Sin1-mediated mTOR signaling in cell growth, metabolism and immune response

The mammalian target of rapamycin(mTOR) is an evolutionarily conserved Ser/Thr protein kinase with essential cellular function via processing various extracellular and intracellular inputs. Two distinct multi-protein m TOR complexes(mTORC), m TORC1 and m TORC2, have been identified and well characterized in eukaryotic cells from yeast to human. Sin1, which stands for ■,also known as mitogen-activated protein kinase■ (MAPKAP)1, is an evolutionarily conserved adaptor protein. Mammalian Sin1 interacts with many cellular proteins, but it has been widely studied as an essential component of m TORC2, and it is crucial not only for the assembly of m TORC2 but also for the regulation of its substrate specificity. In this review, we summarize our current knowledge of the structure and functions of Sin1, focusing specifically on its protein interaction network and its roles in the m TOR pathway that could account for various cellular functions of m TOR in growth,metabolism, immunity and cancer.

Powdery Li^(+)-imprinted manganese oxides adsorbent was widely used to the recovery of Li^(+),but there are some difficulties,such as poor stability in acid solution,inconvenience of operation and separation.In this work,a useful hydrogel composite based H_(4)Mn_(3.5)Ti_(1.5)O_(12)/reduced graphene oxide/polyacrylamide(HMTO-rGO/PAM)was fabricated by thermal initiation method with promising stable,conductive and selective properties.The resulting materials were characterized by field emission scanning electron microscope,infrared absorption spectrum,X-ray diffraction,X-ray photoelectron spectroscopy and electrochemical techniques.The recovery of Li^(+)was investigated using HMTO-rGO/PAM from brine by a separated two-stage sorption statically and electrically switched ion exchange desorption process.The adsorption capacity of 51.5 mg·g^(-1)could be achieved with an initial Li^(+)concentration of 200 mg·L^(-1)in pH 10,at 45℃ for 12 h.Li^(+)ions could be quickly desorbed by cyclic voltammetry(CV)in pH 3,0.1 mol·L^(-1)HCl/NH;Cl accompanying the exchange of Li^(+)and H+(NH;)and the transfer of LMTO-rGO/PAM to HMTO-rGO/PAM.

Proper control of B cell growth and metabolism is crucial for B-cell-mediated immunity,but the underlying molecular mechanisms remain incompletely understood.In this study,Sin1,a key component of mTOR complex 2(mTORC2),specifically regulates B cell growth and metabolism.Genetic ablation of Sin1 in B cells reduces the cell size at either the transitional stage or upon antigen stimulation and severely impairs metabolism.Sin1 deficiency also severely impairs B-cell proliferation,antibody responses,and anti-viral immunity.At the molecular level,Sin1 controls the expression and stability of the c-Myc protein and maintains the activity of mTORC1 through the Akt-dependent inactivation of GSK3 and TSC1/2,respectively.Therefore,our study reveals a novel and specific role for Sin1 in coordinating the activation of mTORC2 and mTORC1 to control B cell growth and metabolism.

目的基于全球营养领导层诊断营养不良标准(global leadership initiative on malnutrition,GLIM)分析老年住院患者的营养状况。方法选择2021年3月至2022年3月在四川大学华西上锦医院住院的5236例老年患者(年龄≥65岁)为研究对象,采用营养风险筛查2002评分并遵循GLIM标准步骤分析老年住院患者营养状况。结果老年住院患者营养风险发生率为30.29%,GLIM标准下营养不良发生率为15.64%。根据GLIM标准分为营养不良组和非营养不良组。营养不良组血红蛋白、白蛋白、甘油三酯、胆固醇、体重指数均低于非营养不良组,差异有统计学意义(P均<0.05);非营养不良组住院时间较营养不良组更短、住院花费更少,差异有统计学意义(P均<0.001)。结论GLIM标准适用于对老年住院患者的营养状况进行诊断。老年住院患者营养不良发生率较高,应重视老年住院患者营养状况。

Phosphorus atomic chains, the narrowest nanostructures of black phosphorus （BP）, are highly relevant to the in-depth development of BP-based one-dimensional （1D） nano-electronics components. In this study, we report a top-down route for the preparation of phosphorus atomic chains via electron beam sculpturing inside a transmission electron microscope （TEM）. The growth and dynamics （i.e., rupture and edge migration） of 1D phosphorus chains are experimentally captured for the first time. Furthermore, the dynamic behavior and associated energetics of the as-formed phosphorus chains are further investigated by density functional theory （DFT） calculations. It is hoped that these 1D BP structures will serve as a novel platform and inspire further exploration of the versatile properties of BP.

目的采用人胚肺二倍体细胞(human embryonic lung diploid cell)KMB17株对柯萨奇病毒B族(Coxsackievirus B,CVB)1、2、3型进行适应性培养,探索应用KMB17细胞培养CVB1-3型病毒的相关参数,为基于KMB17细胞研发CVB多价疫苗提供参考。方法将CVB 1-3型分别设置8个不同的感染复数(mulitiplicity of infection, MOI)梯度(0.05、0.1、0.15、0.2、0.25、0.3、0.35、0.4),在KMB17细胞上进行培养观察,对病毒培养结果进行统计分析。结果 CVB 1-3型病毒均能KMB17细胞中复制,并使细胞产生明显的细胞病变(cytopathic effect, CPE)。其中CVB 1型在KMB17细胞上适宜的种毒MOI值为0.25~0.3,病毒收获液的平均滴度为7.25 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为96.1~97.6 h;CVB 2型在KMB17细胞上适宜的种毒MOI值为0.20~0.25,病毒收获液平均滴度为7.375 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为74.8~76.8 h。CVB 3型在KMB17细胞上适宜的种毒MOI值为0.30~0.35,病毒收获液的平均滴度为7.25 lgCCID50/mL,适宜MOI范围内病毒培养时间的95%置信区间为82.7~84.7 h。结论 KMB17为一株理想的用于CVB1、2、3型培养的基质细胞,可为基于KMB17细胞研发CVB多价疫苗提供参考。

一体化仿真模型是智能气田建设的重要基础,如何利用数据驱动模型高效运转已成为亟待解决的关键难题。针对数据源分散、可用性差及与模型交互程度低等问题,开展了以下技术攻关:(1)结合数据服务总线(DSB)与PI OLEDB接口技术,实现多源结构化数据的有效集成与实时抽取,满足一体化模型对不同时间维度生产数据的业务需求;(2)建立数据质量规则库和自动扫描制度,实现数据质量的有效管控,确保一体化模型的正常运行;(3)开发基于数据流引擎技术的数据交互工具,实现了综合数据库与仿真模型的自动交互,其优势在于应用场景改变时,能够应用灵活编排基础服务的方法高效复用前期数据建设成果,大幅提高开发效率。研究表明,与国内外同类技术相比,应用数据流引擎技术突破了一体化模型与外部数据资源的交互壁垒,打破了该领域数据应用定制化开发的服务模式,为智能气田建设奠定了技术基础。
