AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-Cas9 system and analyze phenotypic modifications as well as gene expression *** CRISPR-Cas9 system used was purchased from dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(dOCK2, ZNF503, PdE4 d, ABL1, ABL2, LPAR1, SMAd2, WASF3 and dACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCLUSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

Fruit Set in Orange with Phytohormones and Its Relation to Endogenous Levels of ga3

This research work aims to contribute in increasing the fruit set in citrus, given its importance in determining fruit yield. The authors evaluated the effects of phytohormones （auxins, gibberellins and cytokinins） in the mooring and features of orange fruit in cultivars Washington navel and Thomson （Citrus sinensis （L.） Osb.）, The experiment was established in a split plot randomized complete block design with five treatments and four replications. Flower tissue samples were stored in liquid N until the extraction of gibberellins and the identification and quantification of ga3. The results showed statistically significant difference （P 〈 0.05） between treatments in the number of fruits retained 129 d after flowering and the percentage of final tie. ga3 content ranged, on a dry weight basis, from 1.66 mg＇g1 in the control to 20.79 mg.g-1 in the high dose. The mean dose （32.2 mg.L-1 auxins, gibberellins 32.2 mg L-1 and 83.2 mg.L-1 cytokinins） caused the largest increase in fruit set.

Antimicrobial Potential and Phytochemical Content of Six diverse Sources of Quinoa Seeds (<i>Chenopodium quinoa</i>Willd.)

Antimicrobial potential of quinoa seeds from six diverse seeds, grown in three distinctive geo-graphical zones of Chile, was correlated with their total phenolic content (TPC), total flavonoid content (TFC) and total saponin content (TSC). The six seeds were Ancovinto, Cancosa, Cahuil, Faro, Regalona and Villarrica. A significant influence of quinoa source on chemical composition of seeds was observed. Extracts of all quinoa samples showed antimicrobial activity in the range of 8.3 - 14.8 mm inhibition zone for E. coli and 8.5 - 15.0 mm inhibition zone for S. aureus. Cancosa seeds had the highest antimicrobial action. The TPC and TFC ranged from 3.71 to 16.55 mg ga/100 g d.m. and 7.77 to 14.37 mg QE/100 g d.m., respectively. TSC varied from 1.78 to 3.08 g/100 g d.m. A cor-relation between TFC and antimicrobial activity was found. In conclusion, the six types of quinoa seeds were identified as potential sources of antioxidant compounds and antimicrobial activity.

Importance of the Role of Gibberellic Acid and Potassium Nitrate in Seed Germination Habanero

The habanero chili （Capsicum chinense Jacq.） is a species of great demand in the domestic and international markets. One problem in the cultivation is low percentage of seed germination. There is little information on the use of promoters and imbibition time to increase this parameter, without compromising the quality of seedlings. In this context, authors assessed the independent effect of gas （gibberellic acid） and KNOs at concentrations of 0, 100, 200, 300, 400 and 500 mg·L1. Seeds without preconditioning were the witness. The experimental unit was a Petri dish with 50 seeds on filter paper placed equidistant. There were three replicates per treatment and the experimental design was randomized complete block. Imbibition time was 24 h. The experimental units were kept in an incubator at 22 ± 2 ℃. From the time of planting and for periods of 24 h for 14 d, experimental units were reviewed to determine the time at the beginning of germination and the total cumulative percentage. The authors also determined the number of normal and abnormal seedlings, the length of hypocotyl and radicle, weight of fresh and dry matter of seedlings. On the fifth day after sowing in vitro, concentrations of 500 mg·L^-1 of gas, 300 and 400 mg·L^-1 KNO3, increased 208% the percentage of seed germination of habanero chile. On day 14 after sowing in vitro concentrations of 500 mg·L^-1 ga3, 300 and 400 mg·L^-1 KNO3, increased 23% normal seedlings and 40% dry matter thereof, with respect to seeds obtained without preconditioning. Unconditioned seeds before sowing produced 125% more seedlings.

Pediatric autoimmune neuropsychiatric disorders associated with or without streptococcal and other bacterial infections (PANdAS/CANS) are emerging as a featured pediatric disorder. Although there is some controversy regarding treatment approaches, especially related to the behavioral sequelae, we have hypothesized in other published work that it is characterized by the rapid onset of Reward deficiency Syndrome (RdS) in children. We propose utilizing a multi-systems biological approach involving the coupling of genetic addiction risk testing and pro-dopamine regulation (KB220/POLYGEN®) to help induce “dopamine homeostasis” in patients with PANdAS, especially those with known dNA-induced hypodopaminergia. This case study examines a 12-year-old Caucasian male with no prior psychiatric issues who presented with a sudden onset of severe anxiety, depression, emotional liability, and suicidal ideation. The patient underwent genotyping and the genetic addiction risk score (gaRS) testing, which revealed risk polymorphisms in the dopamine d2 (-dRd2/ANKK (Taq1A), OPRM1 (A/G), dRd3 (C/T), and MAOA (4R) genes. These polymorphisms have been linked to hypodopaminergia. The patient was subsequently placed on research Id-KB220ZPBMPOLY (POLYGEN®), and albeit the possibility of bias, based upon self and parental assessment, a marked rapid improvement in psychiatric symptoms was observed. In the second phase of treatment (102 days utilizing KB220), the patient received standard antibody testing, which was positive for Lyme. Antibacterial therapy started immediately, and KB220z was discontinued to provide a wash-out period. A monotonic trend analysis was performed on each outcome measure, and a consistently decreasing trend was observed utilizing antibacterial therapy. Our recommendation, albeit only one case, is to utilize and further research a combined therapeutic approach, involving precision-guided dNA testing and pro-dopamine regulation along with antibacterial therapy, as well as

Byrsonima intermedia A Juss. is a species with pharmacological properties from the Brazilian Cerrado that shows difficulties related to sexual propagation. Research on cell viability may provide useful information for the selection of cells with embryogenic potential during the callus culture, Within this context, our research is aimed at establishing the cell viability of calli from Byrsonima intermedia leaf segments. The calli went through three subculture phases, of 60 days each, in MS medium with 0.09 M sucrose, 0.6% agar, pH 5.8 and 4.52 laM 2,4-d. The calli were stored in dark conditions and samples were collected every 10 days from each subculture for viability tests with fluorescein 3,6-diacetate （FdA） and 2,3,5-triphenyltetrazolium chloride （TTC）. The staining methods allowed quantifying cell viability in each subculture. The best results from the FdA tests were obtained at 21, 25 and 29 days for the first, second and third subcultures respectively, with 53,86%, 61.88% and 53.73% viable cells. Regarding the TTC test, the largest absorbance values were obtained at 21, 27 and 28 days for the first, second and third subcultures respectively. Fluorescence and spectrophotometry analyses were efficient for determination of cell viability during callus cultivation period.

Presently, several different graft materials are employed in regenerative or corrective bone surgery. However current misconceptions about these biomaterials, their use and risks may compromise their correct application and development. To unveil these misconceptions, this work briefly reviewed concepts about bone remodeling, grafts classification and manufacturing processes, with a special focus on calcium phosphate materials as an example of a current employed biomaterial. Thus a search on the last decade was performed in Medline, LILACS, Scielo and other scientific electronic libraries using as keywords biomaterials, bone remodeling, regeneration, biocompatible materials, hydroxyapatite and therapeutic risks. Our search showed not only an accelerated biotechnological development that brought significant advances to biomaterials use on bone remodeling treatments but also several therapeutic risks that should not be ignored. The biomaterials specificity and limitations to clinical application point to the current need for developing safer products with better interactions with the biological microenvironments.
