefold lower parasitic capacitance and up to a 140% higher-quality factor and a 230% higher-operation frequency than silicon-core inductors.A 33 MHz boost converter mounted with an air-core toroidal inductor achieves an efficiency of 68.2%,which is better than converters mounted with a Si-core inductor(64.1%).Our inductors show good thermal cycling stability,and they are mechanically stable after vibration and 2-m-drop tests.

Parkinson’s disease and affective disorder: The temporal relationship

In relationship between the affective disorders and Parkinson’s disease (PD) it was found that comorbidity was higher than expected in the majority of the studies. Patients with PD are at increased risk of developing depression and, conversely, recent studies have shown that patients with depressive disorders have increased risk of incident PD. However, the temporal associations between the disorders are not fully elucidated. From this review it could be learned that the temporal aspects strongly suggest that a neurobiological association exists between affective disorder and PD. This is illustrated with hitherto unpublished data. Some of these issues may be investigated in case register studies, e.g. by linkage of registers of somatic and psychiatric illness, and suggestions for future research are given. For GP’s, psychiatrists, geriatricians, and neurologists these new findings will lead to a better understanding and better treatment for patients with complicated comorbid conditions. Here timing is important!
