Relationship of Polymorphisms of Glutathione S-Transferase GSTT1 and GSTM1 With the Response to Chemotherapy In Mexican Women with Advanced Breast Cancer

Introduction: Breast cancer is a common disease diagnosed in Mexican women and the first leading cause of death [1]. Heterogeneity in patients’ response to treatment is consistently observed across populations. Glutathione S-transferases (GSTs) are involved in the metabolism of environmental carcinogens, reactive oxygen species and chemotherapeutic agents by catalyzing the glutathione with electrophilic compounds. The deletion of GSTT1 and GSTM1 genes result in loss of enzyme activity. A few studies evaluated the response to treatment and the polymorphisms of GSTT1 and GSTM1. The aim of this work is to make the association of the null polymorphisms of GSTT1 and GSTM1 with the response to chemotherapy basically doxorubicin and cyclophosphamide. Methods: The genotyping of thirty patients with breast cancer was made with the Polymerase chain reaction, to identify the polymorphisms of GSTT1 and GSTM1. We determine the status of Her-2 neu, estrogen and progesterone receptors, then the response to treatment was made with an ultrasound and pathological data. We made the association with the χi2 statistics using a p≤0.05. Results: Using the Sigma Stat 3.5 program and the chi-squared analysis, we do not observe a significant association with the GSTT1+/GSTM1+, GSTT1-/GSTM1+ and GSTT1-/GSTM1-polymorphisms and the better or worse response to cyclophosphamide and doxorubicin. With the Her-2 neu, estrogen and progesterone receptors status, we neither found an association with the response to the therapy. Conclusion: This study suggests that GSTT1 and GSTM1 polymorphisms have no statistical significance between the genotype of women with advanced breast cancer and the response to neoadjuvant chemotherapy, but we can see a clear tendency toward better response with the null genotype.

effect of Unilateral Low-Frequency Stimulation of Hippocampus on Rapid Kindling—Induced Seizure Development in Rats

Since the last decade deep brain stimulation has been proposed as an alternative treatment for patients who do not become seizure-free with the current pharmacological treatments and cannot undergo resective surgical procedure. However, the optimal stimulation parameters remain undetermined and active research in humans and animals is necessary. The present study was designed to investigate the effect of unilateral Low Frequency Stimulation (LFS) of hippocampus on seizure development by using the hippocampal rapid kindling method (hRK) in rats. We used male Wistar rats implanted with electrodes in the ventral hippocampus. All rats underwent hRK (biphasic square wave pulses, 20 Hz for 10 seconds) during three consecutive days (twelve stimulations per day). The control group (hRK;n = 6) received only RK stimulus, while the treated group (LFS-hRK;n = 8) received also LFS (biphasic square wave pulses, 1 Hz for 30 seconds) immediately before the RK stimulus, during three consecutive days. At the end of behavioral testing on day 3, 62% (P e animals receiving LFS treatment were still not fully kindled staying in stages 0-III (P e number of stimulations needed to achieve generalized seizures (stage IV-V of Racine scale) was significantly higher (P e LFS group with respect to control group. No significant differences in the cumulative daily afterdischarge duration were observed between both groups. These findings suggest that preemptive LFS can significantly decrease the incidence of hippocampus-kindled seizures and delay the progression and secondary generalization of focal seizures.

