An evaluation of garlic lectin as an alternative carrier domain for insecticidal fusion proteins

The mannose-binding lectin GNA （snowdrop lectin） is used as a＂carrier＂ domain in insecticidal fusion proteins which cross the insect gut after oral ingestion. A similar lectin from garlic bulb, ASAII, has been evaluated as an alternative ＂carrier＂. Recombinant ASAII delivered orally to larvae of cabbage moth （Mamestra brassica; Lepidoptera） was subsequently detected in haemolymph, demonstrating transport. Fusion proteins comprising an insect neurotoxin, ButaIT （Buthus tamulus insecticidal toxin; red scorpion toxin） linked to the C-terminal region of ASAII or GNA were produced as recombinant proteins （GNA/ ButaIT and ASA/ButaIT） by expression in Pichia pastoris. In both cases the C-terminal sequence of the lectin was truncated to avoid post-translational proteolysis. The GNA- containing fusion protein was toxic by injection to cabbage moth larvae （Ld50≈ 250μg/g）, and when fed had a negative effect on survival and growth. It also decreased the survival of cereal aphids （Sitobion avenae; Homoptera） from neonate to adult by 〉70% when fed. In contrast, the ASA-ButaIT fusion protein was non-toxic to aphids, and had no effect on lepidopteran larvae, either when injected or when fed. However, intact ASA-ButaIT fusion protein was present in the haemolymph of cabbage moth larvae following ingestion, showing that transport of the fusion had occurred. The stabilities of GNA/ButaIT and ASA/ButaIT to proteolysis in vivo after injection or ingestion differed, and this may be a factor in determining insecticidal activities.

Food-derived peptides with inhibitory capacity for HMG-CoA reductase activity: a potential nutraceutical for hypercholesterolemia

Cardiovascular diseases (CVds) are the leading global cause of mortality and disease burden. Statins are the most prescribed lipid-lowering drugs to treat hypercholesterolemia and prevent CVds. The biochemical mechanism of statins consists of competitive inhibition of the 3-hydroxy-3-methylglutaryl-CoA reductase enzyme (HMG-CoAR)， the limiting enzyme in cholesterol biosynthesis. due to statin intolerance in some patient groups， the search for new inhibitors is a field of great interest. This review focusses on the studies reporting the inhibitory effect of protein hydrolysates and biopeptides on the HMG-CoAR enzyme activity. The analysis of the action mechanism and physicochemical characteristics of the HMG-CoAR inhibitory peptides revealed that the molecular weight， amino acid composition， charge， and polarity are key aspects of the interaction with the HMG-CoAR enzyme. In conclusion， this review reveals the potential of using food peptides as new cholesterol-lowering agents and opens a new interesting field of research. However， clinical approaches are mandatory to confirm their therapeutic hypercholesterolemic effect.

Since the advent of sequencing technologies,the determination of microbial diversity to predict microbial functions,which are the major determinants of soil functions,has become a major topic of interest,as evidenced by the 900 publications dealing with soil metagenome published up to ***,the detection of a gene in soil does not mean that the relative function is expressed,and the presence of a particular taxon does not mean that the relative functions determined in pure culture also occur in the studied *** critical step is to link microbial community composition or function to the product analyzed to determine flux ***,flux rates might not only be highly dynamic,but several metabolites can depend on different reactions,which makes the link to one process of interest difficult or even *** review also discusses biases caused by sampling,storage of samples,dNA extraction and purification,sequencing(*** sequencing),and bioinformatic data *** and the limits of predicting microbial interactions by network inference methods are critically discussed,and finally,future directions for a better understanding of soil functions by using measurements of microbial diversity are presented.

Pure and Mg-doped self-assembled ZnO nano-particles for the enhanced photocatalytic degradation of 4-chlorophenol

A novel self-assembled pure and Mg doped ZnO nano-particles （NPs） were successfully synthesized by a simple low temperature co-precipitation method. The prepared photocatalysts were characterized by X-ray diffraction, high resolution scanning electron mi- croscopy, high resolution transmission electron microscopy, diffuse reflectance spectroscopy and photoluminescence （PL） spectroscopy. The results indicated that the prepared photocatalysts showed high crystallinity with a uniform size distribution of the NPs. The degradation of cholorphenols is highly mandatory in today＇s scenario as they are affecting the environment adversely. Thus, the photocatalytic degradation of 4-chlorophenol （4-CP）, a potent endocrine disrupting chemical in aqueous medium was investigated by both pure and Mg-doped ZnO NPs under UV-light irradiation in the present study. The influence of the Mg content on the structure, morphology, PL character and photocatalytic activity of ZnO NPs were investigated systematically. Furthermore,the effect of different parameters such as 4-CP concentration, photocatalyst amount, pH and UV-light wavelength on the resulting photocatalytic activity was investigated.
