RAGE is associated with prognosis and angiogenesis in human oral squamous cell carcinoma

Purpose: RAGE is a multi ligand receptor. We previously reported the RAGE is closely associated with tumor progression and metastasis in gastrointestinal cancer, prostate cancer and malignant transformation of colorectal adenoma. In this study, we examined the role of RAGE in oral squamous cell carcinoma. Methods: Firstly, RAGE expression was examined by immunohistochemistry in 74 cases with OSCC and compared with clinicopathological parameters. Secondly, RAGE concentration were examined by enzyme-linked immunosorbent assay (ELISA) in OSCC samples and compared with other parameters. Finally, in vitro examinations were performed using human OSCC cell line, HSC-3 and HSC-4 cells. Results: 1. RAGE immunoreacctivity was associated with depth of invasion (P<0.0001), local recurence (P<0.0001) and RAGE expression was an independent prognostic factor for disease free survival by multivariate analysis in OSCC (P=0.0022). *** RAGE concentration was associated with higher tumor microvessel density (MVD) (P=0.0123) and tumor vascular endothelial growth factor (VEGF) concentration (P=0.0344), but not with lymph vessel density (LVD) and VEGF-C concentration. 3. Treatment with RAGE ligand, HMGB 1 increased the secretion of VEGF but not that of VEGF-C in HSC-3 and HSC-4 cells. The effect of HMGB 1 was inhibited by RAGE antisense S-ODN treatment. Conclusion: These results suggest that RAGE may be a key factor in local recurrence and prognosis via angiogenesis in OSCC

＜正＞Small-scale reactor named IMR （Integrated Modular Water Reactor） is beingdeveloped in Japan. The primary strategy in the development of small-scale reactors isto construct a new design concept which satisfies the requirements of enhanced safetyand construction cost reduction. For this goal, a reactor vessel with an integratedprimary system configuration is chosen as the most optimum reactor configuration inthe present work. This configuration adopts natural circulation and self-pressurizationsystems, which eliminate pressurizer, reactor coolant pumps and all main coolantpipes. In addition to these basic designs, we have also developed new concepts whichenhance safety and reduce construction cost as well. These incorporate the ideas ofHybrid Heat Transport System （HHTS） and Stand-alone Direct Heat Removal System（SDHS）. The HHTS has been designed to keep the stable and safe control of powerand system pressure. The SDHS was proposed to remove the decay heat directly fromthe reactor vessel to the atmosphere in case of accident.

A dye-sensitized photocatalyst combining Pt-loaded TiO_(2)and Ru(Ⅱ)tris-diimine sensitizer(RuP)was constructed and its activity for photochemical hydrogen evolution was compared with that of Pt-intercalated HCa_(2)Nb_(3)O_(10)*** the sacrificial donor ethylenediaminetetraacetic acid(EDTA)disodium salt dihydrate was used,RuP/Pt/TiO_(2)showed higher activity than RuP/Pt/HCa_(2)Nb_(3)O_(10).In contrast,when NaI(a reversible electron donor)was used,RuP/Pt/TiO_(2)showed little activity due to back electron transfer to the electron acceptor(I3-),which was gener-ated as the oxidation product of I-.By modification with anionic polymers(sodium poly(styrenesulfonate)or sodium polymethacrylate)that could inhibit the scavenging of conduction band electrons by I3-,the H_(2)production activity from aqueous NaI was improved,but it did not exceed that of RuP/Pt/HCa_(2)Nb_(3)O_(10).Transient absorption measurements showed that the rate of semiconductor-to-dye back electron transfer was slower in the case of TiO_(2)than HCa_(2)Nb_(3)O_(10),but the electron transfer reaction to I3-was much *** results indicate that Pt/TiO_(2)is useful for reactions with sacrificial reductants(e.g.,EDTA),where the back electron transfer reaction to the more reducible product can be ***,more careful design of the catalyst will be nec-essary when a reversible electron donor is employed.

