Key factors in developing the trinitrobenzene sulfonic acid-induced post-inflammatory irritable bowel syndrome model in rats

AIM:To investigate the key factors in developing the trinitrobenzene sulfonic acid(TNBS)-induced postinflammatory irritable bowel syndrome(PI-IBS)model in rats. METHODS:TNBS was administered to rats at the following conditions:(1)with different doses(20,10,5 mg/0.8 mL per rat);(2)with same dose in different concentrations(20 mg/rat,25,50 mg/mL);(3)in different ethanol percentage(25%,50%);and(4)at depth either 4 cm or 8 cm from *** 5 d and 4 wk after TNBS administration,inflammation severity and inflammation resolution were *** 4 and 8 wk after TNBS application,visceral hyperalgesia and enterochromaffin(EC)cell hyperplasia were assayed by abdominal withdrawal reflex test,silver staining and capillary electrophoresis. RESULTS:Our results showed that:(1)TNBS induced dose-dependent acute inflammation and inflammation *** 5 d post TNBS,the pathological score and myeloperoxidase(MPO)activity in all TNBS treated rats were significantly elevated compared to that of the control(9.48±1.86,8.18±0.67,5.78± 0.77 vs 0,and 3.55±1.11,1.80±0.82,0.97±0.08 unit/mg vs 0.14±0.01 unit/mg,P<0.05).At 4 wk post TNBS,the pathological score in high and median dose TNBS-treated rats were still significantly higher than that of the control(1.52±0.38 and 0.80±0.35 vs 0,P<0.05);(2)Intracolonic TNBS administration position affected the persistence of visceral *** 4 wk post TNBS,abdominal withdrawal reflex (AWR)threshold pressure in all TNBS-treated groups were decreased compared to that of the control(21.52 ±1.73 and 27.10±1.94 mmHg vs 34.44±1.89 mmHg,P<0.05).At 8 wk post TNBS,AWR threshold pressure in 8 cm administration group was still significantly decreased(23.33±1.33 mmHg vs 36.79±2.29 mmHg,P<0.05);(3)Ethanol percentage affected the TNBS-induced inflammation severity and visceral *** TNBS-25%ethanol-treated group,the pathological score and MPO activity were significantly lowered compared to that of the TNBS-50%ethanoltreated group,while AWR thres

Horseshoe crabs,the most well-known example of“living fossils”,are iconic and ecologically important macroinvertebrates in coastal and estuarine *** blood is a crucial resource for manufacturing Limulus or Tachypleus amebocyte lysate to detect bacterial endotoxins or fungal contamination in drug and medical *** enhanced understanding of their ecological roles and trophic interactions in the food webs is fundamental to facilitate resource management for the declining populations in *** information of the Asian species,however,is mainly derived from preliminary,scattered reports from a limited number of study *** this study,resource utilization,trophic niche dynamics,and trophic interaction of the juvenile tri-spine horseshoe crab,Tachypleus tridentatus(instars 1–12,approximately 0.5–8 years old)across ontogeny was assessed in diversified nursery habitats along the northern Beibu Gulf,China,using carbon and nitrogen stable *** results suggest that:(i)*** are ecological generalists given the vast range of carbon isotopic values and trophic niche width estimates exhibited between multiple instar groups;(ii)juvenile *** across most habitat types predominantly assimilated energy from a variety of basal production sources in the food web,but primarily depended on sedimentary organic matter and seagrass resource pools;(iii)ontogenetic shifts in juvenile dietary proportions were evident,with decreased reliance on sedimentary organic matter,coupled with increased reliance on benthic macroinvertebrate grazers,detritivores,and omnivores with age;and(iv)nearly all juvenile instars occupied similar trophic positions in the food web with slight shifts in trophic position present with increasing *** findings indicate that resource availability and ontogenetic diet shifts strongly influence horseshoe crab trophic dynamics,and age should be accounted when formulating habitat conservation measures based on resour

Bevacizumab for neovascular age-related macular degeneration in Chinese patients in a clinical setting

