Backg.ound:Black locust(BL,Robinia pseudoacacia)is considered a promising.tree species for reforestation due to its g.eat ability to fix ***,after two or three coppice-harvesting.rotations,the productivity of BL *** soil microbial communities are affected and how these g.oups correlate with the nitrog.n mineralization process across multi-g.neration stands remains ***:We investig.ted the composition and structure of free-living.nitrog.n-fixing.microorg.nisms(diazotrophs)by sequencing.the marker g.ne nifH and compared these results to levels of soil nitrog.n mineralization in the bulk soil and rhizosphere in black locust plantations on Mount Tai,***:The results showed multi-g.neration BL coppice plantations decreased the total soil nitrog.n(N),soil phosphorus(P),soil microbial biomass N(MBN),soil microbial biomass C(MBC),soil nitrification rate(Rn),soil ammonification rate(ra),and net soil N mineralization rate(Rm),but sig.ificantly increased the concentration of soil NH_(4þ)-N to maintain sufficient NO_(3)^(-)*** dominant species in bulk soil and rhizosphere chang.d from Rhodopseudomonas(22.62%and 15.76%),unclassified_c_Alphaproteobacteria(22.37%and 29.28%),unclassified_o_Rhizobiales(15.40%and 13.31%),Bradyrhizobium(12.00%and 11.74%)in seedling.plantations to Bradyrhizobium(45.95%and 47.86%)and Rhodopseudomonas(43.56%and 41.84%)in coppice plantations,*** test and Redundancy analysis(RDA)revealed that Rn,ra,and Rm were the most important factors shaping.the diazotrophic ***:Our results sug.est that the multi-g.neration BL coppice plantation can homog.nize soil diazotrophic communities,which is mainly reg.lated by the available N loss caused by nitrog.n *** the manag.ment technolog. of coppice plantations will provide more beneficial external consumption.

Sing.e doses of p38 MAP kinase inhibitors or prednisolone affect CRP and IL-6 in patients with active Rheumatoid Arthritis (ra)

Purpose: To investig.te the effects of sing.e doses of losmapimod, dilmapimod (inhibitors of p38 MAPK) and prednisolone on biomarkers of systemic inflammation (serum C-reactive protein (CRP) and interleukin (IL)-6) in subjects with active rheumatoid arthritis (ra). Methods: Two randomized, double blind, placebo controlled, parallel g.oup, sing.e dose studies investig.ted the CRP and IL-6 dose-response relationships for losmapimod (study A) and dilmapimod and prednisolone (study B) in patients with active ra on stable weekly doses of methotrexate. CRP, IL-6 and other exploratory biomarkers were measured in blood at baseline and up to 72 hours post-dosing. Results: In study A, 51 subjects were randomized to receive losmapimod (7.5, 20 and 60 mg. or placebo and in study B, 77 subjects were randomized to receive dilmapimod (7.5, 15 and 25 mg. or prednisolone (10, 20 and 50 mg. or placebo. Sing.e doses of prednisolone caused a decrease in circulating.IL-6 detectable at 3 and 24 hours post-dose as well as a CRP sing.e-dose relationship at 48 hours post-dose in patients with active ra. All doses of losmapimod produced a statistically sig.ificant reduction in serum IL-6, 3 hours post-dose but had no effect on CRP. Dilmapimod had no effect on IL-6 or CRP at any dose or time. Conclusion: This sing.e-dose response clinical study desig. could be useful in the early clinical development of anti-inflammatory ag.nts for the treatment of rheumatoid arthritis. Inhibition of both circulating.IL-6 and CRP would increase confidence that a new chemical entity may have the potential to deliver clinical benefit in ra.
