Ginsenoside modified lipid-coated perfluorocarbon nanodroplets: A novel approach to reduce complement protein adsorption and prolong in vivo circulation

Lipid-coated perfluorocarbon nanodroplets(lp-NDs)hold great promise in bio-medicine as vehicles for drug delivery,molecular imaging and vaccine agents.However,their clinical utility is restricted by limited targeted accumulation,attributed to the innate immune system(IIS),which acts as the initial defense mechanism in humans.This study aimed to optimize lp-ND formulations to mini-mize non-specific clearance by the IIS.Ginsenosides(Gs),the principal components of Panax ginseng,possessing complement inhibition ability,structural similarity to cholesterol,and comparable fat solubi-lity to phospholipids,were used as promising candidate IIS inhibitors.Two different types of ginsenoside-based Ip-NDs(Gs Ip-NDs)were created,and their efficacy in reducing IS recognition was examined.The Gs p-NDs were observed to inhibit the adsorption of C3 in the protein corona(PC)and the generation of SC5b-9.Adding Gs to Ip-NDs reduced complement adsorption and phagocytosis,resulting in a longer blood circulation time in vivo compared to lp-NDs that did not contain Gs.These results suggest that Gs can act as anti-complement and anti-phagocytosis adjuvants,potentially reducing non-specific clear-ance by the IS and improving lifespan.

Ferroelectric-controlled graphene plasmonic surfaces for all-optical neuromorphic vision

Artificial visual systems can recognize desired objects and information from complex environments, and are therefore highly desired for pattern recognition, object detection, and imaging applications. However, state-of-the-art artificial visual systems with high recognition performances that typically consist of electronic devices face the challenges of requiring huge storage space and high power consumption owing to redundant data. Here, we report a terahertz(THz) frequency-selective surface using a graphene split-ring resonator driven by ferroelectric polarization for efficient visual system applications. The downward polarization of the ferroelectric material offers an ultrahigh electrostatic field for doping p-type graphene with an anticipated Fermi level. By optimizing the geometric parameters of the devices and modulating the carrier behaviors of graphene, our plasmonic devices exhibit a tunable spectral response in a range of 1.7–6.0 THz with continuous transmission values. The alloptical neural network using graphene plasmonic surfaces designed in this study exhibited excellent performance in visual preprocessing and convolutional filtering and achieved an ultrahigh recognition accuracy of up to 99.3% in training the Modified National Institute of Standards and Technology(MNIST) handwritten digit dataset. These features demonstrate the great potential of graphene plasmonic devices for future smart artificial vision systems.

A major impedance to neuronal regeneration after peripheral nerve injury (PNI) is the activation of various programmed cell death mechanisms in the dorsal root ganglion. Ferroptosis is a form of programmed cell death distinguished by imbalance in iron and thiol metabolism, leading to lethal lipid peroxidation. However, the molecular mechanisms of ferroptosis in the context of PNI and nerve regeneration remain unclear. Ferroportin (Fpn), the only known mammalian nonheme iron export protein, plays a pivotal part in inhibiting ferroptosis by maintaining intracellular iron homeostasis. Here, we explored in vitro and in vivo the involvement of Fpn in neuronal ferroptosis. We first delineated that reactive oxygen species at the injury site induces neuronal ferroptosis by increasing intracellular iron via accelerated UBA52-driven ubiquitination and degradation of Fpn, and stimulation of lipid peroxidation. Early administration of the potent arterial vasodilator, hydralazine (HYD), decreases the ubiquitination of Fpn after PNI by binding to UBA52, leading to suppression of neuronal cell death and significant acceleration of axon regeneration and motor function recovery. HYD targeting of ferroptosis is a promising strategy for clinical management of PNI.

DPPS: A novel dual privacy-preserving scheme for enhancing query privacy in continuous location-based services

Since smartphones embedded with positioning systems and digital maps are widely used,location-based services(LBSs)are rapidly growing in popularity and providing unprecedented convenience in people’s daily lives;however,they also cause great concern about privacy leakage.In particular,location queries can be used to infer users’sensitive private information,such as home addresses,places of work and appointment locations.Hence,many schemes providing query anonymity have been proposed,but they typically ignore the fact that an adversary can infer real locations from the correlations between consecutive locations in a continuous LBS.To address this challenge,a novel dual privacy-preserving scheme(DPPS)is proposed that includes two privacy protection mechanisms.First,to prevent privacy disclosure caused by correlations between locations,a correlation model is proposed based on a hidden Markov model(HMM)to simulate users’mobility and the adversary’s prediction probability.Second,to provide query probability anonymity of each single location,an advanced k-anonymity algorithm is proposed to construct cloaking regions,in which realistic and indistinguishable dummy locations are generated.To validate the effectiveness and efficiency of DPPS,theoretical analysis and experimental verification are further performed on a real-life dataset published by Microsoft,i.e.,GeoLife dataset.

