First isolation and identification of H1N1 swine influenza viruses in Colombian pig farms

The pig industry in Colombia has grown 30% in the last decade achieving high levels of technology and efficiency;in spite of that, respiratory diseases remain a constraint. Since 1970, serological evidence and histological findings suggested the role of swine influenza virus (SIV) as part of the porcine respiratory disease complex;nevertheless, elusive and molecular typing isolates are missing. This study was aimed at isolating SIV from intensive pig farms and to achieve molecular characterization to determine strains circulating in the field. In order to accomplish this goal, 242 samples were taken from nasal swabs, 25 from bronchial washes and 8 from lung tissue. Samples were collected during a period of three years, between 2008 and 2010 and were originated from 78 farms of the three main pig production regions of the country. The samples were transported in BHI broth with 2% antibiotic and antimycotic solution and stored at –70?C until processed. The swabs were inoculated in 9 - 11 days old embryo chicken eggs and in MDCK (Madin Darby Canine Kidney) cell cultures with the addition of trypsin. The isolates were identified by the HA (hemoagglutination) test and by RT-PCR targeting the HA (hemagglutinin), NA (Neuraminidase) and M (Matrix) genes. Full length sequence of the HA and NA glycoproteins from four selected virus isolates was conducted (Macrogen?. USA). As a result, fifteen SIV isolates from nine farms distributed in the three regions were obtained. Twelve of the isolates are related to the swine origin H1N1 virus that caused the 2009 influenza pandemic. The remaining three viruses were related to classical swine influenza viruses.

Liver Transplantation in a Patient with Situs inversus Associated with Biliary Atresia, Polysplenia and Absence of Inferior Vena Cava: A Case Report

Biliary atresia (BA) is the result of a process of unknown etiology which can occur as an isolated lesion or in association with several congenital abnormalities and remains the main indications of liver transplantation among infants. One of the malformations associated with biliary atresia is Situs inversus totalis, which is a rare congenital abnormality found in 10% - 20% of infants with biliary atresia and presents an incidence varying from 1:5000 to 1:20,000. This case report aims to present an extremely rare case of a patient with Situs inversus associated with biliary atresia, polysplenia and abscence of inferior vena cava that underwent liver transplantation. Case Report: A 10-month-old boy was referred to our hospital with the diagnosis of cirrhosis, due to biliary atresia. Abdominal ultrasound and Doppler scan showed the liver in the left upper quadrant as well as signs of chronic liver disease. In addition, we could notice that the spleen had several lobes, suggesting polysplenia, and was positioned in the right upper quadrant. The patient underwent living donor liver transplant. His mother was the donor and the graft, segment III, was positioned in the left upper quadrant associated with a Y en Roux reconstruction of the biliary tract. During the first post-operatory day, the patient presented signs of disseminated sepsis. Laboratory and imaging diagnostic tests were performed presenting a small intestinal malrotation associated with diffuse ischemia. The patient died at the third day of PO due to complications related to this condition. Conclusion: Any patient with Situs inversus with acute abdomen should be aggressively investigated for other structural and biochemical abnormalities. If liver transplant is indicated, the procedure can be successfully performed.

Cancer is a global problem that in addition to physical, emotional and physiological causes economic and social impacts. The p53 gene is a tumor suppressor gene found in many malignant and benign tumors;this has the primary function of keeping cells at rest after damaging to DNA. The p53 acts in the maintenance of cellular homeostasis, mainly through autophagy, playing a role in cell cycle arrest, when necessary, thus avoiding mutated DNA replication. When in the oncogenic environment in many cases it is mutated, losing much of its efficiency allowing tumor development. Studies show that exercise can in the regular part of its pro-autophagic function even in the oncology setting. Stimuli of moderate-intensity aerobic and predominance of submaximal seem to trigger the protective function of p53 in various cancer settings. Among the many changes that these pathology triggers were the objective of this mini review is to relate the changes that exercise generates in p53 protein functions and their possible influence on tumor cells.

Dengue is one of the most prominent tropical epidemic diseases present in the Rio de Janeiro city and Southeast part of Brazil, due to the widespread conditions of occurrence of the dengue vector, the mosquito Aedesaegypti, such as high-temperature days interlaced with afternoon or nocturnal rainstorms in summer. This work has the objective of investigating the relationships between variabilities of the El Ni?o-South Oscillation (ENSO) and greater epidemics of dengue in Rio de Janeiro city. To accomplish this goal, the analysis and signal decomposition by cross-wavelet transform (WT) was applied to obtain the cross variability associated with variations of power and phase of both signals by characteristic periods and along with the time series. Data considered in the analysis are (the decimal logarithm of normalized value) of the monthly available notifications of dengue worsening, provided by the public health system of Brazil, and the Southern Oscillation Index (SOI) Ni?o 3.4 data, provided by the National Oceanic and Atmospheric Administration (NOAA), in the period 2000-2017. A maximum cross-wavelet power close to 0.45 was obtained for the representative period of 1 year and also to periods between 3 and 4 years, associated with the positive phase of the SOI index (i.e. , La Ni?a) or with a transition to the positive phase. The evolution of the combined variability of SOI and dengue can be expressed by progressive differences in phase along the time, eventually resulting in yielding phases (i.e., La Niña-Dengue epidemic).

