background:Very low-protein(VLP)diets negatively impact calcium(Ca)metabolism and *** objective of this study was to investigate the effect of supplemental branched-chain amino acids(bCAA)and limiting amino acids(LAA)on Ca digestibility,absorption and reabsorption in pigs fed with VLP ***-eight piglets were assigned to six treatments:positive control(PC),negative control(NC),and NC containing LAA 25%,LAA 50%,LAA+bCAA 25%(Lb25)and LAA+bCAA 50%(Lb50)more than ***:Relative to PC or NC,Lb25 and Lb50 had higher digestibility of Ca and plasma Ca and phosphorus(P),but lower plasma vitamin ***50 tended to increase vitamin D receptor transcript and protein in the gut,but decreased mRNA or protein abundance of parathyroid hormone 1 receptor(PTH1R),calbindin 1(CALb1),cytochrome P450 family 27 subfamily b member 1 and occludin in small ***50 increased the transcript of cytochrome P450 family 24 subfamily A member 1 and PTH1R but decreased the transcript of transient receptor potential cation channel subfamily V member 5,CALb1 and solute carrier family 17 member 4 in ***:Overall,bCAA increased Ca digestibility through regulating the transcellular and paracellular Ca absorption in the gut and reabsorption in kidney during protein restriction.

To study the local and systemic effects of arbuscular mycorrhizal fungal （AMF） colonization, Nicotiana attenuata plants impaired in their interactions with AMF due to silencing of a calcium- and calmodulin dependent protein kinase （inverted repreat （ir）CCaMK） were grown competitively in pairs with empty vector （EV） plants, with and without two different types of inoculum. When inoculated, EV plants strongly outperformed irCCaMK plants. Foliar transcript profiling revealed that AMF colonization significantly changed gene expression of P-starvation and -transporter genes in irCCaMK plants. The Phtl family phosphate transporter NaPT5 was not only specifically induced in roots after AMF colonization, but also in leaves of AMF-colonized irCCaMK plants, and in plants grown under low Pi conditions in the absence of AMF. The P-starvation signature of inoculated irCCaMK plants corresponded with increases in selected amino acids and phenolic compounds in leaves. We also found a strong AMF-induced increase in amino acids and phenolic metabolites in roots. Plants impaired in their interactions with AMF clearly have a fitness disadvantage when competing for limited soil nutrients with a fully functional isogenic line. The additional role of the AMF-induced Phtl family transporter NaPT5 in leaves under P-starvation conditions will require further experiments to fully resolve.

Long-term monitoring programs for measurement of atmospheric mercury concentrations are presently recognized as powerful tools for local,regional and global studies of atmospheric long-range transport processes,and they could also provide valuable information about the impact of emission controls on the global budget of atmospheric mercury,their observance and an insight into the global mercury cycle. China is believed to be an increasing atmospheric mercury emission source. However,only a few measurements of mercury,to our knowledge,have been done in ambient air over China. The highly-time resolved atmospheric mercury concen-trations have been measured at Moxi base Station (102°72′E 29°92′N,1640 m asl) of the Gongga Alpine Ecosystem Observation and Experiment Station of Chinese Academy of Sciences (CAS) from May 2005 to June 2006 by using a set of Automatic Atmospheric Mercury Speciation Analyzers (Tekran 2537A). Measurements were carried out with a time resolution of every 15 minutes. The overall average total gaseous mercury (TGM) covering the measurement periods was 4±1.38 ng·m-3 (N=57310),which is higher than the global background level of approximately 1.5～2.0 ng·m-3. The measurements in all seasons showed a similar diurnal change pattern with a high concentration during daytime relative to nighttime and maximum concentration near solar noon and minimum concentration immediately before sunrise. The presence of diurnal TGM peaks during spring and summer was found earlier than that during autumn and winter. When divided seasonally,it was found that the concentrations of TGM were highest in winter with 6.13 ± 1.78 ng·m-3 and lowest in summer with 3.17 ± 0.67 ng·m-3. There were no significant differences in TGM among wind sectors during each season. Whereas Hg generally exhibited significant correlations with the parameters,such as temperature,saturated vapor pressure,precipitation,ultraviolet radiation (UV) and atmospheric pressure at the whole measurement stage,

