Certain amino acids changes in the human Na^(+)/K^(+)-ATPase pump,ATPase Na^(+)/K^(+)transporting subunit alpha 1(ATP1A1),cause Charcot-Marie-Tooth disease type 2(CMT2)disease and refractory seizures.To develop in vivo models to study the role of Na^(+)/K^(+)-ATPase in these diseases,we modified the Drosophila gene homolog,Atpα,to mimic the human ATP1A1 gene mutations that cause CMT2.Mutations located within the helical linker region of human ATP1A1(I592T,A597T,P600T,and D601F)were simultaneously introduced into endogenous Drosophila Atpαby CRISPR/Cas9-mediated genome editing,generating the Atpα^(TTTF)model.In addition,the same strategy was used to generate the corresponding single point mutations in flies(Atpα^(I571T),Atpα^(A576T),Atpα^(P579T),and Atpα^(D580F)).Moreover,a deletion mutation(Atpα^(mut))that causes premature termination of translation was generated as a positive control.Of these alleles,we found two that could be maintained as homozygotes(Atpα^(I571T)and Atpα^(P579T)).Three alleles(Atpα^(A576T),Atpα^(P579)and Atpα^(D580F))can form heterozygotes with the Atpαmut allele.We found that the Atpαallele carrying these CMT2-associated mutations showed differential phenotypes in Drosophila.Flies heterozygous for Atpα^(TTTF)mutations have motor performance defects,a reduced lifespan,seizures,and an abnormal neuronal morphology.These Drosophila models will provide a new platform for studying the function and regulation of the sodium-potassium pump.

Static recrystallization behaviors and mechanisms of 7Mo super-austenitic stainless steel with undissolved sigma precipitates during double-stage hot deformation

Static recrystallization(SRX)behaviors and corresponding recrystallization mechanisms of 7Mo super-austenitic stainless steel were studied under different deformation conditions.The order of influence of deformation parameters on static recrystallization behaviors,from high to low,is followed by temperature,first-stage strain and strain rate.Meanwhile,the effect of holding time on static recrystallization behaviors is significantly controlled by temperature.In addition,with the increase in temperature from 1000 to 1200°C,the static recrystallization mechanism evolves from discontinuous static recrystallization and continuous static recrystallization(cSRX)to metadynamic recrystallization and cSRX,and finally to cSRX.The cSRX exists at all temperatures.This is because high stacking fault energy(56 mJ m−2)promotes the movement of dislocations,making the deformation mechanism of this steel is dominated by planar slip of dislocation.Large undissolved sigma precipitates promote static recrystallization through particle-stimulated nucleation.However,small strain-induced precipitates at grain boundaries hinder the nucleation of conventional SRX and the growth of recrystallized grains,while the hindering effect decreases with the increase in temperature.

Globally,grassland degradation is an acute ecological problem.In alpine grassland on the Tibetan Plateau,increased densities of various small mammals in degraded grassland are assumed to intensify the degradation process and these mammals are subject to lethal control.However,whether the negative impact of small mammals is solely a result of population size or also a result of activity and behavior has not been tested.In this study,we use plateau pika as a model to compare population size,core area of colony,and the number of burrow entrances and latrines between lightly and severely degraded grassland.We test whether the alleged contribution of pika to grassland degradation is a result of increased population size or increased burrowing activities of individuals in response to lower food abundance.We found that grassland degradation resulted in lower plant species richness,plant height,and biomass.Furthermore,the overall population size of pika was not significantly affected by location in lightly and severely degraded grassland.However,pika core areas in severely grassland degradation were significantly larger and had significantly higher densities of burrows and latrines.Our study provides convincing evidence that habitat-induced changes in the behavior of small,burrowing mammals,such as pika,can exacerbate grassland degradation.This finding has significant implications for managing small mammals and restoring degraded grassland ecosystems.

Mild traumatic brain injury(mTBI)-induced post-traumatic headache(PTH)is a pressing public health concern and leading cause of disability worldwide.Although PTH is often accompanied by neurological disorders,the exact underlying mechanism remains largely unknown.Identifying potential biomarkers may prompt the diagnosis and development of effective treatments for mTBI-induced PTH.In this study,a mouse model of mTBI-induced PTH was established to investigate its effects on cerebral structure and function during short-term recovery.Results indicated that mice with mTBI-induced PTH exhibited balance deficits during the early post-injury stage.Metabolic kinetics revealed that variations in neurotransmitters were most prominent in the cerebellum,temporal lobe/cortex,and hippocampal regions during the early stages of PTH.Additionally,variations in brain functional activities and connectivity were further detected in the early stage of PTH,particularly in the cerebellum and temporal cortex,suggesting that these regions play central roles in the mechanism underlying PTH.Moreover,our results suggested that GABA and glutamate may serve as potential diagnostic or prognostic biomarkers for PTH.Future studies should explore the specific neural circuits involved in the regulation of PTH by the cerebellum and temporal cortex,with these two regions potentially utilized as targets for non-invasive stimulation in future clinical treatment.