Dermatoscopy in the Diagnosis of Vulvar Basal Cell Carcinoma:A Case Report

Introduction: Basal cell carcinoma (BCC) is the most common human malignancy commonly in white people, but in less than 1% of cases it may appear on unexposed areas, like the perineal and anal regions. Vulvar BCC is often diagnosed late because it grows slowly and tends to be asymptomatic, with no specific physical findings. Here, we present a rare Latin patient with a 10-year history of vulvar BCC with dermatoscopic and histopathologic *** report: A 65-year-old Latin woman presented with a nodule on her left labium majus, which she first noted 10 years ago, and that had grown slowly but constantly. She also reported a 20-year history of pruritus and occasional pain. Physical examination showed a 3.5 cm × 3.0 cm erythematous lesion with a central rough without inguinal lymphadenopathy. Contact dermatoscopy (Dermalite DL3 Gen.) showed one blurred cluster of arborising vessels in a pinkish background with white homogeneous areas and lines radial converging. An incisional biopsy was realized, and histopathology diagnosed ***: Vulvar BCC is a rare malignancy that affects mainly Caucasian women over 70 years of age. The most common etiology for BCC is ultraviolet radiation, but as the vulva is unexposed its cause is unclear. Mutations in tumor suppressor and regulatory genes such as p53 are present in 50% of cases. Gorlin syndrome, chronic radiation, chronic arsenic exposure, xeroderma pigmentosum, and immunosuppression have been considered as risk ***: Vulvar BCC characteristics are the same as other cutaneous forms, featuring blue ovoid nests and arborizing telangiectasia, confused with inflammatory diseases, such as eczema, psoriasis, and chronic infection (especially if it is accompanied by pruritus), this makes diagnosis complex, with a delay of 5 to 6 years on average, with an average size of 2.1 cm. Diagnosis is enhanced with dermatoscopy, as the preferred treatment for most BCC cases is wide surgical excision.

Chemical pesticides tend to accumulate in soil, resulting in human and environmental health risks. Hence, alternative methodologies involving chemical pesticides are beneficial for the control of agricultural pests. Metarhizium anisopliae is an entomopathogenic fungus that acts on different developmental stages of pest insects such as Diatraea saccharalis, a holometabolic lepidopteran with high potential for infestation in sugarcane crops. The present study evaluated the biocontrol effect of M. anisopliae isolates MT and E9 on D. saccharalis eggs at different ages by investigating the external and internal morphological alterations in treated eggs. Conidial suspensions of M. anisopliae isolated from MT and E9 at concentrations of 107 conidia/mL were applied to eggs of D. saccharalis aged 0, 24, 48, 72, 96 and 120 h. The eggs were observed every 24 h during development (0 h to 144 h). Samples were collected for observational, histological, and ultrastructural analyses. We found that the MT isolate caused 100% inviability of eggs aged 0 - 72 h, 144 h after the bioassays, while the effect of the E9 isolate varied between 49.40% and 93.75%. M

Metals obtain optimum conditions of metallurgical and physical properties through a heat treatment. Brass is one of the copper alloys which has many applications in everyday life and in the industry. In this work, the influence of the precipitation heat treatment temperature on the metallurgical microstructure, structure, thermal properties, and microhardness of an alpha brass is analyzed. Samples were heat treated by precipitation for 2 h at 300°C, 400°C, 500°C, 600°C, and 700°C. The best mechanical properties were found at 500°C of precipitation heat treatment temperature. Specimens were characterized by scanning electron microscopy, X-ray diffraction, Vickers microhardness, photothermal radiometry, and photoacoustic to study the thermal diffusivity and conductivity, as well as the heat capacity. The inverse of the full width at the half maximum analysis showed that the crystallinity decreased as the precipitation heat treatment temperature increased. Metallurgical microstructure and microhardness were correlated to the precipitation heat treatment temperatures to determine the effect on the metallurgical and mechanical properties, as well as the effect on the thermal properties of alpha brass.

Fast Determination of Benzodiazepines in Human Urine via Liquid-Liquid Extraction with Low Temperature Partitioning and LC-HRMS

A simple and high-throughput method to simultaneously determine selected benzodiazepines (i.e., diazepam, lorazepam, clonazepam, and bromazepam) in urine was developed and validated. The entire methodology consisted of the application of an innovative extraction/cleanup procedure, namely liquid-liquid extraction with low-temperature partitioning (LLE-LTP), and analysis by liquid chromatography combined with high-resolution mass spectrometry (LC-HRMS). The LLE-LTP procedure was optimized via factorial design and by evaluating crucial variables, specifically the freezing mode (either slow or fast), the urine/acetonitrile volume ratio, and the sample ionic strength. The benzodiazepines were quantified using matrix-matched calibration curves where the following parameters were assessed by validation protocol: in general, linearity range of 17 - 200 μg?L–1 (r > 0.9957);limits of detection lower than 5 μg?L–1;relative standard deviations (RSD) lower than 12.5%;and accuracy ranging from 72.3 to 117%. To test this procedure’s performance, the method was applied to determine the content of diazepam in actual urine samples. The validation results obtained for the method demonstrated that the present methodology could be potentially applied in proficient laboratories as a routine approach for determining benzodiazepines compounds content in urine.

Existing therapies for the treatment of chronic heart failure still have some limitations and there is a pressing need for the development of new therapeutic modalities. The amniotic membrane has been used for the treatment of various diseases, such as conjunctive defects;however, the mechanisms behind its repair functions are still unclear. Regenerative medicine is seeking newer alternatives and among them, biomaterials have emerged in recent years for developing and manipulating molecules, cells, tissues or organs grown in laboratories in order to replace human body parts. Many such materials have been used for this purpose, either synthetically or biologically, in order to provide new medical devices. This review provides a wider view of the regeneration potential of the use of amniotic membrane as a potential biomaterial to facilitate the implementation of new research in surgical procedures. Amniotic membrane appears to be an alternative source of stem cells as well as an excellent biomaterial for cell-based therapeutic applications in engineering heart tissue.
