Tailoring the partitioning of proteins using ionic liquids as adjuvants in polymer-polymer aqueous biphasic systems

Aqueous biphasic systems(ABS)are promising for proteins purification;however,when dealing with samples comprising several proteins,the selectivity towards a target protein is difficult to *** this work,the addition of ionic liquids(ILs)as adjuvants(5 wt%and 10 wt%)in ABS composed of polyethylene glycol(pEG)and dextran was investigated to tailor proteins partitioning between the coexisting *** liquid-liquid phase diagrams were determined at 298 K,followed by partition studies of three proteins(bovine serum albumin(BSA),immunoglobulin G(IgG),and cytochrome C(Cyt C)).partition coefficients of IgG and BSA indicate the preference of both proteins to the dextran-rich phase,whereas Cyt C has no preferential partitioning between the *** addition of chloride-based ILs as adjuvants allows to increase or decrease the partition coefficients,thus tailoring the proteins partitioning between the *** partitioning essentially depends on the IL content in each phase,whereas Cyt C and IgG partitioning is ruled by the ILs chemical structure and established *** docking was carried out to address the ILs effect on the proteins partitioning,supporting experimental observations,while identifying the specific interactions *** partition of each protein in polymer-salt ABS with ILs as adjuvants was determined,demonstrating the higher tailoring ability of polymer-polymer ABS when adding ***,the partition of each protein in presence of the remaining ones was determined,as well as the selectivity of the studied systems to separate each pair of proteins,paving the way for their use in liquidliquid chromatography.

Nutrient content affecting Spodoptera frugiperda and Dalbulus maidis occurrence in corn

This work aims to study the effect of the nutrient contents in the leaves of corn on the incidence of Spodopterafrugiperda （Smith）（Lepidoptera： Noctuidae） and Dalbulus maidis （Delong ＆ Wolcott）（Homoptera： Cicadellidae）. The treatments were represented by unfertilized corn plants and corn plants fertilized with 250 kg of the formulation 4-14-8 （N- p-K）＋ 100 kg of ammonium sulfate （AS）/ha and 500 kg of the formulation 4-14-8 （N-p-K）＋200 kg of AS/ha, arranged in randomized blocks with four replicates. Attacks by S. frugiperda and D. maidis were higher as the nitrogen, sulfur, calcium and copper content in the leaves of corn increased. The effects of calcium and copper found in this study were not consistent with that of lower susceptibility associated with higher levels of these nutrients mentioned in the literature. We believe that there was a cumulative effect of the nutrients altogether, building up a nutritiously better vegetable which in turn had less reserve for defense, or that the content of both nutrients in the leaves were not high enough to induce such effects.

Hyperacusis and stapes surgery:An observation in fifty patients after stapedotomy

Objective:To assess hyperacusis after stapedotomy and its possible influencing *** design:prospective,interventional,and longitudinal ***:A tertiary referral ***:Fifty consecutive patients(35 females,mean age=46.8 years).Intervention:All patients underwent *** validated portuguese version of the“Hyperacusis Questionnaire”(HQ)was administered before and two weeks and one month after ***:No hyperacusis was reported by any patient before *** two weeks after surgery,all patients experienced hyperacusis,with a mean HQ at 16.88±6.54(range 4e25).One month after surgery,hyperacusis had already resolved in most ***,preoperative presentation or surgeon had no influence on HQ scores(p>0.05).patients with previous contralateral stapedotomy showed lower HQ scores(p=0.001).Audiological parameters improvement measured at one month after surgery(pTA,SRT and contralateral SRT)were associated with HQ higher ***:This study confirms that hyperacusis is a common complaint after stapedotomy that usually resolves in one month after *** HQ highest scores were registered among patients with the highest audiological gain after *** suggests that hyperacusis may be a positive prognostic factor for audiological success after stapedotomy.