This study presents direct 2D and 3D co-culture model of mesenchymal stem cells(MSCs)line with chondrocytes isolated from patients with osteoarthritis(unaffected area).MSCs differentiation into chondrocytes after 14,17 days was checked by estimation of collagen Ⅰ,Ⅱ,Ⅹ,aggrecan expression using ***,localization of cells on Hyaff-11 was performed using enzymatic technique and THUNDER Imaging *** showed,that MSCs/chondrocytes 3D co-culture induced suitable conditions for chondrocytes grow and MSCs differentiation than 2D *** that differentiated cells on Hyaff-11 expressed collagen Ⅹ,they showed high collagen Ⅱ(80%)and aggrecan(60%)expression with simultaneous decrease of collagen Ⅰ expression(10%).The high concentration of differentiated cells on Hyaff-11,indicate that this structure has an impact on cells cooperation and *** conclusion,we suggest that high expression of collagen Ⅱ and aggrecan in 3D co-culture model,indicate that cooperation between different subpopulations may have synergistic impact on MSCs chondrogenic *** the high concentration and localization of cells growing in deeper layers of membrane in 3D co-culture,indicate that induced microenvironmental enhance cell migration within ***,we suggest that co-culture model might be useful for construction a bioactive structure for cartilage tissue regeneration.

Impact of comorbidity on waiting list and post-transplant outcomes in patients undergoing liver retransplantation

AIM To determine the impact of Charlson comorbidity index(CCI) on waiting list(WL) and post liver retransplantation(LRT) *** Comparative study of all adult patients assessed for primary liver transplant(PLT)(n = 1090) and patients assessed for LRT(n = 150), 2000-2007 at our centre. Demographic, clinical and laboratory variables were recorded. RESULTS Median age for all patients was 53 years and 66% were men. Median model for end stage liver disease(MELD) score was 15. Median follow-up was 7- years. For retransplant patients, 84(56%) had ≥ 1 comorbidity. The most common comorbidity was renal impairment in 66(44.3%). WL mortality was higher in patients with ≥ 1 comorbidity(76% vs 53%, P = 0.044). CCI(OR = 2.688, 95%CI: 1.222-5.912, P = 0.014) was independently associated with WL mortality. Patients with MELD score ≥ 18 had inferior WL survival(LogRank 6.469, P = 0.011). On multivariate analysis,CCI(OR = 2.823, 95%CI: 1.563-5101, P = 0.001), MELD score ≥ 18(OR 2.506, 95%CI: 1.044-6.018, P = 0.04), and requirement for organ support prior to LRT(P ber of transplanted grafts(Log-Rank 18.455, P ferior to those of PLT at 88% vs 73%, P bidity increases WL and post-LRT mortality. Patients with MELD ≥ 18 have increased WL mortality. Patients with comorbidity or MELD ≥ 18 may benefit from earlier LRT. LRT for ≥ 3 grafts may not represent appropriate use of donated grafts.