Background:Findings on the association of genetic factors and colorectal cancer(CRC)survival are limited and inconsistent,and revealing the mechanism underlying their prognostic roles is of great importance.This study aimed to explore the relationship between functional genetic variations and the prognosis of CRC and further reveal the possible mechanism.Methods:We first systematically performed expression quantitative trait locus(eQTL)analysis using The Cancer Genome Atlas(TCGA)dataset.Then,the Kaplan-Meier analysis was used to filter out the survival-related eQTL target genes of CRC patients in two public datasets(TCGA and GSE39582 dataset from the Gene Expression Omnibus database).The seven most potentially functional eQTL single nucleotide polymorphisms(SNPs)associated with six survival-related eQTL target genes were genotyped in 907 Chinese CRC patients with clinical prognosis data.The regulatory mechanism of the survival-related SNP was further confirmed by functional experiments.Results:The rs71630754 regulating the expression of endoplasmic reticulum aminopeptidase 1(ERAP1)was significantly associated with the prognosis of CRC(additive model,hazard ratio[HR]:1.43,95%confidence interval[CI]:1.08-1.88,P=0.012).The results of dual-luciferase reporter assay and electrophoretic mobility shift assay showed that the A allele of the rs71630754 could increase the binding of transcription factor 3(TCF3)and subsequently reduce the expression of ERAP1.The results of bioinformatic analysis showed that lower expression of ERAP1 could affect the tumor immune microenvironment and was significantly associated with severe survival outcomes.Conclusion:The rs71630754 could influence the prognosis of CRC patients by regulating the expression of the immune-related gene ERAP1.Trial Registration:No.NCT00454519(https://clinicaltrials.gov/)

This review addresses the growing disparity between the current state of fertilizer production in China and the evolving demands of green agriculture in the 21st century. It explores major advances in fertilizers, proposes the concept of green intelligent fertilizers and develops new strategies aligned with the principles of green development in fertilizer industry and agriculture.Green intelligent fertilizers may be designed to maximize the synergistic effects among plants, soils, microorganisms, nutrient sources and the environment. This concept emphasizes the integration of industry and agriculture toward green development for entire industry chain, using an interdisciplinary approach to drive the green transformation of fertilizer industry, and promote green and sustainable development of agriculture. By bridging the gap between the current state of fertilizer industry and a growing need for environmentally responsible agricultural practices, this review highlights a path toward harmonizing fertilizer production with the imperatives of green agriculture.

Effective E.coli inactivation of core-shell ZnO@ZIF-8 photocatalysis under visible light synergize with peroxymonosulfate:Efficiency and mechanism

How to utilize inexhaustible solar light as a means of disinfection technology for its cheap and green remains a challenge.In this work,core-shell ZnO@ZIF-8 was synthesized and used for bacterial inactivation synergizing with peroxymonosulfate(PMS)under visible light irradiation.It took 50 min to achieve thorough sterilization for 7.5-log Escherichia coli(E.coli)cells in vis/PMS/ZnO@ZIF-8 system,compared with that 4.5-log reduction completed in vis/PMS/ZnO system under the same conditions.The enhanced photocatalytic disinfection mechanisms of fabricated ZnO@ZIF-8 were investigated by UV-vis diffuse reflectance spectra,electrochemical impedance spectra and Mott-Schottky plots.The promoted bactericidal efficiency was attributed to higher charge-separation efficiency and stronger oxidation ability of photo-generated holes.Moreover,it was found that^(1)O_(2)and·OH induced bacterial cell lesion process,and the former was the main active species.The external reactive oxygen species(ROS)caused a series of cell wall damage,intercellular ROS up-regulation and genome DNA unwinding,finally resulted in irreversible bacterial death.A two-route mechanism in vis/PMS/ZnO@ZIF-8 system was proposed,in which the generation of^(1)O_(2)was supposed as the product of the oxygen oxidation of photo-generated holes and PMS dissociation.Our work is expected to provide advanced information about a low-cost water disinfection technology of visible light photocatalysis.