Radiotherapy for multiple brain metastases from hepatocellular carcinomas

A 78-year-old man with liver cirrhosis was found to have multiple hepatocellular carcinomas （HCCs） and underwent 3 sessions of transcatheter arterial chernoernbolization. Fourteen months after diagnosis, the patient presented with left herniparesis. Contrast- enhanced magnetic resonance imaging showed multiple metastases with ring-shaped enhancement in the cerebrum and cerebellum. There were no metastases to other organs. The metastatic lesions almost completely disappeared after whole-brain radiotherapy with a total dose of 50 Gy. Neurologic symptoms decreased, and the patient＇s quality of life improved. The patient underwent 2 more sessions of transcatheter arterial chemoembolization. Twelve months after the diagnosis of brain metastasis, the patient remains alive. The present case indicates that radiotherapy can improve quality of life and prolong survival in some patients with brain metastases from HCCs.

Photoreaction of 9-vinylcarbazole in acetonitrile in the presence of titanium dioxide and a catalytic amount of magnesium perchlorate gave 3,6-di(9-carbazolyl)-1,2-dioxane as a photooxygenated product via photodimerization of *** photoreaction proceeds via an electron transfer mechanism,where magnesium perchlorate accelerated formation of the photo-oxygenated product.

目的分析MRL/lpr和(NZB×NZW)F1两种狼疮鼠成年发病时,广泛浸润于多脏器的T细胞克隆T细胞受体(TCR)Vβ基因编码的氨基酸基序特点,揭示该T细胞克隆的抗原特异性。方法用反转录聚合酶链反应(RT-PCR)分别扩增不同成年发病狼疮鼠个体的肾脏、大脑中的TCRVβ6基因,对扩增产物进行基因克隆和DNA测序分析;单链构象多态性分析法(SSCP)鉴定广泛浸润的T细胞克隆TCRVβ6基因编码的氨基酸基序。结果两种成年发病狼疮鼠的不同个体之间,广泛浸润于多脏器的T细胞克隆TCRVβ6基因的互补决定区3(CDR3)中出现相同的氨基酸基序。结论不同个体成年发病狼疮鼠的多脏器中广泛浸润的T细胞克隆TCRVβ基因的CDR3中出现相同的氨基酸,表明该克隆针对特定的自身抗原。

Dipeptidyl peptidase Ⅳ (DPPIV), which belongs to the prolyl oligopeptidase family of serine proteases, is known to have a variety of regulatory biological functions and has been shown to be implicated in type 2 diabetes. It is therefore important to develop selective human DPPIV (hDPPIV) inhibitors. In this study, we determined the crystal structure of apo hDPPIV at 1.9 A resolution. Our high-resolution crystal structure of apo hDPPIV revealed the presence of sodium ion and glycerol molecules at the active site. In order to elucidate the hDPPIV binding mode and substrate specificity, we determined the crystal structure of hDPPIV-diprotin B (Val-Pro-Leu) complex at 2.1 A resolution, and clarified the difference in binding mode between diprotin B and diprotin A (Ile-Pro-Ile) into the active site of hDPPIV. Comparison between our crystal structures and the reported apo hDPPIV structures revealed that positively charged functional groups and conserved water molecules contributed to the interaction of ligands with hDPPIV. These results are useful for the design of potent hDPPIV inhibitors.

A 38-year-old woman was referred to our institution due to epigastralgia. She presented with obstructive jaundice and eosinophilia. Endoscopic retrograde cholangiopancreatography showed diffuse narrowing from the distal common bile duct to the bifurcation of the hepatic ducts. An endoscopic plastic biliary stent was inserted; the specimen obtained from the common bile duct wall revealed dense infiltration by eosinophils. Treatment was started with prednisolone 60 mg daily. The patient's biliary stenosis and eosinophilia gradually improved. Eosinophilic infiltration in the lungs or stomach is relatively common, but it is rare in the common bile duct. Most of the reported cases of eosinophilic cholangitis presented with eosinophilia; our patient's eosinophil count was over 1000/mm3. Since our patient had allergies to pollen and house dust, a relationship between the allergies and the eosinophilic cholangitis was suspected, but no cause was identified.
