Background:The transient receptor potential canonical(TRPC) channels,which are abundantly expressed in central nervous system(CNS),are Ca permeable nonselective cation channels with different biological *** studies indicated that TRPC1 and TRPC6 channels were likely to be involved in the pathological process of mechanical hyperalgesia and nociceptor *** whether TRPC channels are involved in melittin- or formalin-induced inflammatory nociceptive responses remains ***:In this study,we used behavioral tests combined with pharmacological manipulations to explore the potential roles of TRPC channels in inflammatory pain and ***:In this study,we used the well-established bee-venom(melittin) and formalin *** flinch test was to examine whether TRPC channels are involved in inflammatory paininduced primary *** pain sensitivity of rats was determined by testing paw withdrawal thermal latency(PWTL,s) in response to heat stimuli applied to the injected *** pain hypersensitivity was tested as the paw withdrawal mechanical threshold(PWMT,g).Results:Post- and pre-treatment of the primary injury site with *** of SKF96365,a TRPC3/6/7 antagonist,significantly suppressed the development of inflammatory pain-evoked persistent spontaneous nociception(PSN) and hypersensitivity(thermal and mechanical). Background:The effects of acupuncture have varied in different randomized controlled trials(RCTs),and there are many factors that influence treatment effect of acupuncture in different outcomes,with conflicting ***:To identify factors and their impact on the treatment effect of acupuncture in different ***:Acupuncture RCTs were searched from 7 databases including Medline(Pub Med),Embase,Cochrane Central Register of Controlled Trials,china National Knowledge Infrastructure,Wanfang Database,VIP Database,and china Biology Medicine disc between January 1st,2015 and December 31st,*** studies must compare acupuncture to no acupuncture,sham acupuncture,or waiting lists,and report at least 1patient-important outcome.A multi-level meta-regression was conducted using a 3-level robust mixed model and univariate analyses were performed for all independent variables,even those excluded from the multivariable model due to *** used thresholds of 0.2 and 0.4 for the difference of standardized mean differences(SMDs),categorising them as small(0.4) ***:The pain construct analysis involved 211 effect estimates from 153 studies and 14 independent ***-frequency acupuncture treatment sessions produced larger effects compared to low-frequency sessions [large magnitude,the difference of adjusted SMDs 0.46,95% confidence interval(CI) 0.07 to 0.84;P=0.02].The non-pain symptoms construct analysis comprised 323 effect estimates from 231 studies and 15 independent *** acupuncture showed moderately larger effects when compared to non-penetrating acupuncture(0.30,95% CI 0.06 to 0.53;P=0.01).The function construct analysis included 495 effect estimates from 274 studies and 14 independent *** acupuncture and the flexible acupuncture regimen showed moderately larger effects,compared to non-penetrating acupuncture and fixed regimen,respectively(0.40,95% CI 0

Use of Evidence-Based Research Approach in RCTs of Acupuncture-Related Therapies for Primary Dysmenorrhea: A Meta-Research

Objective To assess the use of evidence-based research(EBR)approach in randomized controlled trials(RCTs)of acupuncture-related therapies for primary dysmenorrhea(PD).Methods PubMed,EMBASE,Cochrane Library,china National Knowledge Infrastructure,Wanfang Database,Chinese biomedical Database,and china Science and Technology Journal Database were searched from January 2013 to December 2022 for RCTs of acupuncture on *** full text and references of each RCT were read to assess whether systematic reviews(SRs)or other types of studies with similar research questions and end-users’perspectives were cited to justify and design the *** addition,the discussion section were analyzed to evaluate whether trials placed the new result in the existing SRs to draw a *** logistic regression was used to find variables that associated with 3 aspects of EBR approach:(1)citing clinical studies for justification,(2)citing relevant studies that obtain the perspectives of end users,and(3)citing clinical studies for results *** Of 473 RCTs included,45.67%(216)of the trials cited relevant similar studies,21.56%(102)referenced to the studies that collected end-users’perspectives,and 10.99%(52)placed result in the context of the previous *** RCTs appropriately applied EBR *** all the included studies,3.17%(15)of the trials used SRs to inform study questions but none of them used updated SRs with acceptable quality;1.05%(5)of the trials cited SRs of end-user’s perspectives in the justification and design of the study,and only 1 trial added results in existing SR to draw a *** of publication,language,funding,registration,ethical approval and number of sites were significantly associated with 1 of the 3 aspects of EBR *** Few RCTs in acupuncture-related therapies for PD used the EBR approach to minimize research ***,research institutes,funding agencies,ethics committees,journals and peer

