Diet switch and omega-3 hydroxy-fatty acids display differential hepatoprotective effects in an obesity/nonalcoholic fatty liver disease model in mice

AIM To study the effect of 18-hydroxy-eicosapentaenoic acid(18-HEPE) and 17-hydroxy-docosahexaenoic acid(17-HDHA) in a murine model of obesity/nonalcoholic fatty liver *** C57 BL/6 mice were fed with standard chow diet(CD) or high-fat, fructose-enriched diet(HFD) for 16 wk. Then, three groups were treated for 14 d with either, diet switch(HFD for CD), 18-HEPE, or 17-HDHA. Weightand fasting glucose were recorded on a weekly basis. Insulin tolerance test was performed at the end of treatment. Histological analysis(HE and Masson's trichrome stain) and determination of serum insulin, glucagon, glucagon-like peptide 1(GLP-1), glucose-dependent insulinotropic polypeptide, adiponectin and resistin were carried out as well as liver proteins by western *** Mice treated with hydroxy-fatty acids 18-HEPE and 17-HDHA displayed no weight loss or improved insulin sensitivity. However, these mice groups showed a significant amelioration on serum GLP-1, adiponectin and resistin levels. Also, a significant reduction on inflammatory infiltrate was observed at both portal and lobular zones. Furthermore, up-regulation of PPARα/γ protein levels was observed in liver tissue and it was associated with decreased levels of NF-κB also determined by western blot analysis. On the other hand, diet switch regimen resulted in a marked improvement in most parameters including: weight loss, increased insulin sensitivity, decreased steatosis, restored levels of insulin, glucagon, leptin, adiponectin and resistin. However, no significant changes were observed regarding inflammatory infiltrate in this last *** 18-HEPE and 17-HDHA differentially exert hepatoprotective effects through up-regulation of nuclear receptors PPARα/γ and amelioration of serum adipokines profile.

The growing demand for air travel has led to the saturation of air traffic *** methods of adding routes to alleviate congestion and reduce delays may not achieve the desired effect and even degrade system *** this paper,we explore the application of Braess’s Paradox in the reduction of air traffic *** counterintuitive phenomenon shows that adding new connections to a network can actually increase the overall network *** study uses Hidden Markov methods and the Viterbi algorithm to match air traffic flow with routes,a machine learning approach and a mathematical method to construct cost functions for flight time and traffic volume,and finally uses genetic algorithm and the A*algorithm to detect Braess’s Paradox *** uses ADS-B data from the busy month of July 2019 for a case study of the air traffic network over the UK *** results show that Braess’s Paradox is also applicable to multi-flight level air route *** such network edges can improve system *** one day’s case,the total flight time of the day’s traffic volume decreased from 11509.24 minutes to 10459.97 *** equates to an average savings of 4.99 minutes of flight time per flight,which is significant in controlling delay performance.

Excretion of biliary compounds during intrauterine life

In adults,the hepatobiliary system,together with the kidney,constitute the main routes for the elimination of several endogenous and xenobiotic compounds into bile and urine,***,during intrauterine life the biliary route of excretion for cholephilic compounds,such as bile acids and biliary pigments,is very *** very early in pregnancy the fetal liver produces bile acids,bilirubin and biliverdin,these compounds cannot be efficiently eliminated by the fetal hepatobiliary system,owing to the immaturity of the excretory machinery in the fetal ***,the potentially harmful accumulation of cholephilic compounds in the fetus is prevented by their elimination across the *** to the presence of detoxifying enzymes and specifi c transport systems at different locations of the placental barrier,such as the endothe-lial cells of chorionic vessels and trophoblast cells,this organ plays an important role in the hepatobiliary-like function during intrauterine *** relevance of this excretory function in normal fetal physiology is evident in situations where high concentrations of biliary compounds are accumulated in the *** may result in oxidative stress and apoptosis,mainly in the placenta and fetal liver,which might affect normal fetal development and challenge the fate of the *** present article reviews current knowledge of the mechanisms underlying the hepatobiliary function of the fetal-placental unit and the repercussions of several pathological conditions on this tandem.

Multitemporal landslide inventory analysis of an intertropical mountain in west-central Mexico——Basis for hazard management

Landslides in intertropical mountainous areas of Mexico are a natural hazard that could potentially generate a disaster. Despite this, many areas lack landslide inventories, making it difficult to monitor and efficiently manage the associated risks for local populations. For this reason, the aim of this research was to undertake a multitemporal landslide inventory and analyze its conditioning factors in the Pico de Tancítaro stratovolcano, which, in recent decades, has presented several high-risk geomorphological events with disastrous consequences for nearby localities. The spatial distribution of landslides and its conditioning factors were mapped at a 1:50,000 scale for the 1995-2020 period, through visual interpretation of orthophotos and satellite images(Landsat, SPOT, and SENTINEL). The conditioning factors that link the most to landslides were Quaternary andesite and basaltic andesite rocks;volcanic cones, mountain river valleys, and very steep to steep slopes(30° to >45°), oriented to the South, North, and East;Andosols and Leptosols;and the oyamel fir and mixed forests. 505 landslide events classified into slides and flows that occurred in 1995, 2004, 2010, and 2015 were identified. New scars generated by the reactivation of landslide processes were also observed. The landslides in the study area were triggered by torrential rains caused by hurricanes and tropical storms;such as hurricanes Alex and Patricia. The inventory and analysis of conditioning factors highlighted the distribution and dynamics of slides and flows in the study area. In addition, the most important factors for studies of landslide susceptibility, hazards, and risks in the Pico de Tancítaro stratovolcano were identified, contributing to the management of geomorphological risks in intertropical mountain areas.

