Graphene as adsorbent for highly efficient extraction of modified nucleosides in urine prior to liquid chromatography-tandem mass spectrometry analysis

RNA modifications have been involved in numerous biological processes, and aberrations of these modifications are tightly associated with various diseases including cancer. Herein, we developed graphenebased solid-phase extraction and robust ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) combined with stable isotope-dilution for simultaneous enrichment and accurate determination of 17 modified nucleosides in human urine. We found graphene could effectively adsorb various modified nucleosides in human urine samples. With this method, we identified and quantified these modified nucleosides in urine samples collected from lung cancer patients and healthy *** revealed that the levels of 12 modified nucleosides were all diminished in urine from lung cancer patients, compared with healthy controls. It is worth noting that we demonstrated, for the first time, the presence of 5,2-O-dimethyluridine(m~5U_m) in human urine. Together, we established a robust analytical method for simultaneous determinations of 17 modified nucleosides in human urine, and our results revealed a close correlation between the concentrations of urinary modified nucleosides and the occurrence of lung cancer, implying the potential applications of these modified nucleosides as noninvasive biomarkers for the early detection of lung cancer. Moreover, this study will stimulate future investigations on the regulatory roles of RNA modifications in the initiation and progression of lung cancer.

The coupling control of biological precursors and environmental factors onβ-carotane enrichment in alkaline lacustrine source rocks:A case study from the fengcheng formation in the western Junggar Basin,NW China

The organic-rich mudstones and dolostones of the Permian fengcheng Formation(Fm.)are typically alkaline lacustrine source rocks,which are typified by impressively abundantβ-*** β-carotane has been well acknowledged as an effective indicator of biological sources or depositional ***,the specific biological sources of β-carotane and the coupling control of biological sources and environmental factors on the enrichment of β-carotane in the fengcheng *** *** on a comprehensive investigation of the bulk,molecular geochemistry,and organic petrology of sedimentary rocks and the biochemistry of phytoplankton in modern alkaline lakes,we proposed a new understanding of the biological precursors of β-carotane and elucidated the enrichment mechanism of β-carotane in the fengcheng *** results show that the biological precursors crucially control the enrichment of β-carotane in the fengcheng *** haloalkaliphilic cyanobacteria are the primary biological sources of β-carotane,which is suggested by a good positive correlation between the 2-methylhopane index,7-+8-methyl heptadecanes/C_(max),C_(29%),and β-carotane/C_(max)in sedimentary rocks and the predominance of cyanobacteria with abundantβ-carotene in modern alkaline *** enrichment of β-carotane requires the reducing condition,and the paleoredox state that affects the enrichment of β-carotane appears to have a *** paleoclimate conditions do not considerably impact the enrichment of β-carotane,but they have some influence on the water's paleosalinity by affecting evaporation and *** it does not directly affect the enrichment of β-carotane in the fengcheng Fm.,paleosalinity does have an impact on the cyanobacterial precursor supply and the preservation conditions.

STGNN-LMR:A Spatial–Temporal Graph Neural Network Approach Based on sEMG Lower Limb Motion Recognition

Lower limb motion recognition techniques commonly employ Surface Electromyographic Signal(sEMG)as input and apply a machine learning classifier or Back Propagation Neural Network(BPNN)for ***,this artificial feature engineering technique is not generalizable to similar tasks and is heavily reliant on the researcher’s subject *** contrast,neural networks such as Convolutional Neural Network(CNN)and Long Short-term Memory Neural Network(LSTM)can automatically extract features,providing a more generalized and adaptable approach to lower limb motion *** this approach overcomes the limitations of human feature engineering,it may ignore the potential correlation among the sEMG *** paper proposes a spatial–temporal graph neural network model,STGNN-LMR,designed to address the problem of recognizing lower limb motion from multi-channel ***-LMR transforms multi-channel sEMG into a graph structure and uses graph learning to model spatial–temporal *** 8-channel sEMG dataset is constructed for the experimental stage,and the results show that the STGNN-LMR model achieves a recognition accuracy of 99.71%.Moreover,this paper simulates two unexpected scenarios,including sEMG sensors affected by sweat noise and sudden failure,and evaluates the testing results using hypothesis *** to the experimental results,the STGNN-LMR model exhibits a significant advantage over the control models in noise scenarios and failure *** experimental results confirm the effectiveness of the STGNN-LMR model for addressing the challenges associated with sEMG-based lower limb motion recognition in practical scenarios.