Antibody-drug conjugates(ADCs)are a new type of targeting antibodies that conjugate with highly toxic anticancer drugs via chemical linkers to exert high specificity and efficient killing of tumor cells,thereby attracting considerable attention in precise oncology ***(Cet)is a typical antibody that offers the benefits of good targeting and safety for individuals with advanced and inoperable cutaneous squamous cell carcinoma(cSCC);however,its anti-tumor activity is limited to a single ***(CisPt)shows good curative effects;however,its adverse effects and non-tumor-targeting ability are major *** this study,we designed and developed a new ADC based on a new cytotoxic platinum(IV)prodrug(C8Pt(IV))and *** so-called antibody-platinum(IV)prodrugs conjugates,named Cet-C8Pt(IV),showed excellent tumor targeting in ***,it accurately delivered C8Pt(IV)into tumor cells to exert the combined anti-tumor effect of Cet and ***,metabolomic analysis showed that Cet-C8Pt(IV)promoted cellular apoptosis and increased DNA damage in cSCC cells by affecting the vitamin B6 metabolic pathway in tumor cells,thereby further enhancing the tumor-killing ability and providing a new strategy for clinical cancer treatment using antibody-platinum(IV)prodrugs conjugates.

The dysfunction of innate immunity components is one of the major drivers for ulcerative colitis(UC),and increasing reports indicate that the gut microbiome serves as an intermediate between genetic mutations and UC ***,we find that the IL-17 receptor subunit,CMTM4,is reduced in UC patients and dextran sulfate sodium(DSS)-induced *** deletion of CMTM4(Cmtm4^(-/-))in mice leads to a higher susceptibility to DSS-induced colitis than in wild-type,and the gut microbiome significantly changes in *** causal role of the gut microbiome is confirmed with a cohousing *** further identify that S100a8/9 is significantly up-regulated in Cmtm4^(-/-)colitis,with the block of its receptor RAGE that reverses the phenotype associated with the CMTM4 ***4 deficiency rather suppresses S100a8/9 expression in vitro via the IL17 pathway,further supporting that the elevation of S100a8/9 in vivo is most likely a result of microbial *** together,the results suggest that CMTM4 is involved in the maintenance of intestinal homeostasis,suppression of S100a8/9,and prevention of colitis *** study further shows CMTM4 as a crucial innate immunity component,confirming its important role in UC development and providing insights into potential targets for the development of future therapies.

Subcutaneous injection of bee venom causes long-term neural activation and hypersensitization in the dorsal horn of the spinal cord,which contributes to the development and maintenance of various pain-related *** unique behavioral 'phenotypes' of nociception and hypersensitivity identified in the rodent bee venom test are believed to reflect a complex pathological state of inflammatory pain and might be appropriate to the study of phenotype-based mechanisms of pain and *** this review,the spinal processing of the bee venom-induced different 'phenotypes' of pain and hyperalgesia will be *** accumulative electrophysiological,pharmacological,and behavioral data strongly suggest that different 'phenotypes' of pain and hyperalgesia are mediated by different spinal signaling *** the phenotype-based mechanisms of pain might be useful in development of novel therapeutic drugs against complex clinic pathological pain.

＜正＞Objective:Previous behavioral studies have shown that peripheral tissue injury could produce marked inflammatory responses,causing dramatic spontaneous pain and hypersensitivity,accompanied by plastic changes in both peripheral and central nervous *** date,mechanisms underlying pain-associated spinal cord plasticity have been largely investigated,however,it is still unclear whether there exists cortical plasticity at the higher level of pain processing（such as the primary somatosensory cortex,SI） and if so,what is the specific mechanism for this ***:We performed simultaneous recordings of ensemble neuronal encoding in primary somatosensory cortex of freely moving rats,observed the organization and response properties of deeper layer wide-dynamic-range（WDR） neurons that were preferentially responsive to noxious mechanical stimulation,and nociceptive specific（NS） neurons that were responsive to only noxious *** then examined the stimulusresponse functions of the two classes of neurons obtained during noxious thermal *** bee venom （B V） test model,we further evaluated the inflammatory pain-induced changes in firing patterns of these neuronal ***:A total of 106 neurons were encountered in *** population of nociceptive neurons included 63（59%） WDR and 15（14%） NS *** neurons were distributed over a wider area of the cortex than NS *** noxious thermal stimuli activated both the nociceptive WDR and NS *** BV test model, we demonstrated that in the first hour after BV injection,most of the neurons within S1 showed enhancement of spike frequency and mono-phasic excitation which lasted for 20-60 *** than 70%of neurons showed responsiveness to the BV-induced persistent *** with NS neurons,the average spontaneous activity of WDR neurons showed a significant increase after ***:Our results strongly suggest that the SI is involved in nociception in freely m