Bacille-Calmette-Guerin modulates human macrophage and dendritic cell response to SARS-CoV-2 S-glycoprotein

Background:Given that epidemiological evidence suggests a potential protective role for Bacille-Calmette-Guerin against COVID-19,we aimed to explore whether pre-exposure of human monocyte-derived macrophages and dendritic cells to BCG could modulate their response to SARS-CoV-2 ***:Dual THP-1 cells containing 2 reporter plasmids for transcription factors NF-κB,and IRF were differ-entiated into macrophages over 3 days using phorbol 12-myristate 13-acetate,or into dendritic cells over 6 days using commercial monocyte-dencritic cell differentiation media and matured with recombinant tumor necrosis factor-α.Cells were exposed to BCG for 24 h and then stimulated with SARS-CoV-2 S-glycoprotein for 24 ***:Pre-exposure of human macrophages and DCs to BCG increased IRF and NF-kb activation in response to the SARS-CoV-2 ***:Our results showed that pre-exposure of both types of cells to BCG exhibited an increase in inflam-matory transcription factors upon stimulation with ***-induced trained immunity may be an important tool for reducing susceptibility to SARS-CoV-2 infection and severity of *** findings help in the design of future BCG-based therapeutic approaches in the treatment of diseases caused by viral infections.

Clinical relevance of the relationship between changes in gut microbiota and bile acid metabolism in patients with intrahepatic cholangiocarcinoma

Although only 10-15%of liver cancers are cholangiocarcinomas(CCAs),which derive from the epithelial cells of the biliary tree,these cancers constitute a serious and growing health problem *** rate of mortality due to this tumor is very high because CCA usually appears in aged people and very often,in more than 70%of cases,are diagnosed late because of:(I)their anatomical localization in a big organ like the liver;(II)the lack of specific symptoms during most time of their development;and(III)the absence of available accurate,specific and sensitive biomarkers(1).These characteristics reduce the rate of success of surgical resection,which is the only curative option for CCA patients,because the response of this aggressive cancer to the available pharmacological armamentarium is extremely feeble(2).This justifies the interest of many groups to identify non-invasive markers to achieve early diagnosis that also permit to distinguish CCA from non-malignant biliary diseases and from hepatocellular carcinoma(HCC),the most frequent liver cancer,which derives from hepatocytes(3).

Office Furniture Design for Wheelchair User

There are an estimated 1.19 million wheelchair users in Spain. People use wheelchairs for a variety of reasons, the most common one is paralysis from spinal cord injuries. Current estimates indicate there are around 504,000 people alive in Spain with spinal cord injuries. Among other reasons for using wheelchairs are： fatigue from multiple sclerosis, muscle weakness from muscular dystrophy, lower limb spasticity from cerebral palsy, and missing limbs due to amputation. People who use wheelchairs may encounter a variety of obstacles at their workstations depending on their limitations. Some possible accommodations could be adjustable height desk or table for a person who cannot work comfortably at an existing desk, accessible filing system or office supplies, and frequently used materials on most accessible shelves or drawers for a person who cannot reach upper and lower shelves and drawers. This paper presents the graphical design of a workstation for wheelchair users, in this case, technical, anthropometric, ergonomic, aesthetics and scientists aspects have been analyzed. The design of composite structure conduct to explicit safety constraints applied to office furniture requirements kept continuously current and updated as part of innovation. Finally, results are showed in a three-dimensional model.

BACKGROUND Matrix metalloproteinases(MMPs)participate in the degradation of extracellular matrix compounds,maintaining the homeostasis between fibrogenesis and fibrolytic processes in the ***,there are few studies on the regulation of liver MMPs in fibrosis progression in *** To assess the production activity and regulation of matrix metalloproteinases in liver fibrosis stages in chronic hepatitis C(CHC).METHODS A prospective,cross-sectional,multicenter study was *** patients were categorized in fibrosis grades through FibroTest®and/or FibroScan®.Serum MMP-2,-7,and-9 were determined by western blot and multiplex suspension array *** were validated by the Kruskal-Wallis and Mann-Whitney U *** Spearman correlation coefficient and area under the receiver operating characteristic curve were *** and gelatinase activity was determined through the Azocoll substrate and zymogram test,whereas tissue inhibitor of metalloproteinase-1 production was determined by dot blot *** Serum concentrations of the MMPs evaluated were higher in CHC patients than in healthy ***-7 distinguished early and advanced stages,with a correlation of 0.32(P<0.001),and the area under the receiver operating characteristic displayed moderate sensitivity and specificity for MMP-7 in F4(area under the receiver operating characteristic,0.705;95%confidence interval:0.605-0.805;P<0.001).Collagenolytic activity was detected at F0 and F1,whereas gelatinase activity was not detected at any fibrosis *** inhibitor of metalloproteinase-1 determination showed upregulation in F0 and F1 but downregulation in F2(P<0.001).CONCLUSION High concentrations of inactive MMPs were present in the serum of CHC patients,reflecting the impossibility to restrain liver fibrosis *** could be good diagnostic candidates and therapeutic targets for improving novel strategies to reverse liver fibrosis in CHC.
