Identification of an immune-related gene signature for predicting prognosis and immunotherapy efficacy in liver cancer via cell-cell communication

BACKGROUND Liver cancer is one of the deadliest malignant tumors *** has provided hope to patients with advanced liver cancer,but only a small fraction of patients benefit from this treatment due to individual *** immune-related gene signatures in liver cancer patients not only aids physicians in cancer diagnosis but also offers personalized treatment strategies,thereby improving patient survival *** several methods have been developed to predict the prognosis and immunotherapeutic efficacy in patients with liver cancer,the impact of cell-cell interactions in the tumor microenvir-onment has not been adequately *** To identify immune-related gene signals for predicting liver cancer prognosis and immunotherapy *** Cell grouping and cell-cell communication analysis were performed on single-cell RNA-sequencing data to identify highly active cell groups in immune-related *** active immune cells were identified by intersecting the highly active cell groups with B cells and T *** significantly differentially expressed genes between highly active immune cells and other cells were subsequently selected as features,and a least absolute shrinkage and selection operator(LASSO)regression model was constructed to screen for diagnostic-related *** genes that were selected more than 5 times in 10 LASSO regression experiments were included in a multivariable Cox regression ***,3 genes(stathmin 1,cofilin 1,and C-C chemokine ligand 5)significantly associated with survival were identified and used to construct an immune-related gene *** The immune-related gene signature composed of stathmin 1,cofilin 1,and C-C chemokine ligand 5 was identified through cell-cell *** effectiveness of the identified gene signature was validated based on experi-mental results of predictive immunotherapy response,tumor mutation burden analysis,immune cell infiltrat

Unveiling the Re,Cr,and I diffusion in saturated compacted bentonite using machine-learning methods

The safety assessment of high-level radioactive waste repositories requires a high predictive accuracy for radionuclide diffusion and a comprehensive understanding of the diffusion *** this study,a through-diffusion method and six machine-learning methods were employed to investigate the diffusion of ReO_(4)^(−),HCrO_(4)^(−),and I−in saturated compacted bentonite under different salinities and compacted dry *** machine-learning models were trained using two *** dataset contained six input features and 293 instances obtained from the diffusion database system of the Japan Atomic Energy Agency(JAEA-DDB)and 15 *** other dataset,comprising 15,000 pseudo-instances,was produced using a multi-porosity model and contained eight input *** results indicate that the former dataset yielded a higher predictive accuracy than the *** gradient-boosting exhibited a higher prediction accuracy(R2=0.92)and lower error(MSE=0.01)than the other machine-learning *** addition,Shapley Additive Explanations,Feature Importance,and Partial Dependence Plot analysis results indicate that the rock capacity factor and compacted dry density had the two most significant effects on predicting the effective diffusion coefficient,thereby offering valuable insights.

Joint UAV trajectory and communication design with heterogeneous multi-agent reinforcement learning

Unmanned aerial vehicles(UAVs) are recognized as effective means for delivering emergency communication services when terrestrial infrastructures are unavailable. This paper investigates a multiUAV-assisted communication system, where we jointly optimize UAVs’ trajectories, user association, and ground users(GUs)’ transmit power to maximize a defined fairness-weighted throughput metric. Owing to the dynamic nature of UAVs, this problem has to be solved in real time. However, the problem’s non-convex and combinatorial attributes pose challenges for conventional optimization-based algorithms, particularly in scenarios without central controllers. To address this issue, we propose a multi-agent deep reinforcement learning(MADRL) approach to provide distributed and online solutions. In contrast to previous MADRLbased methods considering only UAV agents, we model UAVs and GUs as heterogeneous agents sharing a common objective. Specifically, UAVs are tasked with optimizing their trajectories, while GUs are responsible for selecting a UAV for association and determining a transmit power level. To learn policies for these heterogeneous agents, we design a heterogeneous coordinated QMIX(HC-QMIX) algorithm to train local Q-networks in a centralized manner. With these well-trained local Q-networks, UAVs and GUs can make individual decisions based on their local observations. Extensive simulation results demonstrate that the proposed algorithm outperforms state-of-the-art benchmarks in terms of total throughput and system fairness.

Convenient non-invasive flow monitoring would facilitate the operation and control in microfluidic chips,but is challenging due to the small space of microchannels and complex operation required in traditional optical *** this work,we propose a novel non-invasive strategy to probe microfluidic flows via streaming potential *** sealing one side of the microchannel with a piece of hydrogel film,streaming potential inside the channel can be clearly detected by electrodes at outer surface of the hydrogel due to ion diffusion in the *** is detected without sensors contacting with the internal ***,the electrodes shape like a tiny probe,which can move around mapping the flow distribution in a chip with the spatial resolution of 1 mm and flow rate detection limit of 3μL·min–*** inside the channels can also be detected,due to the fluctuation of streaming voltage when gas-liquid interface flows through the electrode,showing an easy and potential way for multi-functional flow monitoring in microfluidic chips.

Study of the reaction mechanism for preparing powdered activated coke with SO_(2)adsorption capability via one-step rapid activation method under flue gas atmosphere

In this study,the impact of different reaction times on the preparation of powdered activated carbon(PAC)using a one-step rapid activation method under flue gas atmosphere is investigated,and the underlying reaction mechanism is *** indicate that the reaction process of this method can be divided into three stages:stage I is the rapid release of volatiles and the rapid consumption of O_(2),primarily occurring within a reaction time range of 0-0.5 s;stage II is mainly the continuous release and diffusion of volatiles,which is the carbonization and activation coupling reaction stage,and the carbonization process is the main in this *** stage mainly occurs at the reaction time range of 0.5 -2.0 s when SL-coal is used as material,and that is 0.5-3.0 s when JJ-coal is used as material;stage III is mainly the activation stage,during which activated components diffuse to both the surface and interior of *** stage mainly involves the reaction stage of CO_(2)and H2O(g)activation,and it mainly occurs at the reaction time range of 2.0-4.0 s when SL-coal is used as material,and that is 3.0-4.0 s when JJ-coal is used as ***,the main function of the first two stages is to provide more diffusion channels and contact surfaces/activation sites for the diffusion and activation of the activated components in the third *** the reaction mechanism would serve as a crucial reference and foundation for designing the structure,size of the reactor,and optimal positioning of the activator nozzle in PAC preparation.