Analysing vaccine efficacy evaluated in phase 3 clinical trials carried out during outbreaks

In this paper we examine several definitions of vaccine efficacy(VE)that we found in the literature,for diseases that express themselves in outbreaks,that is,when the force of infection grows in time,reaches a maximum and then *** fact that the disease occurs in outbreaks results in several problems that we *** propose a mathematical model that allows the calculation of VE for several *** trials usually needs a large number of volunteers that must be ***,all volunteers should be enrolled in approximately the same time,but this is generally impossible for logistic reasons and they are enrolled in a fashion that can be replaced by a continuous density function(for example,a Gaussian function).The outbreak can also be replaced by a continuous density function,and the use of these density functions simplifies the ***,for example Gaussian functions,one of the problems one can immediately notice is that the peak of the two curves do not occur at the same *** model allows us to conclude:First,the calculated vaccine efficacy decreases when the force of infection increases;Second,the calculated vaccine efficacy decreases when the gap between the peak in the force of infection and the peak in the enrollment rate increases;Third,different trial protocols can be simulated with this model;different vaccine efficacy definitions can be calculated and in our simulations,all result are approximately the *** final,and perhaps most important conclusion of our model,is that vaccine efficacy calculated during outbreaks must be carefully examined and the best way we can suggest to overcome this problem is to stratify the enrolled volunteer's in a cohort-by-cohort basis and do the survival analysis for each cohort,or apply the Cox proportional hazards model for each cohort.

primary Liver Large B-Cell Lymphoma: A Rare Diagnosis

primary liver lymphoma is extremely rare, in most of cases it is a B cell lymphoma. Usually the diagnosis is made in middle-aged individuals and most of them have a relatively short life expectancy. In this article, the authors present a case report of a 75-year-old woman with symptoms of three weeks of evolution of diffuse abdominal pain, asthenia and anorexia. The analysis showed cyto cholestasis and the radiological image with lush hepatomegaly because of a large hepatic mass. The patient started chemotherapy and actually is well twenty months after the end of the chemotherapy.

Introduction:The histological diagnosis of prostate cancer is commonly based on morphological *** presence of malignant tissue mixed with benign tissue,or the presence of carcinoma that mimics benignity may generate difficulty in the diagnostic ***,the application of immunohistochemistry contributes its diagnostic ***:To evaluate the 34βE12 marker in the detection of adenocarcinoma(ADn),atypical small acinar proliferation(ASAp),regular prostatic tissue(RpT)and regular prostatic tissue alternated by atrophy spotlights(RpTa)in transrectal biopsy guided by ultrasonography of patients with suspected prostate ***:Analysis of 34 patients who underwent ultrasound-guided transrectal biopsy with subsequent analysis by H&E staining and 34βE12 labeling for elucidation of neoplasms or diseased tissues with doubtful ***:The marker 34βE12 showed negativity in 100%of the neoplasms ADn,positivity in 100%of the benign prostatic tissues(RpT and RpTa);the patients with ASAp presented positivity(20%)and negativity(80%).The chi-square test(χ2)showed that there is an association(χ2=29.55 and pps,that is,the 34βE12 marker has a significant value(ppatients with prostatic neoplasia and benign prostatic *** and Conclusion:With the early screening of prostate cancer in the modern era,pathologists have become increasingly challenged to diagnose small outbreaks of cancer when only a few atypical glands are present in transrectal biopsy-guided *** 34βE12 marker becomes an important tool in elucidating diagnoses such as ADn and ASAp.

Diagnostic Value between 1984 and 2018 of Transrectal Biopsy Guided by Ultrasonography after Radical prostatectomy

Objectives—to determine correlation between GSs(Gleason scores)on needle biopsy and Rp(radical prostatectomy),evaluating diagnostic tests on biopsy and Rp within the last years,between 1984 and ***—analysis of 100 patients,diagnosed with pCa(prostate cancer)needle biopsy using 18-gauge needle,who underwent Rp with lymphadenectomy and for which preoperative and postoperative GSs were *** group analysis used three categorization schemes for differentiation:mild,moderate and poor for the whole group and we determined SE(sensitivity),Sp(specificity),pVpR(positive predictive value),negative predictive value and ***—we found that 42%of the patients had no changes between GS on biopsy and prostatectomy,while 20%were overgraded and 38%undergraded by needle *** of+1 point in GS(32%)or-1 point(17%)was the most *** patients were classified as moderately differentiated by biopsies(78 and 35%in scheme 1 and 2 or 3,respectively),while 43%of patients received an intermediate differentiation *** accuracy varied from 44 to 76%for the analysis of all three ***—there are differences in correlation between GS on biopsy and on surgical specimen,and Gleason’s graduation also depends on the experience of the *** have shown that sextant biopsies using 18-gauge and a same group of pathologists showed acceptable concordance values(42%)between the GS on biopsy and prostatectomy.