AIM：To determine the outcome of non-investigational treatment with intravitreal bevacizumab（IVB） in neovascular age-related macular degeneration（AMD）patients.· METHODS：Retrospective chart review of 81 eyes with neovascular AMD followed-up for at least 12 mo and received 3-monthly loading IVB ***-treat was based upon the individual clinician＇s *** visual acuity（BCVA） and optical coherence tomography measurements of central foveal thickness outcomes were evaluated at 12,24 mo.· RESULTS：Eighty-one eyes（of 75 patients） completed12 mo of follow-up and 44 eyes（of 41 patients） completed24 mo of *** mean baseline logMAR BCVA significantly improved from 0.94±0.69 to 0.85±0.68 at12mo（P〈0.001） and from 0.91±0.65 to 0.85±0.60（P=0.004）at 24 *** proportion of eyes that lost 〈15 logMAR letters at 12 mo was 90.1%and at 24 mo was 81.8%.IVB was effective in improving visual acuity in both treatment naive and previous photodynamic therapy（PDT）-treated *** naive patients required significantly fewer injections than patients with prior *** regression analysis identified that poorer baseline visual acuity was associated with greater improvement in visual acuity（P=0.015）.· CONCLUSION：Fewer injections in clinical practice may result in suboptimal visual outcomes compared with clinical trials of IVB in neovascular AMD *** baseline visual acuity and prior PDT treatment may also improve vision after *** safety and durability of effect was maintained at 24 mo.

A Systematic Approach for Making 3D-Printed Patient-Specific Implants for Craniomaxillofacial Reconstruction

Craniomaxillofacial reconstruction implants,which are extensively used in head and neck surgery,are conventionally made in standardized *** surgery,the implant must be bended manually to match the anatomy of the individual *** bending process is time-consuming,especially for inexperienced ***,repetitive bending may induce undesirable internal stress concentration,resulting in fatigue under masticatory loading in v iv o and causing various complications such as implant fracture,screw loosening,and bone *** have been reports on the use of patient-specific 3D-printed implants for craniomaxillofacial reconstruction,although few reports have considered implant *** this paper,we present a systematic approach for making 3D-printed patientspecific surgical implants for craniomaxillofacial *** approach consists of three parts:First,an easy-to-use design module is developed using Solidworks®software,which helps surgeons to design the implants and the axillary fixtures for *** engineers can then carry out the detailed design and use finite-element modeling(FEM)to optimize the ***,the fabrication process is carried out in three steps:0 testing the quality of the powder;(2)setting up the appropriate process parameters and running the 3D printing process;and (3)conducting post-processing treatments(i.e.,heat and surface treatments)to ensure the quality and performance of the ***,the operation begins after the final checking of the implant and *** the surgery,postoperative rehabilitation follow-up can be carried out using our patient tracking *** this systematic approach,we have successfully conducted a total of 41 surgical cases.3D-printed patient-specific implants have a number of advantages;in particular,their use reduces surgery time and shortens patient recovery ***,the presented approach helps to ensure implant quality.

Lycium barbarum, a traditional Chinese anti-aging herb, has been shown to protect retinal ganglion cells （RGCs） in a rat chronic ocular hypertension （COH） model. Here, we investigated the expression of endothelin-1 （ET-1）, a strong vasoconstrictor, and its receptors, ETA and ETB, in the COH model and assessed the effects of Lycium barbarum on the ET-1 axis. Elevated intraocular pressure （IOP） was induced in the right eye of SD rats using argon laser photocoagulation. （1） The expression of ET-1, ETA and ETB in normal and COH retinas was studied. （2） Some COH rats were fed daily with Lycium barbarum Polysaccharides （LBP） using 1 mg/kg or phosphate-buffered saline （PBS） for 3 weeks （started 1 week before photocoagulation）. The effects of LBP on the expression of ET-1 and its receptors, ETA and ETB, in COH retina were evaluated. A semi-quantitative analysis of staining intensity was used to evaluate the expression levels of ET-1, ETA and ETB in retinal vasculature. We found that （1） Under COH condition, the immunoreactivity of ET-1 was increased in retina associated with an increase of ETB receptor immunoreactivity and a decrease of ETA receptor immunoreactivity. （2） After feeding COH rats with LBP, the expression of ET-1 was decreased with an increase of ETA expression and a decrease of ETB expression in the retina, especially in RGCs. （3） By comparing the staining intensity in the vasculature of COH retina in LBP-fed group with PBS-fed group, there was a decrease in the expression of ET-1 and ETA and an increase in ETB. In summary, ET-1 expression was up-regulated in the retina in COH model. LBP could decrease the expression of ET-1 and modulate the expression of its receptors, ETA and ETB, under the condition of COH. The neuroprotective effect of LBP on RGCs might be related to its ability to regulate the ET-1-mediated biological effects on RGCs and retinal vasculature.

