Extract of Yokukansan improves anxiety-like behavior and increases serum brain-derived neurotrophic factor in rats with cerebral ischemia combined with amyloid-β42 peptide

OBJECTIVE: To examine the effects of Yokukansan(YKS) extract on two endogenous modulators of anxiety, brain-derived neurotrophic factor(BDNF)and serotonin (5-HT)_(2A) receptors pharmacologically, in the ischemic rat model of ***: The cerebral ischemia(CI) was induced by bilateral occlusion of the vertebral and common carotid arteries(4-vessel occlusion ischemia). The CI was combined with the amyloid-β42 peptide(A42) injected intracerebroventricularly, and referreβdto as CI+Aβ. Anxiety-like behaviors were assessed by elevated plus maze(enclosed arm), light/dark transition test(dark chamber), and open-field ***-dog shakes were induced by the 5-HT_(2A) receptor agonist 2, 5-dimethoxy-4-iodoamphetamine(DOI). The concentration of BDNF in serum was determined by enzyme-linked immuno sorbent ***: CI + Aβ increased anxiety, as demonstrated by the increase of time spent in the enclosed arms and dark chambers, and locomotion in the outer zone of the open field(thigmotaxis). CI + Adecreased the serum concentration of BDNF. YKS reβ-duced the anxiety-like behaviors, suppressed the DOI-induced wet-dog shakes and increased serum BDNF ***: Our findings suggest that YKS extract improves CI + Aβ-induced anxiety by antagonizing 5-HT_(2A) receptors and increasing BDNF.

Is hepatic arterial infusion chemotherapy effective treatment for advanced hepatocellular carcinoma resistant to transarterial chemoembolization?

AIM:To evaluate the effectiveness of hepatic arterial infusion chemotherapy(HAIC) for advanced hepatocellular carcinoma(HCC) resistant to transarterial chemoembolization(TACE).METHODS:This study was conducted on 42 patients who received HAIC for advanced HCC between 2001and 2010 at our hospital.5-fluorouracil(5-FU) was administered continuously for 24 h from day 1 to day 5 every 2-4 wk via an injection ***-arterial cisplatin or subcutaneous interferon was administered in combination with the *** patients enrolled in this retrospective study were divided into two groups according to whether or not they fulfilled the criteria for resistance to TACE proposed by the Japan Society of Hepatology in 2010(written in Japanese);one group of patients who did not fulfill the criteria for TACE resistance(group A,n = 23),and another group who fulfilled the criteria for TACE resistance(group B,n = 19).We compared the outcomes in terms of the response and survival rates between the two ***:Both the response rate and tumor suppression rate following HAIC were significantly superior in group A than in group B(response rate:48% vs 16%,P = 0.028,tumor suppression rate:87% vs 53%,P = 0.014).Furthermore,both the progression-free survival rate and survival time were significantly superior in group A than in group B(3-,6-,12-,and 24-mo = 83%,70%,29% and 20% vs 63%,42%,16% and 0%,respectively,P = 0.040,and 9.8 mo vs 6.2 mo,P = 0.040).A multivariate analysis(Cox proportional hazards regression model) showed that resistance to TACE was an independent predictor of poor survival(P = 0.007).CONCLUSION:HAIC administrating 5-FU was not effective against advanced HCC resistant to *** tools for treatment,i.e.,molecular-targeting agents may be considered for these cases.

Toll-like receptor 9 gene mutations and polymorphisms in Japanese ulcerative colitis patients

Abnormal innate immune responses toward luminal bacteria play an important role in the pathogenesis of inflammatory bowel *** has been demonstrated that bacteria having CpG DNA ameliorate experimental colitis in mice,and Toll-like receptor 9 (TLR9) signaling mediates the anti-inflammatory effects in mouse colonic inflammation.A gene variation in NOD2/CARD15 has been reported in Crohn's disease (CD) patients in Western countries,but this variation has not been identified in Japanese CD ***,we hypothesized that TLR9 is a key factor in the development of ulcerative colitis (UC),and we investigated gene mutations and polymorphisms of TLR9 in Japanese UC *** single nucleotide polymorphisms (SNPs) in TLR9 were identified in healthy controls,and were assessed in 48 UC patients and 47 healthy *** subjects were matched for age,sex and date of blood sampling from among a subgroup of *** found that TLR9-1486CC,1174GG and 2848AA increase the risk of UC [odds ratio (OR) 2.64,95% confidence interval (95% CI):1.73-6.53,P=0.042],and TLR9-1486TT,1174AA and 2848GG decrease the risk of UC (OR 0.30,95% CI:0.10-0.94,P=0.039),although there were no correlations between SNPs and disease phenotype or TLR9 mRNA *** findings suggest that TLR9 polymorphisms are associated with increased susceptibility to UC.

Dirac reduction of binary asteroid systems with symmetry and stability of relative equilibria

In this paper, we study symmetry reduction for a binary asteroid system modeled by a rigid body and a particle. In particular, we demonstrate how translational and rotational symmetry reduction appeared in the binary asteroid system can be carried out in the context of Dirac reduction by stages and with the associated reduction of implicit Hamiltonian systems. Then we investigate Stability of relative equilibria of the asteroid pair and show stability regions by using the energy-momentum method. Lastly, we illustrate some numerical simulations for stable and unstable orbits near from relative equilibria of the Collinear and T configurations.C 2013 The Chinese Society of Theoretical and Applied Mechanics. Simultaneously disrupting AtPrx2,AtPrx25 and AtPrx71 alters lignin content and structure in Arabidopsis stem

Plant class III heme peroxidases catalyze lignin polymerization. Previous reports have shown that at least three Arabidopsis thaliana peroxidases, AtPrx2, AtPrx25 and AtPrx71, are involved in stem lignification using T-D NA insertion mutants, atprx2, atprx25, and atprx71. Here, we generated three double mutants, atprx2/atprx25, atprx2/atprx71, and atprx25/atprx71, and investigated the impact of the simultaneous deficiency of these peroxidases on lignins and plant growth. Stem tissue analysis using the acetyl bromide method and derivatization followed by reductive cleavage revealed improved lignin characteristics, such as lowered lignin content and increased arylglycerol-β-aryl （β-O-4） linkage type, especially β-O-4 linked syringyl units, in lignin, supporting the roles of these genes in lignin polymerization. In addition, none of the double mutants oexhibited severe growth defects, such as shorter plant stature, dwarfing, or sterility, and their stems had improved ceil wall degradability. This study will contribute to progress in lignin bioengineering to improve iignocellulosic biomass.

