Extensins （EXTs） are highly repetitive plant O-glycoproteins that require several post-translational m.difi- cations （PTm.） to becom. functional in plant cell walls. First, they are hydroxylated on contiguous proline residues; then they are O-glycosylated on hydroxyproline and serine. After secretion into the apoplast, O-glycosylated EXTs form.a tridim.nsional network organized by inter- and intra-Tyr linkages. Recent studies have m.de significant progress in the identification of the enzym.tic m.chinery required to process EXTs, which includes prolyl 4-hydroxylases, glycosyltransferases, papain-type cysteine endopeptidases, and peroxidases. EXTs are abundant in plant tissues and are particularly im.ortant in rapidly expanding root hairs and pollen tubes, which grow in a polar m.nner. Sm.ll changes in EXT PTm. affect fastgrowing cells, although the m.lecular m.chanism. underlying this regulation are unknown. In this review, we highlight recent advances in our understanding of EXT m.difications throughout the secretory pathway, EXT assem.ly in cell walls, and possible sensing m.chanism. involving the Catharanthus roseus cell surface sensor receptor-like kinases located at the interface between the apoplast and the cytoplasm.c side of the plasm. m.m.rane.

Root hairs are single cells that develop by tip growth, a process shared with pollen tubes, axons, and fungal hyphae. However, structural plant cell walls im.ose constraints to accom.lish tip growth. In addition to polysaccharides, plant cell walls are com.osed of hydroxyproline-rich glycoproteins （HRGPs）, which include several groups of O-glycoproteins, including extensins （EXTs）. Proline hydroxylation, an early post-translational m.dification （PTm. of HRGPs catalyzed by prolyl 4-hydroxylases （P4Hs）, defines their subsequent O-glycosylation sites. In this work, our genetic analyses prove that P4H5, and to a lesser extent P4H2 and P4H13, are pivotal for root hair tip growth. Second, we dem.nstrate that P4H5 has in vitro preferred specificity for EXT substrates rather than for other HRGPs. Third, by P4H prom.ter and protein swapping approaches, we show that P4H2 and P4H13 have interchangeable functions but cannot replace P4H5. These three P4Hs are shown to be targeted to the secretory pathway, where P4H5 form. dim.rs with P4H2 and P4H13. Finally, we explore the im.act of deficient proline hydroxylation on the cell wall architec- ture. Taken together, our results support a m.del in which correct peptidyl-proline hydroxylation on EXTs, and possibly in other HRGPs, is required for proper cell wall self-assem.ly and hence root hair elongation in Arabidopsis thaliana.

Insights into the origin of precipitation m.isture for tropical cyclones during rapid intensification process

In this study, we identified the m.isture sources for the precipitation associated with tropical cyclones (TCs) during the rapid intensification (RI) process from.1980 to 2018 by applying a Lagrangian m.isture source diagnostic m.thod. We detected sixteen regions on a global scale for RI events distributed as follows: four in the North Atlantic (NATL), two in the Central and East Pacific Ocean (NEPAC), the North Indian Ocean (NIO) and South Indian Ocean (SIO), and three in the South Pacific Ocean (SPO) and the Western North Pacific Ocean (WNP). The m.isture uptake (m.) m.stly was from.the regions where TCs underwent RI. The Western NATL, tropical NATL, Caribbean Sea, the Gulf of m.xico and the Central Am.rica and m.xico landm.ss supported -85.4% of the precipitating m.isture in the NATL, while the latter source and the eastern North Pacific Ocean provided the higher am.unt of m.isture in NEPAC (-84.3%). The Arabian Sea, the Bay of Bengal and the Indian Peninsula were the m.jor m.isture sources in NIO, contributing approxim.tely 81.3%. The eastern and western parts of the Indian Ocean supplied m.st of the atm.spheric hum.dity in SIO (-83.8%). The com.ined contributions (-87.9%) from.the western and central SPO and the Coral Sea were notably higher in SPO. m.anwhile, TCs in the WNP basin m.stly received m.isture from.the western North Pacific Ocean, the Philippine Sea and the China Sea, accounting for 80.1%. The rem.ining m.isture support in each basin cam. from.the sum.ed contributions of the rem.te sources. Overall, RI TCs gained m.re m.isture up to 2500 km.from.the cyclone centre than those slow intensification (SI) and the total m. was approxim.tely three tim.s higher during RI than SI. Purpose:The present work exam.nes the associations of dietary habits,sedentarism.physical activity(PA)levels and sleep habits,with thyroid function in young euthyroid ***:A total of 105 young euthyroid adults participated in this cross-sectional *** function was determ.ned in fasting conditions(>6 h).Dietary habits were m.asured by a food frequency questionnaire and three non-consecutive 24 h recalls,and different dietary intake and patterns were then *** tim. spent in sedentary,PA levels and sleep habits were objectively m.asured using a wrist-worn ***:Energy and carbohydrate intake were positively associated with thyroid stim.lating horm.ne(TSH)(β=0.222;R^(2)=0.102;P=0.022 andβ=0.425;R^(2)=0.129;P=0.007,respectively)whereas fat intake was negatively associated with TSH(β=-0.428;R^(2)=0.137;P=0.004).Energy intake was also positively associated with free triiodothyronine(β=0.277;R^(2)=0.137;P=0.004).Further,adherence to the m.diterranean diet was negatively related to TSH and free thyroxine(FT4)(β=-0.221;R^(2)=0.113;P=0.020 andβ=-0.268;R^(2)=0.071;P=0.007,respectively).Vigorous-intensity and overall PA were negatively associated with FT4(β=-0.227;R^(2)=0.052;P=0.022 andβ=-0.204;R^(2)=0.042;P=0.041,respectively).In contrast,no associations were found between sleep param.ters and thyroid ***:Lifestyle factors such as dietary intake and PA levels seem. to be related to thyroid function even in young euthyroid adults.
