How Escherichia coli bacteria develop a particular colonial, 3-D biofilm morphological pattern is still a poorly understood process. Recently, we reported a new E. coli K-12 morphotype exhibited by old macrocolonies described as volcano-like. The formative developmental process of this morphotype has been presented as a suitable experimental model for the study of 3D patterning in macrocolony biofilms. Here, we report the optical microscopy observations and genetic analysis that have unveiled the existence of a novel autoaggregative behaviour which generates massive lumpiness over the surface of the volcano-like macrocolonies. These lumpy formations are generated by the autoaggregation and strong interaction of tightly packed bacterial cells in structures with a chondrule-like appearance which give the colony’s surface its characteristic microscopic lumpy phenotype. Furthermore, they exhibit different levels of maturation from the edge to the center of the colony. Hence, its generation appears to follow a spatiotemporal program of development during the macrocolony’s morphogenesis. Interestingly, the agar’s hardness influences the morphology exhibited by these formations, with high agar concentration (1.5%, 15 g/l) suppressing its development. This new auto-aggregative E. coli’s behaviour does not require the activity of the biofilm master regulator CsgD, the adhesiveness of flagella, pili type 1, adhesin Ag43, β-1,6-N-acetyl-D-glucosamine polymer-PGA, cellulose or colanic acid, but it is under glucose repression and the control of cAMP receptor protein (CRP). The possible physiological role of these chondrule-like formations in the adaptability of the colony to different stressful environmental conditions is discussed.

＜正＞The sprocket pump, patented in 1988, is used in applications such as fuel pumping in boilers, transfer of fluids, water pumping in treatment plants, and oil pumping in lubrication systems. Design modifications to improve its performance are described. These include: （1） changing the cross section of the pins that produce the pumping action, from circular to a combination of circular arcs and straight segments, in order to achieve surface rather than line sealing;（2） introduction of an extension to the sprocket wheel resulting in a hydrostatically balanced condition;and （3） providing pressure zones on the back face of the drive disk to partially balance the force due to the pressure distribution on the front face.

Objective: to evaluate the protective effect of kombucha and its components not polar, in pancreas of diabetic rats treated with streptozotocin. Material and methods: ninety-six male Wistar rats of 120 to 170 g of PC were used;four groups of 24 animals were formed: control (GC), control treatment (GCTx), kombucha treatment (GTxK) and treatment with non-polar components of kombucha (GTxCNP). Diabetes was induced in the last three groups with an intraperitoneal injection of streptozotocin at a dose of 65 mg/kg BW. At the same time, treatment was started in the different groups;GC and GCTx groups were administered, orally, 1 ml of sterile saline solution;to the GTxK group 324 mg/kg of kombucha;and to GTxCNP 0.006 mg/kg of the non-polar components. For the histopathological study, pancreas samples were obtained on days 1, 7, 14 and 21 and fixed in formalin. They were processed with the histological technique, 5-micron-thick sections were made and stained with the hematoxylin-eosin technique. Finally, the number of islets of langerhans per field observed with the 10x objective was quantified. The results were subjected to a one-way analysis of variance and Tukey’s test (p Results: Significant changes in the number of islets were observed in histological sections of the pancreas. The groups treated with streptozotocin showed a significant decrease in the number of islets 7 days after starting treatment;likewise, their number was lower in the GCTx group compared to the GTxK and GTxCNP groups (p ≤ 0.05). At 14 and 21 days, the lowest number of islets was maintained, although only in the GCTx and GTxK groups (p ≤ 0.05). Streptozotocin is a drug that can be used in rats to induce an experimental model of diabetes with an effect that can last up to 21 days. Conclusions: This fermented beverage and its non-polar components can be a complementary treatment alternative for diabetes mellitus, since it prevents further damage to the β cells of the pancreas and the Islets of langerh

The extraction of bioactive compounds has become one of the most interesting areas of modern *** therapeutic reasons,it´s important to obtain antimicrobial agents from natural *** objective of the present study was to determine the inhibitory effect of ethanolic extract of basil(Ocimum basilicum *** Rubin)subjected to different concentrations of potassium(K^(+))on the activity of three bacterial strains that are pathogens in *** was evaluated by inhibition surface and these results were compared to two antibiotics:Gentamicin(GE)and Ciprofloxacin(CPF)for their efficacy against each bacterial *** variables presented significant difference(p≤0.05).According to the results,basil extract evaluated showed positive antibacterial activity against the three strains of Pseudomonas aeruginosa,listeria monocytogenes and Staphylococcus aureus on Mueller Hinton *** was observed highest inhibition areas of different diameters(15.3 mm,21.3 mm and 21.6 mm respectively)when the extract used was obtained from the plants with the highest concentration of potassium(13 mmol l-1).These values were even superior to the highest values showed on the treatments with the antibiotic gentamicin.

The"Crime of Flores Street"is one of the most famous cases of poisoning to divide public opinion in Portugal in the late 19th century, and it also demonstrated the weaknesses of the Portuguese medicolegal system and attested to the importance of toxicological analysis. Vicente Urbino de Freitas was a prominent doctor, graduating from the Faculty of Medicine of the University of Coimbra in 1875. He later became Professor of Physiology at the Porto Medical-Surgical School and author of a number of books on leprosy. In 1877, he married Maria das Dores Basto Sampaio Freitas, and this was followed by the death of a number of her close relatives in suspicious circumstances, notably her brother josé António Sampaio Junior and nephew Mário Guilherme Augusto de Sampaio. This review aims to retell the story of Portugal's first significant medicolegal case as well as the accompanying judicial drama that gave birth to Forensic Toxicology in Portugal and prompted the medicolegal organization that exists today. This research was carried out over a 10-year period and repre-sents undeniable historical value given the rarity of the facts compiled. At the heart of this forensic case was the use of toxicological analyses in court for which the Chemist Antònio Joaquim Ferreira da Silva played a key role. This toxicological report revealed high concen-trations of morphine, delphinine and narceine in viscera and in Mario's urine. The Mario's cause of death was attributed to poisoning by opium alkaloids. Despite the strong judicial evidence, doubts still remains as to whether Vicente Urbino de Freitas was a"monster"or a victim of circumstances and a hapless martyr.

Method Validation for SPE Applied to Determination of PAH in Petroliferous Industry Effluent Water

The presence of polycyclic aromatic hydrocarbons (PAH) in produced water is of environmental concern due to their toxic properties. PAH analysis in complex samples requires pre-treatment to enrich the fraction con- taining analytes, and eliminate matrix interferences. The objective of this work was to develop and validate an analytical methodology for determination of PAH in produced water, using solid phase extraction (SPE) and analysis by gas chromatography with flame ionization detection (GC-FID). Average recoveries of PAH from produced water enriched at two concentration levels varied from 30.9% for naphthalene to 119.1% for chrysene (RSD between 3.8% and 22.2%). The linear range was between 0.5 and 50.0 μg?ml–1, with regres- sion coefficients better than 0.998. Detection limits were between 0.01 and 0.04 μg?l–1, and quantitation lim- its were between 0.05 and 0.16 μg?l–1. The validated method was applied to samples of produced water treated for disposal, in which concentrations varied from 3.5 μg?l–1 for phenanthrene to 44.3 μg?l–1 for naphthalene ( = 177.7 μg?l–1). The method was also applied to seawater samples, in which 13 PAH compounds were detected ( = 60.27 μg?l–1), probably derived from pyrogenic sources.