Comparison of imo and Humphrey field analyzer perimeters in glaucomatous eyes

AIM:To compare the imo perimeter,a new portable head-mounted perimeter unit that enables both eyes to be examined quickly and simultaneously,with the Humphrey field analyzer(HFA)perimeter to investigate correlations and their diagnostic ability in glaucomatous ***:The performance of the equipment in 128 glaucomatous eyes and 40 normal eyes were *** investigated the correlations of mean deviation,pattern standard deviation,visual field index,and the ***:Measurements of mean deviation(r=0.886,P<0.001),pattern standard deviation(r=0.814,P<0.001),and visual field index(r=0.871,P<0.001)in both perimeters were strongly and positively *** sensitivities in the imo perimeter were 80.5%for mean deviation,81.2%for pattern standard deviation,and 80.5%in visual field index;those in the HFA were 63.3%for mean deviation,74.5%for pattern standard deviation,and 80.5%for visual field *** perimeters demonstrated high diagnostic ***:The parameters by the imo and HFA in glaucomatous eyes show strong positive correlations with favorable sensitivity,specificity,and diagnostic ***,the difference between imo and HFA results increases with the increase in visual field disturbance.

Since ancient times,humans have bred several plants that we rely on ***,little is known about the divergence of most of these *** the present study,we investigated the divergence of Mibuna(Brassica rapa *** ***),a traditional leafy vegetable in Kyoto(Japan),by combining genetic analysis and a survey of ancient *** is considered to have been bred 200 years ago from Mizuna,another traditional leafy vegetable in *** has simple spatulate leaves,whereas Mizuna has characteristic serrated *** quantitative trait loci(QTL)and gene expression analyses suggested that the downregulation of BrTCP15 expression contributed to the change in the leaf shape from serrated to simple ***,the SNP analysis indicated that the genomic region containing the BrTCP15 locus was transferred to Mibuna by ***,we conducted a survey of ancient literature to reveal the divergence of Mibuna and found that hybridization between Mizuna and a simple-leaved turnip might have occurred in the ***,the genomic analysis of multiple turnip cultivars showed that one of the cultivars,Murasakihime,has almost the same sequence in the BrTCP15 region as *** results suggest that the hybridization between Mizuna and turnip has resulted in the establishment of Mibuna.

Family empowerment and associated factors in Japanese families raising a child with severe motor and intellectual disabilities

Objectives:Family caregivers raising children with severe motor and intellectual disabilities(SMID)experience the enormous burden of *** concept of family empowerment is one of the important assessment indexes of family nursing from the perspective of providing comprehensive support for these *** objective of this study was to identify the factors associated with the empowerment of families raising a child with SMID in ***:We conducted a nationwide questionnaire survey involving 1659 primary caregivers raising a child with SMID through 89 special *** assessed the main outcomes using the Family Empowerment Scale(FES).We then conducted a multiple linear regression analysis to reveal the factors associated with family ***:In total,1362 primary caregivers were included in our *** results show that factors contributing to high FES scores are higher age of the primary caregiver,higher education,greater recognition of regional support,lower childcare burden,higher utilization of home visit services,higher usage of a childcare institution,higher household income,and stronger family ***:Healthcare professionals should carefully assess the state of family empowerment of the primary caregivers who are younger and those who have low education,low household income,high childcare burden,and fragile bonding with the ***,they should encourage such families to use regional support resources for *** is,policy makers should consider ways to promote home visits and institutional services for the care of children with SMID,aiming especially for the provision of well-coordinated care and services.

Transcriptional regulation of human polo-like kinases and early mitotic inhibitors

Human polo-like kinases （PLK1-PLK4） have been implicated in mitotic regulation and carcinogenesis. PLK1 phosphorylates early mitotic inhibitor 1 （Emil） to ensure mitosis entry, whereas Emi2 plays a key role during the meiotic cell cycle. Transcription factor E2F is primarily considered to regulate the G1/S transition of the cell cycle but its involvement in the regulation of mitosis has also been recently suggested. A gap still exists between the molecular basis of E2F and mitotic regulation. The present study was designed to characterize the transcriptional regulation of human PLK and Emi genes. Adenoviral overexpression of E2F1 increased PLK1 and PLK3 mRNA levels in A549 cells. A reporter gene assay revealed that the putative promoter regions of PLK1, PLK3, and PLK4 genes were responsive to activators E2F, E2F1-E2F3. We further characterized the putative promoter regions of Emil and Emi2 genes, and these could be regulated by activators E2F and E2F1-E2F4, respectively. Finally, PLK1-PLK4, Emil, and Emi2 mRNA expression levels in human adult, fetal tissues, and several cell lines indicated that each gene has a unique expression pattern but is uniquely expressed in common tissues and cells such as the testes and thymus. Collectively, these results indicate that E2F can integrate G1/S and G2/M to oscillate the cell cycle by regulating mitotic genes PLK and Emi, leading to determination of the cell fate.