Objective:To identify the prevalence of soil-transmitted helminthiasis(STH)among school-age children in the Cagayan Valley,the Philippines,assess their level of awareness on the disease,and determine predisposing factors of the ***:A total of 478 GradesⅢ-Ⅴschool-age children in Pamplona and Sanchez-Mira School Districts in the Cagayan Valley answered the questionnaire assessing their knowledge,attitude,and practices on STH,subjected to anthropometric measurements,and provided faecal samples for parasitologic assessment(direct smear,Kato-Katz,and formol-ether concentration techniques).Results:The participants of the study,with 55.86%females,were 8 to 14 years *** nutritional status was assessed‘normal’(84.31%),‘severely wasted’(6.49%),‘wasted’(5.23%),‘overweight’(2.72%),and‘obese’(1.26%).The prevalence of infection with at least 1 STH species was 25.99%in Pamplona and 19.40%in ***,the prevalence of heavy intensity was 7.11%for Ascaris lumbricoides and 1.67%for Trichuris *** hookworm infections had light *** majority of the school-age children had a low score in the KAP *** knowledge of STH,‘stunted growth as a symptom of infection’was associated with a lower risk of Ascaris lumbricoides infection(OR 0.448;95%CI 0.212,0.945;P=0.035)while‘playing with soil as a mode of transmission’was associated with an increased risk of Ascaris lumbricoides infection(OR 2.067;95%CI 1.014,4.212;P=0.046).In attitude towards STH,‘I think I have intestinal worm now’was associated with a higher risk of Ascaris lumbricoides infection(OR 1.681;95%CI 1.061,2.662;P=0.027).Conclusions:The prevalence rate of Ascaris lumbricoides among the school-age children in the Cagayan Valley shows the need to further intensify intervention in the area to meet the threshold set by the World Health *** identified predictors of infection,which concerns the school-age children’s knowledge and attitude toward STH,can be used in augmenting

Background:Access to clean and safe drinking water that is free from pathogenic protozoan parasites,especially Cryptosporidium parvum and Giardia lamblia that cause gastrointestinal illness in humans,is still an issue in Southeast Asia(SeA).This study is the first attempt to detect the aforementioned protozoan parasites in water samples from countries in SeA,using real-time polymerase chain reaction(qPCR)***:A total of 221 water samples of 10 l each were collected between April and October 2013 from Malaysia(53),Thailand(120),the Philippines(33),and Vietnam(15).A physicochemical analysis was *** water samples were processed in accordance with the US environmental Protection Agency’s methods 1622/1623.1,microscopically observed and subsequently screened using qPCR ***:Cryptosporidium oocysts were detected in treated water samples from the Philippines(1/10),with a concentration of 0.06±0.19 oocyst/L,and untreated water samples from Thailand(25/93),Malaysia(17/44),and the Philippines(11/23),with concentrations ranging from 0.13±0.18 to 0.57±1.41 oocyst/*** cysts were found in treated water samples from the Philippines(1/10),with a concentration of 0.02±0.06 cyst/L,and in untreated water samples from Thailand(20/93),Vietnam(5/10),Malaysia(22/44),and the Philippines(16/23),with concentrations ranging from 0.12±0.3 to 8.90±19.65 cyst/*** pathogens *** and *** were detected using using qPCR assays by targeting the 138-bp fragment and the small subunit gene,*** was detected in untreated water samples from the Philippines(1/23)and Malaysia(2/44),whilst,*** detected was detected in treated water samples from the Philippines(1/10)and in untreated water samples from Thailand(21/93),Malaysia(12/44),and the Philippines(17/23).Nitrate concentration was found to have a high positive correlation with(oo)cyst(0.993).Conclusion:The presence of(oo)cysts in the water samples means that there is potential risk for

Clinical Facial Study of the Parents of Children with OAV (Oculo-Auriculo-Vertebral) Syndrome. Cases and Controls

Introduction: The oculo-auriculo-vertebral syndrome is a complex disease of the head and neck. Aim: The purpose of this study was to look for facial asymmetry, measuring both sides of the mandible and external ear in the parents of these patients in a clinical way, to see if it could be considered as a minor manifestation of the disease. Patients and Methods: A clinical, observational study was done with 50 trios and their parents’ controls, at the Instituto Nacional de Rehabilitación LGII in 2019, from January to May. Mandible and ear measurements were done. Results: All of our patients had microtia and facial asymmetry, and in 17 cases, other anomalies. We compared both sides of the mandible in the patients’ parents and in the parents’ controls. The results showed differences only in the fathers, and in the comparison with the controls, we found statistical differences only in women, family and non-family cases. The ears did not show any differences. Discussion: It is very interesting that we found differences in the measurements of the mandible of the patients’ mothers and their controls;in the mothers the mandible was bigger than in the controls, this probably means that the development of the cranial structures is modified by some genetic variants. No differences were found in the measurements of the ears. Conclusion: It is very difficult to know exactly if the differences we found are important or not. The clinical measurements could be a good option to establish if the parents have minor manifestations that could explain some heritability.