Advances in Polymethine Dyes for Near-Infrared Organic Photodiodes

Near-infrared organic photodiodes (NIR OPDs) have tremendous potential in industrial, military, and scientific applications, due to their unique features of lightweight, low toxicity, high structural flexibility, cooling-system-free, etc. However, the overall performance of currently available NIR OPDs still lags behind the commercial inorganic photodetectors, ascribed to the critical challenge of realizing organic semiconductors with sufficiently low optical bandgap and excellent optoelectronic properties simultaneously. Among various types of NIR-absorbing organic semiconductors, polymethine dyes not only possess advantages of simple synthesis and structural diversity, but also show fascinating optical and aggregation features in the solid state, making them attractive material candidates for NIR OPDs. In this review, after a brief introduction of NIR OPDs and polymethine dyes, we comprehensively summarize the advances of polymethine dyes for broadband and narrowband NIR OPDs, and further introduce their applications in all-organic optical upconversion devices and photoplethysmography sensors. In particular, the relationship between the chemical structure and the aggregation behaviors of polymethine dyes and the device performance is carefully discussed, providing some important molecular insights for developing high performance NIR OPDs.

Coronavirus disease 2019(COVID-19)highlights the importance of rapid and reliable diagnostic assays for the management of virus transmission.here,we developed a one-pot hydrothermal method to prepare Si-FITC nanoparticles(NPs)for the fluorescent immunoassay of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)nucleocapsid protein(N protein).The synthesis of Si-FITC NPs did not need post-modification,which addressed the issue of quantum yield reduction during the coupling reaction.Si-FITC NPs showed two distinct peaks,Si fluorescence atλem=385 nm and FITC fluorescence atλem=490 nm.In the presence of KMnO_(4),Si fluorescence was decreased and FITC fluorescence was enhanced.Briefly,in the presence of N protein,catalase(CAT)-linked secondary antibody/reporter antibody/N protein/capture antibody immunocomplexes were formed on microplates.Subsequently,hydrogen peroxide(H_(2)O_(2))and Si-FITC NPs/KMnO_(4)were injected into the microplate together.The decomposition of H_(2)O_(2)by CAT resulted in remaining of KMnO_(4),which changed the fluorescence intensity ratio of Si-FITC NPs.The fluorescence intensity ratio correlated significantly with the N protein concentration ranging from 0.02 to 50.00 ng/mL,and the detection limit was 0.003 ng/mL,which was more sensitive than the commercial ELISA kit with a detection limit of 0.057 ng/mL.The N protein concentration can be accurately determined in human serum.Furthermore,the COVID-19 and non-COVID-19 patients were distinguishable by this method.Therefore,the ratiometric fluorescent immunoassay can be used for SARS-CoV-2 infection diagnosis with a high sensitivity and selectivity.

Blood neutrophil extracellular traps:a novel target for the assessment of mammary health in transition dairy cows

Background:Mammary health is important for transition dairy cows and has been well recognized to exert decisive effects on animal welfare.However,the factors influencing mammary health are still unclear.Differential somatic cell count(DSCC)could reflect the mastitis risk since it is the percentage of neutrophils plus lymphocytes in total somatic cells and could be reflective of mammary health of dairy cows.This work aimed to investigate the assessment and prognosis of the health of transition cows based on blood neutrophil extracellular traps(NETs).Results:Eighty-four transition Holstein dairy cows were selected.The serum was sampled in all the animals at week 1 pre-and postpartum,and milk was sampled at week 1 postpartum.Based on the DSCC in milk at week 1,cows with lower(7.4%±4.07%,n=15)and higher(83.3%±1.21%,n=15)DSCCs were selected.High DSCC cows had higher levels of red blood cell counts(Phemoglobin(P=0.07),and hematocrit(P=0.05),higher concentrations of serum oxidative variables[reactive oxygen species(Pher levels of serum and milk NETs(Phe assessments performed using levels of the three prepartum serum parameters(NETs,deoxyribonuclease I andβ-casein),the area under the curve(0.973)of NETs was the highest.In addition,the sensitivity(1.00)and specificity(0.93)were observed for the discrimination of these cows using NETs levels with a critical value of 32.2 ng/mL(Phe formation of NETs in blood in transition dairy cows may damage the integrity of the blood-milk barrier and thereby increase the risk for mastitis in postpartum cows.

A novel multiscale simulation framework for low-dimensional memristors

In recent years,the memristor has been widely considered an emerging device,but it has rarely been simulated.An obstacle is the change in the intrinsic atomic level when it works.Using the density functional theory(DFT),this atomic level change in structure cannot be demonstrated.Using molecular dynamics(MD),memristor electronic transport properties cannot be calculated.In this study,we propose a novel multiscale simulation framework merging MD,DFT,and the nonequilibrium Green’s function method,which can reveal not only a memristor’s basic working mechanism but also its transport character.To verify our framework’s availability in guiding innovative memristor design,a new type of memristor,a planar monolayer MoS_(2)-based memristor,is simulated for the first time.The popped S atoms’effect on its carrier transport is revealed,which clarifies the working mechanism of the planar monolayer MoS_(2)-based memory device.We hope that this framework can shed light on the analysis and design of low-dimensional memristors.