Legacy of herbicides in water from Hailun City,Northeast China:Occurrence,source,and ecological risk assessment

Herbicides(HBCs)are extensively used in modern ***,their potential negative impacts on environmental media have emerged as a significant environmental *** this study,we employed positivematrix factorization(PMF)to identify the potential sources of ***,we utilized amulti-matrix ecological riskmodel to assess the risks associated with HBCs in both surface water and groundwater in the black soil region of Northeast *** findings revealed that the levels of15HBCs in surface water and groundwater ranged from585.84 to 6466.96 ng/L and 4.80 to 11,774.64 ng/L,*** PMF results indicated that surface runoff and erosion accounted for 50%of the total HBCs in water,serving as the primary *** tested HBCs exhibited acute risk values within acceptable *** risk index for the∑15HBCs was categorized as“moderate risk”in 31%of the surface waters and 13%of the ***,4%of the groundwater sampling sites reached the“high risk”*** chronic risk quotient of∑15HBCs in surface water and groundwater was 92%and 62%at the“high risk”level,***,noncarcinogenic HBCs contributed more significantly to the ecotoxicology of the aquatic system than carcinogenic *** study provides comprehensive information on the legacy of HBCs in water bodies and emphasizes the potential risks posed by HBCs to aquatic *** results obtained from this study could help relevant management authorities in developing and implementing effective regulations tomitigate the ecological and environmental risks associated with HBCs.

Mirizzi syndrome is a serious complication of gallstone *** is caused by the impacted stones in the gallbladder neck or cystic *** of the features of Mirizzi syndrome is severe inflammation or dense fibrosis at the Calot’s *** our clinical practice,bile duct,branches of right hepatic artery and right portal vein clinging to gallbladder infundibulum are often observed due to gallbladder infundibulum adhered to right hepatic *** intraoperative damage of branches of right hepatic artery occurs more easily than that of bile duct,all of which are hidden pitfalls for *** resonance cholangiopancreatography(MRCP)and endoscopic retrograde cholangiopancreatography(ERCP)are the preferable tools for the diagnosis of Mirizzi *** cholecystectomy in Mirizzi syndrome is easy to damage branches of right hepatic artery and bile duct due to gallbladder infundibulum adhered to right hepatic *** cholecystectomy is an easy,safe and definitive approach to Mirizzi *** combined with the application of ERCP,a laparoscopic management of Mirizzi syndrome by well-trained surgeons is feasible and *** objective of this review was to highlight its existing problems:(1)low preoperative diagnostic rate,(2)easy to damage bile duct and branches of right hepatic artery,and(3)high concomitant gallbladder ***,the review aimed to discuss the possible therapeutic strategies:(1)to enhance its preoperative recognition by imaging findings,and(2)to avoid potential pitfalls during surgery.

Traditional therapies such as surgery and endocrine therapy no longer meet the clinical needs in prostate cancer treatment,and more effective treatments are urgently *** studies have reported that targeted inhibition of the transcription factor cyclin dependent kinase 7(CDK7)could effectively suppress prostate cancer ***,the toxicity of CDK7 inhibitors such as THZ1 is the main limitation of the clinical *** this work,we synthesized Cys8E(C8E)nanoparticles(NPs)loaded with THZ1(C8E@THZ1),a novel GSH-targeting and stimuli-responsive nano-delivery platform,and investigated its anti-tumor potential and biosafety *** vitro,C8E@THZ1 potently inhibited the proliferation and promoted the apoptosis of prostate cancer *** tumor-bearing mice,C8E@THZ1 inhibited tumors by up to 85%,while the damage of THZ1 to liver function was effectively *** results confirmed that inhibition of CDK7 can effectively block the progression of prostate cancer,and that Cys8E NPs is a highly prospective delivery platform to promote the clinical application of CDK7 inhibitors.

Microstructure and texture evolution of Cu-Cr-Co-Ti alloys during the two-stage cryorolling

This study entailed an investigation of the mechanical properties,microstructural and texture orientation evolutions of Cu-Cr-Co-Ti alloys prepared via twostage cryorolling and intermediate aging *** this end,X-ray diffraction and electron backscatter diffraction were *** results indicate that the two-stage cryorolling and intermediate aging treatments led to the development of profuse twin bundles and significantly enhanced the mechanical *** initial cryorolling led to coplanar slip and developed a strong Y({111})orientation,accelerating the formation of Goss({011})orientation and a Brass-type *** intermediate aging treatment relieved the restriction on dislocation slip and reoriented the grains toward the Copper({112})and Z({111})*** Z orientation,with a relatively high volume fraction,dominated the *** cryorolling intensified twinning and shear banding,transforming the Copper-type shear bands into Brass-type shear bands with rhomboidal prism *** areas inside the Brasstype shear bands exhibited a Y orientation,and the areas outside the shear bands exhibited a stable Brass-type *** evident decrease in the weighted Schmid factors demonstrated that the two-stage cryorolling and intermediate aging treatment can modify the texture evolution and aid the design of high-performance Cu alloys.
