Translational pancreatic cancer research:a comparative study on patient-derived xenograft models

AIM To assess the viability of orthotopic and heterotopic patient-derived pancreatic cancer xenografts implanted into nude *** This study presents a prospective experimental analytical follow-up of the development of tumours in mice upon implantation of human pancreatic adenocarcinoma samples. Specimens were obtained surgically from patients with a pathological diagnosis of pancreatic adenocarcinoma. Tumour samples from pancreatic cancer patients were transplanted into nude mice in three different locations(intraperitoneal, subcutaneous and pancreatic). Histological analysis(haematoxylin-eosin and Masson's trichrome staining) and immunohistochemical assessment of apoptosis(TUNEL), proliferation(Ki-67), angiogenesis(CD31) and fibrogenesis(α-SMA) were performed. When a tumour xenograft reached the target size, it was reimplanted in a new nude mouse. Three sequential tumour xenograft generations were generated(F1, F2 and F3).RESULTS The overall tumour engraftment rate was 61.1%. The subcutaneous model was most effective in terms of tissue growth(69.9%), followed by intraperitoneal(57.6%) and pancreatic(55%) models. Tumour development was faster in the subcutaneous model(17.7 ± 2.6 wk) compared with the pancreatic(23.1 ± 2.3 wk) and intraperitoneal(25.0 ± 2.7 wk) models(P = 0.064). There was a progressive increase in the tumour engraftment rate over successive generations for all three models(F1 28.1% vs F2 71.4% vs F3 80.9%, P < 0.001). There were no significant differences in tumour xenograft differentiation and cell proliferation between human samples and the three experimental models among the sequential generations of tumour xenografts. However, a progressive decrease in fibrosis, fibrogenesis, tumour vascularisation and apoptosis was observed in the three experimental models compared with the human samples. Extension of Calcifying Epithelial Odontogenic Tumor to the Maxillary Sinus: A Case Report

The calcifying epithelial odontogenic tumor (CEOT) is a rare and benign odontogenic epithelial neoplasm. This tumor accounts for less than 1% of all odontogenic tumors. It normally affects patients between 30 and 50 years old, and it is typically located in the posterior region of the mandible. Involvement of the maxillary sinus has previously been published only in six cases. This report presents a single case of CEOT that invades the maxillary sinus in a 69-year-old male. We performed a left partial maxillectomy and immediate reconstruction of the defect with a temporalis muscle flap. A comprehensive immunohistochemical study was reported. No recurrences have been found after 8 years of follow-up.

Pecan[Carya illinoinensis(Wangenh.)***.]is a deciduous tree whose fruits(nuts)are of high economic value and offer excellent nutritional ***,soils high in sulphates can limit its growth and *** with 5-year-old trees of‘Western Schley’pecan grown in soils high in sulphates,the levels of proline and oxidative metabolism were recorded in the *** showed that different levels of visible leaflet damage(‘sufficiency’,‘low’,‘moderate’or‘severe’)were associated with different levels of leaflet sulphates(mg kg^(−1)):‘sufficiency’(≤40),‘low’(41–60),‘moderate’(61–80)and‘severe’(80–100).‘Severe’sulphate damage was associated with significant reductions in chlorophyll(TChl)(17.04μg g^(−1)),relative water content(RWC)(50%)and leaf area(LA),and with increases in the concentrations of total carotenoids(TC)and proline(Prl).Increases were also observed in the activities of the oxidative metabolism enzymes:superoxide dismutase(SOD)(1.82 units min^(−1) g^(−1)),catalase(CAT)(2.86μmol H_(2)O_(2) min^(−1) g^(−1))and antioxidant capacity(AC)(87%DPPH inhibition).However,guaiacol peroxidase(GP)showed a reduction(2.97 nmol GSH min^(−1) g^(−1)).An inverse relationship was found between the sulphate concentration in the leaflets with respect to the evaluated parameters of TChl,TC,RWC,LA,AC,and *** synthesis and antioxidant enzymatic activity indicate salt stress in pecan leaflets in orchards irrigated with deep-well water high in sulphates.

Objective To determine whether topical applications of thiosulfinate-enriched Allium sativum extract(TASE)can accelerate acute cutaneous wound healing(WH)in a murine *** Keratinocyte viability and in vitro wound closure were assessed in keratinocyte *** of topical TASE(0.5μg/mL of allicin in 97%ethanol)on acute cutaneous WH were determined in a murine model of acute cutaneous *** mice were alternately assigned to the vehicle-and TASE-treated groups(n=6 per group).Expression levels of mRNA for keratinocyte differentiation marker-related proteins(filaggrin,loricrin and involucrin)and lipid synthetic enzymes(elongation of very long chain fatty acids protein 4(ELOVL4),fatty acid synthase(FA2H),3-hydroxy-3-methyl-glutaryl-coenzyme A reductase(HMGCoA),and serine palmitoyltransferase(SPT))were assessed using real-time quantitative polymerase chain reaction on day 3 and 8 after wounding,while transepidermal water loss(TEWL)rates were measured in wounded *** TASE accelerated WH both in vivo(40%vs.22%reduction in wound area,P<0.01)and in vitro(90%vs.65%reduction in wound area,P<0.01).Moreover,topical applications of TASE upregulated the expression levels of epidermal mRNA for ELOVL4,HMGCoA,SPT,filaggrin,loricrin and involucrin(P<0.05 ***-treated controls)on day 3 after ***,TASE significantly lowered TEWL rates in comparison with vehicle alone on day 8(33.06±2.09 g/(m^2·h)vs.24.60±2.04 g/(m^2·h),P<0.01).Conclusions Topical applications of TASE stimulated keratinocyte proliferation and formation of epidermal permeability barrier function,leading to acceleration of acute cutaneous *** products containing TASE could be used to manage acute cutaneous WH.