Associations of daily sedentary behavior,physical activity,and sleep with irritable bowel syndrome:A prospective analysis of 362,193 participants

Background:Irritable bowel syndrome(IBS)substantially affects quality of life and requires early prevention.This study aimed to elucidate the relationships between IBS and daily behaviors,including sedentary behavior(SB),physical activity(PA),and sleep.In particular,it seeks to identify healthy behaviors to reduce IBS risk,which previous studies have rarely addressed.Methods:Daily behaviors were retrieved from self-reported data of 362,193 eligible UK Biobank participants.Incident cases were determined by self-report or health care data according to RomeⅣcriteria.Results:A total of 345,388 participants were IBS-free at baseline,during a median follow-up of 8.45 years,19,885 incident IBS cases were recorded.When examined individually,SB and shorter(≤7 h/day)or longer(>7 h/day)sleep duration were each positively associated with increased IBS risk,and PA was associated with lower IBS risk.The isotemporal substitution model suggested that replacing SB with other activities could provide further protective effects against IBS risk.Among people sleeping≤7 h/day,replacing 1 h of SB with equivalent light PA,vigorous PA,or sleep was associated with 8.1%(95%confidence interval(95%CI):0.901-0.937),5.8%(95%CI:0.896-0.991),and 9.2%(95%CI:0.885-0.932)reduced IBS risk,respectively.For people sleeping>7 h/day,light and vigorous PA were associated with a 4.8%(95%CI:0.926-0.978)and a 12.0%(95%CI:0.815-0.949)lower IBS risk,respectively.These benefits were mostly independent of genetic risk for IBS.Conclusion:SB and unhealthy sleep duration are risk factors for IBS.A promising way to mitigate IBS risk for individuals sleeping≤7 h/day and for those sleeping>7 h/day appears to be by replacing SB with adequate sleep or vigorous PA,respectively,regardless of the genetic predisposition of IBS.

Fast Blood Oxygenation through Hemocompatible Asymmetric Polymer of Intrinsic Microporosity Membranes

Membrane technology has attracted considerable attention for chemical and medical applications,among others.Artificial organs play important roles in medical science.A membrane oxygenator,also known as artificial lung,can replenish O_(2) and remove CO_(2) of blood to maintain the metabolism of patients with cardiopulmonary failure.However,the membrane,a key component,is subjected to inferior gas transport property,leakage propensity,and insufficient hemocompatibility.In this study,we report efficient blood oxygenation by using an asymmetric nanoporous membrane that is fabricated using the classic nonsolvent-induced phase separation method for polymer of intrinsic microporosity-1.The intrinsic superhydrophobic nanopores and asymmetric configuration endow the membrane with water impermeability and gas ultrapermeability,up to 3,500 and 1,100 gas permeation units for CO_(2) and O_(2),respectively.Moreover,the rational hydrophobic–hydrophilic nature,electronegativity,and smoothness of the surface enable the substantially restricted protein adsorption,platelet adhesion and activation,hemolysis,and thrombosis for the membrane.Importantly,during blood oxygenation,the asymmetric nanoporous membrane shows no thrombus formation and plasma leakage and exhibits fast O_(2) and CO_(2) transport processes with exchange rates of 20 to 60 and 100 to 350 ml m^(−2) min^(−1),respectively,which are 2 to 6 times higher than those of conventional membranes.The concepts reported here offer an alternative route to fabricate high-performance membranes and expand the possibilities of nanoporous materials for membrane-based artificial organs.

A thermodynamic perspective on electrode poisoning in solid oxide fuel cells

A critical challenge to the commercialization of clean and high-efficiency solid oxide fuel cell(SOFC)technology is the insuf-ficient stack lifespan caused by a variety of degradation mechanisms,which are associated with cell components and chemical feedstocks.Cell components related degradation refers to thermal/chemical/electrochemical deterioration of cell materials under operating conditions,whereas the latter regards impurities in feedstocks of oxidant(air)and reductant(fuel).This article provides a thermodynamic perspective on the understanding of the impurities-induced degradation mechanisms in SOFCs.The discussion focuses on using thermodynamic ana-lysis to elucidate poisoning mechanisms in cathodes by impurity species such as Cr,CO_(2),H_(2)O,and SO_(2) and in the anode by species such as S(or H_(2)S),SiO_(2),and P_(2)(or PH_(3)).The author hopes the presented fundamental insights can provide a theoretical foundation for search-ing for better technical solutions to address the critical degradation challenges.