Delirium in hospitalized patients: Cohort observational in infirmary of medical clinic

Objectives: To determine prevalence, incidence and risk factors of delirium in hospitalized patients in the infirmary of medical clinic of the Lauro Wanderley University Hospital. Method: An observational cohort study evaluating 140 patients over 18 years between admission and discharge. The survey instrument was the Confusion Assessment Method. Results: The prevalence of delirium was 5.7% (8/140) and the incidence, 2.1%. In patients with delirium were observed alteration of sleep-wake cycle (6/75%), altered level of consciousness (6/75%), disorganized thinking (5/62, 5%) and psychomotor retardation (5/62 5%). There was a significant association of delirium and age, mechanical restraint in bed, prior history of cognitive impairment, auditory and visual sensory deficit, psychiatric hospitalization, poor health status and mortality. Conclusions: The prevalence of delirium at admission was lower than that found in other studies, but it was a predictor factor of hospital mortality.

The objective of this work was to verify the responses in agronomic and physiological characters of lettuce seedlings when submitted to different doses of humic acids and the presence or absence of inoculation of the bacterium Azospirillum brasilense. The experiment was carried out at the protected Cultivation Station and Biological Control Teacher Mário César Lopes, belonging to the Experimental Stations Nucleus of the State University of the West of paraná. The experimental design was a randomized block design, in a 5 × 2 factorial scheme, with four repetitions, totaling 40 plots. The first factor consisted of five solutions containing humic acid (100%, 75%, 50%, 25% and 0%) and the second factor was the presence or absence of the diazotrophic bacterium Azospirillum brasilense obtained by commercial product. Germination tests were carried out in addition to the determination of agronomic characters such as the number of fully expanded leaves, plant height, stem diameter, main root length, shoot dry mass, root dry mass, total dry mass, and Dickson quality index. The association of diazotrophic bacteria and humic acids did not influence the emergency rate, mean emergence time, mean speed of emergency, emergence speed index, plant height, and root length of lettuce seedlings. The dilutions of humic acids, regardless of inoculation with bacteria diazotrophic favored the development of the number of leaves, stem diameter and Dickson quality index. Inoculation of lettuce seeds with diazotrophic bacteria associated with the addition of humic acids, favored the development of the dry mass of root and total dry matter.

Eucalyptol prevents pilocarpine-induced seizure and neuronal damage in mice, through the cholinergic, monoaminergic and antioxidant pathways

Current antiepileptic drugs can inhibit seizure occurrence but are not effective in preventing its onset. Moreover, they produce several side effects, which may impact the efficacy of the treatment. In addition, it has been stimulating the prospection of new molecules isolated from aromatic plants, with potential anticonvulsant and neuroprotective activities and less side effects. This study aimed to evaluate, though behavioural and neurochemical methodologies, the anticonvulsant, and neuroprotective effects of eucalyptol on mice subjected to the pilocarpine-induced seizure model. Eucalyptol (100, 200 and 400 mg/kg, p.o.) was administered in mice prior to pilocarpine (350 mg/kg, i.p.) and the following behavioural parameters were assessed: Latency to First Seizure (LFS), Seizure Intensity (SI) and Latency to Death (LD). In addition, an oxotremorine-induced tremors test was performed to evaluate the cholinergic system involvement on the eucalyptol effects. Neurochemical tests were also performed, including determination of hippocampal (HC) concentration of thiobarbituric acid reactive substances (TBARS) and nitrite/nitrate and striatal (ST) concentration of noradrenaline, dopamine, and serotonin. Lastly, histopathological, and morphometric hippocampal analysis were conducted. Eucalyptol increased the latency to first seizure and latency to death, inhibited oxotremorine-induced tremors, decreased hippocampal TBARS and nitrite/nitrate overproduction, increased striatal noradrenaline and dopamine levels and prevented hippocampal neurodegeneration. These results demonstrate the potential anticonvulsant, neuroprotective and antioxidant effects of eucalyptol, probably through a conjunction of mechanisms including muscarinic cholinergic antagonism, oxidative stress mitigation and the monoaminergic system modulation, which appears to effectively control the seizure onset.
