The Phytochemical investigation on MeOH extract on the bark of Aristolochia brasiliensis Mart.&Zucc(Aristolochi-aceae)led to the isolation of major compound(1)as light brown grainy *** compound was identified as 1-(4-hydroxybenzyl)-1,2,3,4-tetrahydroisoquinoline-6,7-diol(higenamine)on the basis of spectroscopic analysis,including 1D and 2D NMR *** compound was evaluated for its antimycobacterial activity against Mycobacterium indicus pranii(MIP),using Galleria mellonella larva as an in vivo infection *** survival of MIP infected larvae after a single dose treatment of 100 mg/kg body weight of higenamine was 80%after 24 *** the compound exhibited a dose dependent activity,as evidenced by the reduction of colony density from 10^(5) to 10^(3) CFU for test concentrations of 50,100,150 and 200 mg/kg body weight *** IC50 value for higenamine was 161.6 mg/kg body weight as calculated from a calibration *** analysis showed that,a complete inhibition of MIP in the *** could be achieved at 334 mg/kg body *** the fact that MIP has been found to be highly resistant against isoniazid(INH)in an in vitro assay model,in this study the microbe was highly susceptible to this standard anti-TB *** isolation of higenamine from the genus Aristolochia and the method used to evaluate its in vivo antimycobacterial activity in *** are herein reported for the first time.

In Silico Evaluation of Anti-Malarial Agents from Hoslundia opposita as Inhibitors of Plasmodium falciparum Lactate Dehydrogenase (PfLDH) Enzyme

Malaria has continued to be a health and economic problem in Africa and the world at large. Many anti-malarial drugs have been rendered ineffective due to the emergence of resistant strains of Plamodium falciparum. A key malaria parasite enzyme in glycolytic pathway, P. falciparum lactate dehydrogenase (PfLDH) is specially targeted for anti-malarial drugs development. Thus, the aim of this investigation was to determine the in silico inhibition effects of antimalarial compounds from Hoslundia opposita Vahl. namely hoslundin, hoslundal and hoslunddiol on PfLDH enzyme. The compounds were docked to the three-dimensional structure of PfLDH as enzyme using AutoDock Vina in PyRx virtual screening software. Binding affinity and position of the inhibitors were evaluated using PyMol software. The PfLDH enzyme showed two binding sites: the cofactors binding site (Site A) and secondary binding site (Site B). In the absence of the cofactor all ligands showed higher affinity than NADH, and were bound to the cofactors binding site (Site A). When docked in the presence of the cofactor, site B was the preferred binding site. Binding to cofactor site with higher binding energy than NADH suggests that these ligands could act as preferential competitive inhibitors of PfLDH. However, the binding to site B also suggests that they may be non-competitive allosteric inhibitors. Amino acid residues Gly99, Asn140, Phe100 and Thr97 were indicated to form hydrogen bonds with Hoslundin. Hoslunddiol showed hydrogen bonding with Thr97 and Met30, while Hoslundal formed hydrogen bond with Thr101 and Asn140.

AIM: To develop a tool for detailed analysis of spirally acting anesthetic and analgesic agents. METHODS: Studies were done on visually identified motor neurons in 400 uuuuuuuum thick spinal cord slices from 14-23 d old rats using patch clamp techniques. Ethanol was used as a prototype general anesthetic agent. RESULTS: Cell bodies in the ventrolateral horn identified as motor neurons by retrograde fluorescent labeling had a mean dimension of 32 ± 5 /Mm (x ± s, n = 25). Mean resting potential was - 62. 8 ± 2. 4 mV; input resistance was 44±24 MΩ (n = 19). Threshold was -44± 7 mV, and action potential amplitude 101 ± 9 mV from baseline. Ethanol concentrations at and below 50-200 mmol/L decreased motor neuron excitability to the injected current; there was no effect on resting potential, but a variable reversible increase in input resistance. Analysis of a ten step protocol to decrease postoperative spinal wound infections

AIM To define a ten-step protocol that reduced the incidence of surgical site infection in the spine surgery practice of the senior author and evaluate the support for each step based on current *** In response to unexplained increased infection rates at our institution following spine surgery,a ten-step protocol was implemented:(1) preoperative glycemic management based on hemoglobin A1 c(HbA1 c);(2) skin site preoperative preparation with 2% chlorhexidine gluconate disposable cloths;(3) limit operating room traffic;(4) cut the number of personnel in the room to the minimum required;(5) absolutely no flash sterilization of equipment;(6) double-gloving with frequent changing of outer gloves;(7) local application of vancomycin powder;(8) re-dosing antibiotic every 4 h for prolonged procedures and extending postoperative coverage to 72 h for high-risk patients;(9) irrigation of subcutaneous tissue with diluted povidone-iodine solution after deep fascial closure; and(10) use of Dura Prep skin preparation at the end of a case before skin closure. Through an extensive literature review,the current data available for each of the ten steps was *** Use of vancomycin powder in surgical wounds,routine irrigation of surgical site,and frequent changing of surgical gloves are strongly supported by the literature. Landscape effects on the thermotolerance of carabid beetles and the role of behavioral thermoregulation

Physiological thermotolerance and behavioral thermoregulation are central to seasonal cold adaptation in ectothermic *** species with enhanced mobility,behavioral responses may be of greater importance in the cold stress *** the carabid beetles as a study organism,the current study compared physiological thermotolerance and behavioral thermoregulation in carabid species inhabiting cereal fields in different landscape contexts,from fine grain heterogeneous“complex”landscapes to homogenous“simple”*** thermotolerance was determined via measurement of the CTmin and chill coma *** responses to cold temperature exposure were determined employing a purpose built arena,and thoracic temperature measured to estimate the efficacy of the behavior as a form of behavioral *** revealed an influence of landscape composition on the cold tolerance of carabid beetles,although species differed in their sensitivity to landscape intensification.A reduced effect of landscape on the thermotolerance of larger carabid beetles was observed,thought to be the consequence of greater mobility preventing local acclimation to microclimatic variation along the landscape intensification *** into behavioral thermoregulation of the 3 largest species revealed burrowing behavior to be the main behavioral response to cold stress,acting to significantly raise carabid body *** finding highlights the importance of behavioral thermoregulation as a strategy to evade cold *** use of behavioral thermoregulation may negate the need to invest in physiological thermotolerance,further offering explanation for the lack of landscape effect on the physiological thermotolerance of larger carabids.

Patients suffering from liver cirrhosis(LC) frequently require non-hepatic abdominal surgery,even before liver *** is an important risk factor itself for surgery,due to the higher than average associated morbidity and *** high surgical risk occurs because of the pathophysiology of liver disease itself and to the presence of contributing factors,such as coagulopathy,poor nutritional status,adaptive immune dysfunction,cirrhotic cardiomyopathy,and renal and pulmonary dysfunction,which all lead to poor *** evaluation of these factors and the degree of liver disease can help to reduce the development of complications both during and after abdominal *** the emergency setting,with the presence of decompensated LC,alcoholic hepatitis,severe/advanced LC,and significant extrahepatic organ dysfunction conservative management is preferred.A multidisciplinary,individualized,and specialized approach can improve outcomes;preoperative optimization after risk stratification and careful management are mandatory before *** techniques can also improve *** review the impact of LC on surgical outcome in non-hepatic abdominal surgeries required in this cirrhotic population before,during,and after surgery.