Responses of arbuscular mycorrhizal fungi to straw return and nitrogen fertilizer reduction in a rainfed maize field

Straw return can be used to reduce fertilizer input and improve agricultural sustainability and soil ***,how straw return and reduced fertilizer application affect beneficial soil microbes,particularly arbuscular mycorrhizal fungi(AMF),remains poorly ***,we conducted a five-year field experiment in a rainfed maize field on the Loess Plateau of northwestern *** tested four treatments with straw return combined with four nitrogen(N)application rates,i.e.,100%,80%,60%,and 0%of the common N application rate(225 kg N ha^(-1)year^(-1))in this region,and two reference treatments(full or no N application),with three replicates for each *** colonization was quantified and AMF communities colonizing maize roots were characterized using Illumina *** virtual taxa(VTs)of AMF were identified in root samples,among which VT113(related to Rhizophagus fasciculatus)and VT156(related to Dominikia gansuensis)were the predominant *** root length colonization and AMF VT richness were sensitive to N fertilization,but not to straw return;furthermore,both gradually increased with decreasing N application *** VT composition of the AMF community was also affected by N fertilization,but not by straw return,and the community variation could be well explained by soil available N and phosphorus ***,60%,80%,and full N fertilization produced similar maize ***,our study revealed the response patterns of AMF to straw return and N fertilizer reduction and showed that straw return combined with N fertilizer reduction may be a promising practice to maintain mycorrhizal symbiosis concomitantly with crop productivity.

Neural Modified Extended State Observer for Autonomous Maritime Transportation Vehicles with Actuator Faults and Unmatched Nonlinearities

This paper investigates extended state observer design problem for autonomous maritime transportation vehicles consisting of multiple unmanned aerial vehicles(UAVs)and unmanned surface vehicles(USVs)via the cooperation–competition *** two coordinate transformations,the dynamic model of UAVs and USVs is transformed into a standard secondorder heterogeneous multi-agent system in unified earth-fixed *** order to recover the unavailable velocities,unpredictable fault,total disturbance as well as to estimate the heterogeneous match/unmatched nonlinearities,neural network(NN)is incorporated into the design process of the modified distributed extended state observer(MESO).Based on the Lyapunov stability analysis,it is proved that all the error signals are uniformly ultimately bounded(UUB)and the bounds could be arbitrarily small by choosing appropriate *** results verify the effectiveness of the proposed method.

Vegetation productivity on the southern edge of the Inner Mongolian Plateau,which plays a vital role in the ecological environment and in the arable and pasto-ral production in this region,can be characterized by the NDVI(normalized difference vegetation index).However,the observed NDVI data span only the last~40 *** growth of Pinus tabulaeformis *** strongly correlated with the NDVI,making it a valuable proxy for extend-ing the length of observed NDVI *** this study,we reconstructed an NDVI series for 1776–2021 for the Daqing Mountains,based on a tree-ring width *** reconstructed data accounted for 55%of the variance in the observed data,and its statistical characteristics and validation indicate that the reconstruction is *** correlation analysis demonstrated the consistency of climate signals in central Inner Mongolia in both the arable and pastoral *** results of superposed epoch analysis revealed a good temporal consistency between drought and flood events and the reconstructed NDVI sequence in this region.

A Data-Oriented Method to Optimize Hydraulic Fracturing Parameters of Tight Sandstone Reservoirs

Based on the actual data collected from the tight sandstone development zone, correlation analysis using theSpearman method was conducted to determine the main factors influencing the gas production rate of tightsandstone fracturing. An integrated model combining geological engineering and numerical simulation of fracturepropagation and production was completed. Based on data analysis, the hydraulic fracture parameters wereoptimized to develop a differentiated fracturing treatment adjustment plan. The results indicate that the influenceof geological and engineering factors in the X1 and X2 development zones in the study area differs ***, it is challenging to adopt a uniform development strategy to achieve rapid production increase. Thedata analysis reveals that the variation in gas production rate is primarily affected by the reservoir thickness andpermeability parameters as geological factors. On the other hand, the amount of treatment fluid and proppantaddition significantly impact the gas production rate as engineering factors. Among these factors, the influence ofgeological factors is more pronounced in block X1. Therefore, the main focus should be on further optimizing thefracturing interval and adjusting the geological development well location. Given the existing well location, thereis limited potential for further optimizing fracture parameters to increase production. For block X2, the fracturingparameters should be optimized. Data screening was conducted to identify outliers in the entire dataset, and adata-driven fracturing parameter optimization method was employed to determine the basic adjustment directionfor reservoir stimulation in the target block. This approach provides insights into the influence of geological,stimulation, and completion parameters on gas production rate.