background： The goal of this study was to compare the antagonism of elevated dietary Cu （250 mg/kg） from CuS04 on three different Zn sources （ZnS04. H20; [Zn bis（-2-hydroxy-4-（methylthio）butanoic acid）], Zn（HMTba）2 a chelated Zn methionine hydroxy analogue; and Zn-Methionine）, as measured using multiple indices of animal performance in ROSS 308 broilers. Methods： Three experiments were conducted in broiler chicks fed a semi-purified diet. All birds were fed a Zn-deficient diet （8.5 mg/kg diet） for 1 wk, and then provided with the experimental diets for 2 wks. Results： Experiment 1 was a 2 x 2 factorial design with two levels of Cu （8 vs. 250 mg/kg diet from CuSO4） and two Zn sources at 30 mg/kg [ZnSO4. H20 vs. Zn（HMTba）2]. Elevated Cu impaired growth performance only in birds fed ZnSO4. Compared to ZnSO4. H20, Zn（HMTba）2 improved feed intake （12 %; P 〈 0.001） and weight gain （12 %, P 〈 0.001） and the benefits were more pronounced in the presence of 250 mg/kg diet Cu. Experiment 2 was a dose titration of ZnSO4- H20 and Zn（HMTba）2 at 30, 4,5, 60, and 75 mg/kg diet in the presence of 250 mg/kg CuSO4. feed：gain was decreased and tibia Zn was increased with increasing Zn levels from 30 to 75 mg/kg. birds fed Zn（HMTba）2 consumed more food and gained more weight compared to birds fed ZnSO4, especially at lower supplementation levels （30 and 45 mg/kg; interaction P 〈 0,05）. Experiment 3 compared two organic Zn sources （Zn（HMTba）2 vs. Zn-Methionine） at 30 mg/kg with or without 250 mg/kg CuSO4. No interactions were observed between Zn sources and Cu levels on performance or tissue mineral concentrations. High dietary Cu decreased weight gain （P 〈 0.01）. Tibia Cu and liver Cu were significantly increased with 250 mg/kg dietary Cu supplementation （P 〈 0.01）. No difference was observed between the two Zn sources. Conclusions： Dietary 250 mg/kg Cu significantly impaired feed intake and weight gain in birds fed ZnSO4. H20, but had less impact in birds fed Zn（HMTba）2. No difference

Dear Editor, Flavonols are synthesized by flavonol synthase （FLS） enzymes （Martens et al., 2010）. These compounds absorb UV-b light in the 280-320-nm region and their concentration increases in plants exposed to environmental abiotic and biotic stresses, including UV-b; consequently, flavonols are thought to act as UV-b filters （Agati et al., 2011）. It has been also suggested that these metabolites function as reactive oxygen species （ROS） scavengers, as they contain an OH- group in the 3-position of the flavonoid skeleton, which allows them to chelate metals, inhibiting the formation of free radicals and ROS accumula- tion, once formed （Agati et al., 2009）. For these reasons, it has been suggested that flavonols play uncharacterized roles in UV responses （Verdan et al., 2011）. Nevertheless, despite the fact that the role of flavonols in UV-b protection has been inferred, the protection conferred by flavonols on the target sites of UV-b-damage has not been directly proven in planta.

Is hepatic steatosis associated with left ventricular mass index increase in the general population?

AIM To investigate the association between hepatic steatosis and change in left ventricular mass index(LVMI) over five years, and examine whether systolic and diastolic blood pressures are mediators of the association between hepatic steatosis and LVMI using a general population *** We analyzed data from the Study of Health in Pomerania. The study population comprised 1298individuals aged 45 to 81 years. Hepatic steatosis was defined as the presence of a hyperechogenic pattern of the liver together with elevated serum alanine transferase levels. Left ventricular mass was determined echocardiographically and indexed to height2.7. Path analyses were conducted to differentiate direct and indirect paths from hepatic steatosis to LVMI encompassing systolic and diastolic blood pressure as potential mediating *** Hepatic steatosis was a significant predictor for all measured echocardiographic characteristics at baseline. Path analyses revealed that the association of hepatic steatosis with LVMI change after five years was negligibly small(β =-0.12, s.e. = 0.21, P = 0.55). Systolic blood pressure at baseline was inversely associated with LVMI change(β =-0.09, s.e. = 0.03, P between diastolic blood pressure at baseline and LVMI change was evident(β = 0.03, s.e. = 0.05, P = 0.56). The effect of the indirect path from hepatic steatosis to LVMI via systolic baseline blood pressure was small(β =-0.20, s.e. = 0.10, P = 0.07). No indirect effect was observed for the path via diastolic baseline blood pressure(β = 0.03, s.e. = 0.06, P = 0.60). Similar associations were observed in the subgroup of individuals not receiving beta-blockers, calcium channel blockers, or drugs acting on the reninangiotensin *** baseline associations between hepatic steatosis and LVMI do not extend to associations with LVMI change after five years. More studies are needed to study the longitudinal effects of hepatic steatosis on LVMI.