Since the reform started inof 1978,China’s overall economic performance has been *** average annual GDP growth rate reached 9.43% in ***,in the last few years,China’s economic growth rate has been questioned.A deflation started to appear was evident at the end of *** spite of the Chinese government’s many efforts,the deflation has continued.A deflation in an economy in general is accompanied by stagnation or slow GDP ***,China’s GDP growth rate reached 7.68% annually during the deflation period in 1998-2002,which was the fastest growth rate in the world,.Moreover,the energy consumption dropped in 1998 and *** abnormality which prompted some economists to question the reliability of China’s *** the paper,I the author will analyze why it was was possible for China to maintain high growth with reduction of energy consumption during the deflation period and suggests the way for China to absorb the excess capacity and get out of the deflation.I The author will also discuss the prospect for China’s long-term growth.

Chinese Herbal Medicine for Chemotherapy Induced Gastrointestinal Side Effects: A Systematic Review of Randomized Controlled Trials

Objective: To determine how safe and effective Chinese Herbal Medicine(CHM) is in alleviating the nausea, vomiting, oral ulceration,diarrhea and constipation for cancer patients with ***: Data sources: A systematic review of Chinese and English articles using Ovid SP, CNKI, VIP Database and Traditional Chinese Medicine Database System. Study selection: Only randomized controlled trials(RCTs) for the prevention or treatment of any one of gastrointestinal side effects, namely nausea, vomiting, oral ulceration, diarrhea and constipation, of CHM with or without western medicine(WM) vs WM, placebo or no treatment were included. Data Extraction: Independent extraction of articles was first performed by four medical students using predefined data fields. Then, all data, including study quality indicators, was checked by two ***: Eighty-six RCTs involving 7076 cancer patients were found and analyzed in this review. Because of the heterogeneity of study design and low overall methodological quality, only descriptive summaries were performed. Beneficial effects were found in some CHM interventions,regardless of being taken alone or taken with WM. Moreover, none of serious adverse effect was reported. However, same intervention had not been repeatedly investigated by different research ***: Implications of the analysis support the efficacy and safety of CHM for the management of gastrointestinal side ***, definite clinical recommendation for particular CHM intervention still cannot be made due to low methodological quality of included studies and lack of duplicated verification. Further large scale and high quality RCTs on the same CHM interventions are suggested.

Positive-sense RNA viruses modify intracellular calcium stores,endoplasmic reticulum and Golgi apparatus(Golgi)to generate membranous replication organelles known as viral *** factories provide a conducive and substantial enclave for essential virus replication via concentrating necessary cellular factors and viral proteins in ***,we identified the vital role of a broadspectrum antiviral,peruvoside in limiting the formation of viral ***,we revealed the pleiotropic cellular effect of Src and PLC kinase signaling via cyclin-dependent kinase 1 signaling leads to Golgi-specific brefeldin A-resistance guanine nucleotide exchange factor 1(GBF1)phosphorylation and Golgi vesiculation by peruvoside *** ramification of GBF1 phosphorylation fosters GBF1 deprivation consequentially activating downstream antiviral signaling by dampening viral factories *** investigation showed signaling of ERK1/2 pathway via cyclin-dependent kinase 1 activation leading to GBF1 phosphorylation at Threonine 1337(T1337).We also showed 100%of protection in peruvoside-treated mouse model with a significant reduction in viral titre and without measurable cytotoxicity in *** findings highlight the importance of dissecting the broad-spectrum antiviral therapeutics mechanism and pave the way for consideration of peruvoside,host-directed antivirals for positive-sense RNA virus-mediated disease,in the interim where no vaccine is available.
