mEK inhibition activates STAT signaling to increase breast cancer immunogenicity via mHC-I expression

Aim:Immunotherapy and immune checkpoint inhibitors(ICI)have changed cancer care for many patients;however,breast cancers have exhibited minimal response to single agent ICI *** is a significant need to identify novel targets capable of increasing cancer cell immunogenicity and response to ICIs in breast *** activated protein kinase(mAPK)signaling is essential for many cellular processes but the relationship between mAPK signaling and cancer cell immunogenicity is less well *** reports suggest that mEK inhibition(mEKi)affects the tumor-immune microenvironment by altering the expression of interferon responsive PD-L1 and mHC-I through unknown ***:Using western blotting and flow cytometry,we sought to determine whether mEKi affects JAK-STAT signaling upstream of PD-L1 and mHC-I expression in a panel of mouse mammary cancer and triple negative breast cancer cell ***:The cell lines tested exhibited increased STAT activation in response to mEKi ***,mEKi-induced mHC-I and PD-L1 expression are dependent upon STAT1 in mmTV-Neu ***,mEKiinduced STAT activation and interferon-responsive protein expression are abrogated with ErbB-family inhibitor co-treatment in mmTV-Neu cells,suggesting ErbB receptor signaling dependence,but not in basal-like cell ***,analysis of basal-like breast cancer patient samples exhibited an inverse relationship between STAT1 and Ras/mAPK activation ***:These findings suggest that mAPK signaling and STAT activation are inversely related in both mouse and human mammary *** work also supports further study of mEKi to increase STAT signaling and potentially,immunotherapy responses through increased mHC-I and PD-L1 expression.

Feeding behavior and hormoligosis associated with imidacloprid resistance in Asian citrus psyllid,Diaphorina citri

Imidacloprid is a neonicotinoid insecticide used for managing the Asian citrus psyllid,Diaphorina citri Kuwayama,which serves as vector of phytopathogens causing citrus ***,development of resistance to neonicotinoids among populations of *** has coincided with occasional control failures in the *** objectives of this research were to(1)survey current levels of imidacloprid resistance in Florida citrus;(2)compare feeding behavior between imidacloprid-resistant and susceptible *** using electrical penetration graph recordings,and(3)investigate the possible amplification of insecticide hormoligosis associated with *** surveys confirmed that the susceptibility of *** populations to imidacloprid has decreased in commercial Florida citrus groves compared with a laboratory-susceptible *** 12 generations of selection,resistance to imidacloprid increased by 438 fold compared with the susceptible ***-susceptible *** feeding on citrus exhibited significantly more bouts associated with intercellular pathway(C),phloem penetration(D),phloem salivation(E1),and nonprobing(Np)activities than imidacloprid-resistant ***,there were no differences observed in the frequency or duration of phloem ingestion or xylem feeding between susceptible and resistant *** was no statistical difference in fecundity between resistant and susceptible ***,the fecundity of imidacloprid-susceptible female *** treated with a sublethal concentration of imidacloprid(LC_(25))increased significantly compared with controls,while such hormoligosis was less pronounced among imidacloprid-resistant *** results suggest that imidacloprid-resistant psyllids may cease feeding sooner than susceptible counterparts following sublethal exposure to this insecticide,indicative of a behavioral resistance mechanism.

Building-Integrated Photovoltaic(BIPV)on vertical façades is a potential PV application in today’s *** performance of BIPV on façades is significantly influenced by the façade *** tropical cities,the optimum façade orientation,in terms of maximum energy yield and daylight performance,cannot be simply determined,due to relatively symmetrical sun path throughout the *** study therefore aims to determine the optimum orientation for BIPV on tropical building faç*** achieve the objective,experiment,modelling,and computational simulation are conducted to evaluate the BIPV energy yield and to predict the indoor daylight performance in a scale-model building with a 105Wp monocrystalline silicon PV,facing each cardinal orienta-tion in Bandung,*** South orientation yields practically zero ASE_(1000,250),providing the best annual daylight performance,and yielding the most desirable value in four out of five daylight *** greatest annual energy yield is at the North orientation,providing 179-186 kWh(95%prediction interval)per year,but with larger uncertainty compared to that at the South,due to direct sunlight *** on three different objective functions,the South orientation is considered optimum for placing the BIPV panel on the prototype façade in the location.

A Rare Case of Aortic Valve Endocarditis and Acute meningitis Due to Haemophilus influenzae

HACEK organisms represent a rare but important group of causative pathogens in endocarditis. These bacteria have historically been associated with culture-negative endocarditis;however, modern laboratory techniques have made this less common. In this case, we present a 74-year-old man who presented with acute onset altered mentation, fever, and sepsis. He was ultimately found to have Haemophilus influenzae meningitis, cerebral empyema, aortic valve endocarditis, psoas myositis, and L2 - L3 diskitis with osteomyelitis. Although HACEK organisms are commonly found in the oropharynx and upper respiratory tract in humans, our patient did not report recent preceding dental or ENT procedures. H. influenzae is responsible for approximately 0.16% of all cases of bacterial endocarditis, representing a very limited subset. Although generally considered low virulent pathogens, this case demonstrates the unusual extent of infection from a HACEK organism, H. influenzae, causing aortic valve endocarditis as well as atypical non-cardiac sequelae, including acute meningitis.