Gonadal determination in marine turtles depends on incubation temperature. The mechanisms that spark off this process remain unclear. Previously, we proposed that sensory nerves reaching the gonadal primordium in nominal female embryos of Lepidochelys (L) olivacea may sense and signal incubation temperature. These nerves could later trigger ovarian determination by releasing neurotransmitters in a code constructed based on the thermal information (Gutierrez-Ospina et al., Acetylcholinesterase-positive innervation is present at the undifferentiated stages of the sea turtle Lepidochelys olivacea embryo gonads: implications for temperature-dependent sex determination, J. Comp. Neurol. 410 (1999) 90-98). The hypothesis briefly described, however, has been recently refuted under weak theoretical grounds and experimental misinterpretations (see introduction). Here, we present preliminary results that show that nominal female embryos have sensory neurons located in the dorsal horn laminae I and II of the lumbar spinal cord that display increased c-Fos-like immuno-staining after being incubated either at 15°C or 50°C. Because these spinal neurons are the primary central target of dorsal root ganglion neurons that innervate the urogential crest, these observations keep open the possibility that gonadal sensory nerves indeed signal thermal information that could later be used to trigger or instruct ovarian specification in marine turtles.

目的 :探讨钠氢离子拮抗剂 Cariporide在延迟心室纤颤大鼠心肺复苏中的作用。方法 :将 2 0只延迟的心室纤颤(10分钟 )动物模型的大鼠随机分为 2组 (Cariporide治疗组及对照组 )。在心肺复苏后 6分钟行试验电除颤 (2 J,2 J,4J) ,对未能转复者继续心肺复苏 2分钟后行补救电除颤 -心肺复苏系列。所有复苏动物复苏后观察 2小时。结果 :与对照组比较 ,Cariporide治疗组以较小的心脏按压高度、较少的电除颤次数、较低的除颤能量 ,获得较高的实验转复率、较低的复苏后心律失常发生率、较好的复苏后血流动力学结果及较长的生存时间 (P <0 .0 5 )。结论 :钠氢离子拮抗剂应用于心室纤颤的复苏可提高复苏效果 ,减少心律失常的发生并改善生存。

目的 :探讨对由延迟的心室纤颤引起的心脏骤停的治疗方法。方法 :将 32只延迟心室纤颤 (10分钟心室纤颤 )动物模型的大鼠随机分为 4组 (0 -min组 ,2 -min组 ,4-min组及 6 -min组 ) ,分别在心肺复苏前、心肺复苏后 2分钟、4分钟、6分钟行试验电除颤 ,对未能转复者继续心肺复苏及补救电除颤。结果 :0 -min组 ,2 -min组无大鼠能通过试验电除颤成功恢复自主心率 ,而 6 -min组有 7只大鼠 ;与另外两组相比 ,4-min组及 6 -min组具有更长的生存时间。此外 ,6 -min组恢复自主心率所需的试验电除颤次数、补救电除颤次数及总除颤能量均显著地较其他三组为低。结论 :我们认为对延迟的心室纤颤在电除颤前应进行至少 4～ 6分钟的心肺复苏。

目的　探讨联合应用钠氢离子交换抑制剂(NHe I)Cariporide与肾上腺素在心肺复苏中的效果。方法　 16只历经 12min室颤的大鼠配对分为 2组 (Cariporide组及对照组 ) ,分别在心肺复苏 (CPR)前注入Cariporide或等容量的生理盐水。CPR第 2min结束前始注入肾上腺素 ,使CPP维持在 2 5mmHg以上。对照组与匹配的Cariporide组有对应的除颤时间。对心肺复苏 8min后未能自动复率大鼠进行电除颤。结果　与对照组相比 ,以同样的高度进行胸前心脏按压 ,cariporide组须较小剂量的肾上腺素便可获得较高、持续时间较长的冠脉灌注压 (P e及肾上腺素可提高心室纤颤的复苏效果。
