Simultaneous inhibition of PI3Kα and CDK4/6 synergistically suppresses KRAS-mutated non-small cell lung cancer

Objective: Activating KRAS mutations are the most common drivers in the development of non-small cell lung cancer(NSCLC).However, unsuccess of treatment by direct inhibition of KRAS has been proven. Deregulation of PI3K signaling plays an important role in tumorigenesis and drug resistance in NSCLC. The activity of PI3Kα-selective inhibition against KRAS-mutated NSCLC remains largely ***: Cell proliferation was detected by sulforhodamine B assay. Cell cycle distribution and apoptosis were measured by flow cytometry. Cell signaling was assessed by Western blot and immunohistochemistry. RNA interference was used to down-regulate the expression of cyclin D1. Human NSCLC xenografts were employed to detect therapeutic efficacy in ***: CYH33 possessed variable activity against a panel of KRAS-mutated NSCLC cell lines. Although CYH33 blocked AKT phosphorylation in all tested cells, Rb phosphorylation decreased in CYH33-sensitive, but not in CYH33-resistant cells, which was consistent with G1 phase arrest in sensitive cells. Combined treatment with the CDK4/6 inhibitor, PD0332991, and CYH33 displayed synergistic activity against the proliferation of both CYH33-sensitive and CYH33-resistant cells, which was accompanied by enhanced G1-phase arrest. Moreover, down-regulation of cyclin D1 sensitized NSCLC cells to CYH33. Reciprocally, CYH33 abrogated the PD0332991-induced up-regulation of cyclin D1 and phosphorylation of AKT in A549 cells. Co-treatment with these two drugs demonstrated synergistic activity against A549 and H23 xenografts, with enhanced inhibition of Rb ***: Simultaneous inhibition of PI3Kα and CDK4/6 displayed synergistic activity against KRAS-mutated NSCLC. These data provide a mechanistic rationale for the combination of a PI3Kα inhibitor and a CDK4/6 inhibitor for the treatment of KRASmutated NSCLC.

Tongue cancer is an increasingly common disease with high morbidity,Besides clinical obser-vation,biomedical imaging techniques have been investigated for early detection of *** this paper,we proposed a co-registered dual-modality photoacoustic(PA)and *** imaging technique to simultaneously map the functional and structural information ofhuman tongue,which has the potential to detect and diagnose tongue cancer in early stage,Theimaging probe comprises a 20-MHz side-view focused transducer for ultrasound imaging and PAdetection,a light path constructed by a multimode optical fiber,and a prism for PA *** experiments were conducted to evaluate the performance of the system includingpenetration depth,spatial resolution and signal-to-noise *** vivo imaging of animaltumor and human tongue was carried out to show the feasibility of the proposed technique todetect tumor lesions in human *** results of phantom and in vivo experiments suggestthat the proposed technique has the potential to detect the early-stage cancer lesions in humantongue.

A Cost-Efficient Approach to Storing Users'Data for Online Social Networks

As users increasingly befriend others and interact online via their social media accounts,online social networks (OSNs)are expanding *** with the big data generated by users,it is imperative that data storage be distributed,scalable,and *** one of the most significant challenges about this topic is determining how to minimize the cost without deteriorating system *** many storage systems use the distributed key value store,it cannot be directly applied to OSN storage *** because users'data are highly correlated,hash storage leads to frequent inter-server communications,and the high inter-server traffic costs decrease the OSN storage system's scalability. Previous studies proposed conducting network partitioning and data replication based on social ***,data replication increases storage costs and impacts traffic ***,we consider how to minimize costs from the perspective of data storage,by combining partitioning and *** cost-efficient data storage approach supports scalable OSN storage *** proposed approach co-locates frequently interactive users together by conducting partitioning and replication simultaneously while meeting load-balancing *** experiments are undertaken on two real- world traces,and the results show that our approach achieves lower cost compared with state-of-the-art *** we conclude that our approach enables economic and scalable OSN data storage.

Effects of the planarity and heterogeneity of networks on evolutionary two-player games

We study the effects of the planarity and heterogeneity of networks on evolutionary two-player symmetric games by considering four different kinds of networks, including two types of heterogeneous networks: the weighted planar stochastic lattice(a planar scale-free network) and the random uncorrelated scale-free network with the same degree distribution as the weighted planar stochastic lattice; and two types of homogeneous networks: the hexagonal lattice and the random regular network with the same degree k_0= 6 as the hexagonal lattice. Using extensive computer simulations, we found that both the planarity and heterogeneity of the network have a significant influence on the evolution of cooperation, either promotion or inhibition, depending not only on the specific kind of game(the Harmony, Snowdrift, Stag Hunt or Prisoner's Dilemma games), but also on the update rule(the Fermi, replicator or unconditional imitation rules).

Influence of yttrium on the structure and magnetostriction of Fe_(83)Ga_(17) alloy

Polycrystalline Fe83Ga17 alloy rods with various amounts of yttrium were prepared by high vacuum induction melting. It is found that yttrium addition has a significant effect on the structure and magnetostriction of Fes3Ga17 alloy. The small addition of yttrium alters the solidification character and the grain shape of Fe83Ga17 alloy, and as a result, columnar grains with the 〈100〉 preferential direction are pro- duced. Yttrium addition improves the magnetostrictive performance of the as-cast Fes3Ga17 alloy. The magnetostriction values of the as-cast alloy with 0.32at% and 0.64at% yttrium addition go up to 119×10^-6 and 137×10^-6 under 15 MPa compressive stress, respectively. The energy dispersive spectroscopy （EDS） result shows that almost all of the yttrium atoms exist in the Y2Fe17-xGax phase. A small amount of this kind of secondary phase cannot obviously increase the saturate magnetic field.