In 2019,the Event Horizon Telescope(EHT)released the first-ever image of a black hole event *** are now aiming for higher angular resolutions of distant targets,like black holes,to understand more about the fundamental laws of gravity that govern our *** achieve this higher resolution and increased sensitivity,larger radio telescopes are needed to operate at higher frequencies and in larger *** like the next-generation Very Large Array(ngVLA)and the Square-Kilometer Array(SKA)require building hundreds of telescopes with diameters greater than 10 ms over the next *** has a twofold *** telescope surfaces need to be more accurate to operate at higher frequencies,and the logistics involved in maintaining a radio telescope need to be simplified to support them properly in large *** of these problems can be solved with improved methods for surface metrology that are faster and more accurate with a higher *** leads to faster and more accurate panel alignment and,therefore,a more productive *** this paper,we present the use of binocular fringe projection profilometry as a solution to this problem and demonstrate it by aligning two panels on a 3-m radio telescope *** measurement takes only 10 min and directly delivers feedback on the tip,tilt,and piston of each panel to create the ideal reflector shape.

Breast cancer detection heavily relies on medical imaging, particularly ultrasound, for early diagnosis and effectivetreatment. This research addresses the challenges associated with computer-aided diagnosis (CAD) of breastcancer fromultrasound images. The primary challenge is accurately distinguishing between malignant and benigntumors, complicated by factors such as speckle noise, variable image quality, and the need for precise segmentationand classification. The main objective of the research paper is to develop an advanced methodology for breastultrasound image classification, focusing on speckle noise reduction, precise segmentation, feature extraction, andmachine learning-based classification. A unique approach is introduced that combines Enhanced Speckle ReducedAnisotropic Diffusion (SRAD) filters for speckle noise reduction, U-NET-based segmentation, Genetic Algorithm(GA)-based feature selection, and Random Forest and Bagging Tree classifiers, resulting in a novel and efficientmodel. To test and validate the hybrid model, rigorous experimentations were performed and results state thatthe proposed hybrid model achieved accuracy rate of 99.9%, outperforming other existing techniques, and alsosignificantly reducing computational time. This enhanced accuracy, along with improved sensitivity and specificity,makes the proposed hybrid model a valuable addition to CAD systems in breast cancer diagnosis, ultimatelyenhancing diagnostic accuracy in clinical applications.

Background:Physiological and biochemical processes across tissues of the body are regulated in response to the high demands of intense physical activity in several occupations,such as firefighting,law enforcement,military,and sports.A better understanding of such processes can ultimately help improve human performance and prevent illnesses in the work ***:To study regulatory processes in intense physical activity simulating real-life conditions,we performed a multi-omics analysis of 3 biofluids(blood plasma,urine,and saliva)collected from 11 wildland firefighters before and after a 45 min,intense exercise *** profiles ***-exercise were compared by Student’s t-test followed by pathway analysis and comparison between the different omics ***:Our multi-omics analysis identified and quantified 3835 proteins,730 lipids and 182 metabolites combining the 3 different types of *** blood plasma analysis revealed signatures of tissue damage and acute repair response accompanied by enhanced carbon metabolism to meet energy *** urine analysis showed a strong,concomitant regulation of 6 out of 8 identified proteins from the renin-angiotensin system supporting increased excretion of catabolites,reabsorption of nutrients and maintenance of fluid *** saliva,we observed a decrease in 3 pro-inflammatory cytokines and an increase in 8 antimicrobial peptides.A systematic literature review identified 6 papers that support an altered susceptibility to respiratory ***:This study shows simultaneous regulatory signatures in biofluids indicative of homeostatic maintenance during intense physical activity with possible effects on increased infection susceptibility,suggesting that caution against respiratory diseases could benefit workers on highly physical demanding jobs.

Capturing CO2 Emissions in the George C. Wallace Tunnel: A Case Study

This paper describes the design of a ventilation system to be paired with a carbon capture system. The ventilation system utilizes the geometry of the George C. Wallace tunnel, located in the City of mobile, Alabama, USA to capture and redirect emissions to a direct air capture (DAC) device to sequester 25% of the total CO2 mass generated from inside the tunnel. The total CO2 mass rate for the westbound traffic between the week-day hours of 7 a.m. and 6 p.m. has been estimated between 2,300 to 3,000 lbs./hr. By sequestering these emissions, the overall surrounding air quality was shown to be improved to a level that mirrors that from the pre-US industrial era of 270 ppm.