Transportation Safety Requirements in the Use of Pushchairs on Buses

The safe use of buses by passengers travelling with pushchairs for young children is a mobility problem that has not yet been satisfactorily resolved. The lack of national and international standards in this area in the past led to the development of a research project developed jointly by the Universitat Politecnica Valencia and the Universidad Politecnica Madrid （Spain）. This paper shows a comparative study to evaluate the dynamic behaviour in case of low-g accidents reproducing frontal impacts resembling real traffic events （deceleration 2g, AV 20 km/h） of a new pushchair restraint system against the use of back-restraint by pushchairs as establishes R 107- 06 series of amendments. In the dynamic trials, three pushchair restraint prototypes and a typical wheelchair back-restraint system combined with two worst-case up-to-date pushchair models in misuse and correct use configurations were tested. The results demonstrated the need for preventing children injuries as a consequence of low-g accidents. The new design meets and improves the latest revision of regulation R 107 regarding the use of restraint systems for the transport of wheelchair and pushchair passengers and represents an advance for pushchairs travelling on buses.

Hospital without dyspnea： rationale and design of a multidisciplinary intervention

Dyspnea is a common and disabling symptom of respiratory and heart diseases, which is growing in incidence. During hospital admis- sion, breathlessness is under-diagnosed and under-treated, although there are treatments available for controlling the symptom. We have developed a tailored implementation strategy directed to medical staff to promote the application of these pharmacological and non-pharmacological tools in dealing with dyspnea. The primary aim is to decrease the rate of patients that do not receive an adequate relief of dyspnea. This is a four-stage quasi-experimental study. The intervention consists in two teaching talks that will be taught in Cardiology and Respiratory Medicine Departments. The contents will be prepared by Palliative Care specialists, based on available tools for management of dyspnea and patients＇ needs. A cross-sectional study of dyspnea in hospitalized patients will be performed before and after the intervention to ascertain an improvement in dyspnea intensity due to changes in medical practices. The last phase consists in the creation of consensus protocols for dyspnea management based in our experience. The results of this study are expected to be of great value and may change clinical practice in the near future and promote a changing for the better of dyspnea care.

AIM To determine a potential relationship between serum undercarboxylated(uc OC) concentration and cardiovascular risk factors in type 2 diabetes(T2D) patients and healthy subjects(HS).METHODS A cross-sectional study was conducted on 140 subjects classified into two groups, 70 with T2D and 70 HS. Medical history and physical examination with anthropometric measurements were obtained from all subjects. Body fat percentage was determined by bioelectrical impendency analysis. Serum uc OC concentration was determined by enzyme immunoassay,while serum levels of insulin and hsC RP were obtained using high sensitivity enzyme-linked immunosorbent assay. Insulin resistance was determined using the homeostasis model assessment-IR. Lipid profile [triglycerides,total cholesterol(TC), high-density lipoproteins(HDL-c),low density lipoproteins(LDL-c), very low-density lipoproteins] was determined by spectrophotometry and standard formulas when applicable. RESULTS The T2D patient group showed significantly higher values of waist circumference, waist-to-hip ratio, systolic blood pressure(SBP), diastolic blood pressure(DBP),current smoking, and alcohol use when compared to the HS group(P < 0.05). We observed a significantly lower serum ucO C concentration in T2D than in HS(1.5 ± 1.4vs 2.3 ± 1.8, P < 0.05). In the whole study population,ucO C concentration was inversely correlated with body mass index(BMI)(r =-0.236, P < 0.05), fasting plasma glucose(r =-0.283, P < 0.01) and HDL-c(r =-0.255,P < 0.05); and positively correlated with LDL-c/HDL-c ratio(r = 0.306, P < 0.05) and TC/HDL-c ratio(r =0.284, P < 0.05). In the T2D group, serum uc OC concentration was inversely correlated with BMI(r =-0.310, P < 0.05) and body-fat percentage(r =-0.311,P < 0.05), and positively correlated with DBP(r = 0.450,P < 0.01). We have designed and evaluated a radial-flow bioreactor for three-dimensional liver carcinoma cell culture on a new porous coated *** designed a culture chamber where a radial flow of culture medi-um was continuously delivered through *** this system was established,flow was simulated using flow dy-namics software based on numeric methods to solve Navier-Stockes flow *** within cell culture scaffolds was simulated using a flow velocity of 7 mL/min and found that cell culture medium was distributed unhindered in the bioreactor ***,the bioreactor was built according to the simulated design and was tested with liver carcinoma cells(HepG2)cultured over an L-polylactic acid scaffold whose surface was modified with iodine-doped *** bioreactor was tested under non-flow and in radial flow *** density under radial flow conditions was almost double than that under static conditions and both total protein and albumin output was also increased under radial flow conditions.