Lithium-sulfur （Li-S） battery,as a promising next-generation high-energy-density battery,suffers from lithium polysulfides（LPSs） shuttle and Li dendrites *** crystalline framework-based membranes as separator can effectively absorb and restrain LPSs but without precise structural design principal exploration and mechanism ***,bio-inspired 1D oriented lithium-ion transport channels with molecular negatively charged PO3（-1） have been developed in hybrid tetrazole frameworks for promoting the transmission of Li+*** theoretical calculations and in situ spectroscopy demonstrated the higher binding energy and inhibited diffusion of LPSs in *** assembled PP@PO3Janus separator in Li-S coin cell delivered a high initial capacity of 1410.9 mAh g-1at 0.1 C and a low attenuation rate of 0.038%per cycle over 500 cycles at 5 ***,the high capacities of 862,542 and 409 mAh g-1based on high-sulfur-loading cathodes of 2.6,3.8 and 5.0 mg cm-2at0.5 C were achieved,***,the punch battery of PO3@PP separator with S-cathode of 1.2 mg cm-2has been developed for demonstrating its potentials commercial application,which displays impressive capacity of 873.4 mAh g-1with the retention of～78.9%over 50 cycles.

Non-monotonic behavior of jam probability and stretchedexponential distribution in pedestrian counterflow

We adopt a floor field cellular automata model to study the statistical properties of bidirectional pedestrian flow movingin a straight corridor. We introduce a game-theoretic framework to deal with the conflict of multiple pedestrians tryingto move to the same target location. By means of computer simulations, we show that the complementary cumulative distributionof the time interval between two consecutive pedestrians leaving the corridor can be fitted by a stretched exponentialdistribution, and surprisingly, the statistical properties of the two types of pedestrian flows are affected differently by theflow ratio, i.e., the ratio of the pedestrians walking toward different directions. We also find that the jam probability exhibitsa non-monotonic behavior with the flow ratio, where the worst performance arises at an intermediate flow ratio of around0.2. Our simulation results are consistent with some empirical observations, which suggest that the peculiar characteristicsof the pedestrians may attributed to the anticipation mechanism of collision avoidance.

Clinical observation of mineralized collagen bone grafting after curettage of benign bone tumors

Curettage of benign bone tumor is a common cause for bone *** such bone defect repair,autogenous bone,allogeneic bone and traditional artificial bone graft substitutes have many *** recent years,a biomimetic mineralized collagen(MC)with similar composition and microstructures to the natural bone matrix was developed and used for treating various bone *** this work,a retrospective study analyzed clinical outcomes of patients treated with curettage of benign bone tumors and bone grafting with MC,in comparison to another group treated with the same surgical method and autogenous ***-Sandhu X-ray score of the autogenous bone group was superior to the MC group at 1 month after the operation,but the two groups had no statistical difference at 6 and 12 *** MC group was better in Musculoskeletal Tumor Society scoring at 1 and 6 months after the operation,and the two groups had no statistical difference at 12 ***,the MC performed not as good as autogenous bone in early stage of bone healing but achieved comparable outcomes in long-term ***,the MC has advantages in function recovery and avoided potential complications induced by harvesting autogenous bone.

Experimental investigation on using CO_(2)/H_(2)O emulsion with high water cut in enhanced oil recovery

CO_(2) emulsions used for EOR have received a lot of interest because of its good performance on CO_(2)mobility ***,most of them have been focusing on the high quality CO_(2) emulsion(high CO_(2) fraction),while CO_(2) emulsion with high water cut has been rarely *** this paper,we carried out a comprehensive experimental study of using high water cut CO_(2)/H_(2)O emulsion for enhancing oil ***,a nonionic surfactant,alkyl glycosides(APG),was selected to stabilize CO_(2)/H_(2)O emulsion,and the corresponding morphology and stability were evaluated with a transparent PVT ***,plugging capacity and apparent viscosity of CO_(2)/H_(2)O emulsion were measured systematically by a sand pack displacement apparatus connected with a 1.95-m long capillary ***,a high water cut(40 vol%) CO_(2)/H_(2)O emulsion was selected for flooding experiments in a long sand pack and a core sample,and the oil recovery,the rate of oil recovery,and the pressure gradients were *** results indicated that APG had a good performance on emulsifying and stabilizing CO_(2) *** inversion from H_(2)O/CO_(2) emulsion to CO_(2)/H_(2)O emulsion with the increase in water cut was ***_(2)/H_(2)O emulsions with lower water cuts presented higher apparent viscosity,while the optimal plugging capacity of CO_(2)/H_(2)O emulsion occurred at a certain water ***,the displacement using CO_(2)/H_(2)O emulsion provided 18.98% and 13.36% additional oil recovery than that using pure CO_(2) in long sand pack and core tests,*** work may provide guidelines for EOR using CO_(2) emulsions with high water cut.
