Immune enhancement effect of an herb complex extract through the activation of natural killer cells and the regulation of cytokine levels in a cyclophosphamide-induced immunosuppression rat model

Objective: To investigate the effects of a herb complex extract(HCE) prepared from Cornus officinalis Sieb. Et Zucc., Eriobotrya japonica Lindley, and olive leaves on immune response of mouse spleen NK cells in vitro and in vivo analysis. Methods: The activity of natural killer(NK) cells was measured in splenocytes and YAC-1 cells. Mice were immunosuppressed using cyclophosphamide(5 mg/kg body weight). Three different doses of HCE(200, 400, and 800 mg/kg body weight) and red ginseng extract(800 mg/kg body weight) which was used as standard immunomodulatory herb were administered orally for 4 weeks. The body weight, dietary, water intake, organs(liver, thymus, and spleen) weight, completed blood count, and cytokines(tumor necrosis factor alpha, interferon gamma, and interleukin-2) production was measured. Results: At the maximum concentration of HCE, the activity of NK cells was increased by 48.5%. HCE increased liver, spleen, and thymus weights without altering numbers of white blood cells, lymphocytes, and neutrophils in a cyclophosphamide-induced immunosuppression rat model. However, HCE recovered the inhibited cytokine expression; HCE(800 mg/kg) increased cytokines levels. The results indicate the immune enhancement potential of this HCE. Conclusion: The HCE enhances immunity by increasing NK cell activity, regulating cytokine levels, and maintaining spleen weight. Therefore, it may be used as a potential immunity enhancer.

Comparison of two Nd:YAG laser posterior capsulotomy: cruciate pattern vs circular pattern with vitreous strand cutting

AIM： To investigate the effects and safety of neodymium： yttrium-aluminium-garnet（Nd：YAG） laser posterior capsulotomy with vitreous strand cuttingMETHODS： A total of 40 eyes of 37 patients with symptomatic posterior capsular opacity（PCO） were included in this prospective randomized study and were randomly subjected to either cruciate pattern or round pattern Nd：YAG posterior capsulotomy with vitreous strand cutting（modified round pattern）. The best corrected visual acuity（BCVA）, intraocular pressure（IOP）, refractive error, endothelial cell count（ECC）, anterior segment parameters, including anterior chamber depth（ACD） and anterior chamber angle（ACA） were measured before and 1 mo after the laser posterior capsulotomy. RESULTS： In both groups, the BCVA improved significantly（P〈0.001 for the modified round pattern group, P=0.001 for the cruciate pattern group）; the IOP and ECC did not significantly change. The ACD significantly decreased（P〈0.001 for both） and the ACA significantly increased（P=0.001 for the modified round pattern group and P=0.034 for the cruciate group）. The extent of changes in these parameters was not significantly different between the ***： Modified round pattern Nd：YAG laser posterior capsulotomy is an effective and safe method for the treatment of PCO. This method significantly changes the ACD and ACA, but the change in refraction is not significant. Modified round pattern Nd：YAG laser posterior capsulotomy can be considered a good alternative procedure in patients with symptomatic PCO.

Gyejibokryeong-Hwan improves recovery of injured muscles in mouse model of muscle contusion

OBJECTIVE: To investigate the effects of Gyejibokryeong-Hwan(Guizhifuling-wan, GBH) on muscle injury in a mouse model of muscle ***: C57/BL6 mouse biceps femoris muscles were injured using the drop-mass method and injured animals were treated orally with GBH(50,100, or 500 mg/kg) once a day for 7 d. Open field and treadmill running tests were performed to assess functional recovery from muscle injury. The production of pro-inflammatory cytokines was examined by enzyme-linked immunosorbent assay and Western blotting analysis. Expression of the muscle regeneration biomarkers, myoblast determination(Myo D), myogenic factor 5(Myf5), and smooth muscle actin(α-SMA), in the biceps femoris muscle was investigated at the protein and m RNA level by Western blotting and real time-PCR, respectively. Histological analysis was performed using hematoxylin and eosin staining. Finally, myosin heavy chain production was investigated in differentiated C2C12 myoblasts in the presence of ***: GBH treatment markedly improved locomotion and running behavior. GBH significantly inhibited the secretion of monocyte chemoattractant protein-1 into the bloodstream in muscle-contused animals. The levels of Myo D, Myf5, and α-SMA protein and m RNA were significantly up-regulated by GBH in injured muscle tissue. Histological studies suggested that GBH facilitated recovery from muscle damage. However, GBH did not induce the production of myosin heavy chain in ***: Overall, the present study suggested that GBH improves the recovery of the injured muscles in the mouse model of muscle contusion.

Long-distance migratory birds travel more rapidly in spring than in autumn,as they face temporal breeding ***,several species travel slower in spring owing to environmental influences,such as food availability and wind *** trackers were attached to 17 Whooper Swans(Cygnus cygnus) inhabiting northeastern Mongolia,to determine their migration routes and stopover sites in spring and *** between spring and autumn migrations,migration-influencing parameters,and the effect of spring stopover site temperatures were *** swans completed perfect tours between their wintering and breeding sites,and these data were used for *** migration lasted 57 days,with 49.2 days spent at 3.7 stopover *** migration lasted 21.5 days,with 17.5 days spent at 1.0 stopover ***,the swans traveled more rapidly in autumn than in *** distance,number of stopovers,migration speed,and straightness were important migration determinants in both spring and *** distance,stopover duration,number of stopovers,daily travel speed,travel duration,and migration speed differed significantly between spring and *** spring migration,the temperature at the current stopover sites and that at the future stopover sites displayed significant variations(t=1585.8,df=631.6,p <0.001).These findings are critical for the conservation and management of Whooper Swans and their key habitats in East Asian regions,and the data are anticipated to make a particularly significant contribution toward developing detailed management plans for the conservation of their key habitats.

Low survival rate occurs in patients who initially experience a spontaneous return of circulation after cardiac arrest（CA）. In this study, we induced asphyxial CA in adult male Sprague-Daley rats, maintained their body temperature at 37 ± 0.5°C, and then observed the survival rate during the post-resuscitation phase. We examined neuronal damage in the hippocampus using cresyl violet（CV） and Fluore-Jade B（F-J B） staining, and pro-inflammatory response using ionized calcium-binding adapter molecule 1（Iba-1）, glial fibrillary acidic protein（GFAP）, and tumor necrosis factor-alpha（TNF-α） immunohistochemistry in the hippocampus after asphyxial CA in rats under normothermia. Our results show that the survival rate decreased gradually post-CA（about 63% at 6 hours, 37% at 1 day, and 8% at 2 days post-CA）. Rats were sacrificed at these points in time post-CA, and no neuronal damage was found in the hippocampus until 1 day post-CA. However, some neurons in the stratum pyramidale of the CA region in the hippocampus were dead 2 days post-CA. Iba-1 immunoreactive microglia in the CA1 region did not change until 1 day postCA, and they were activated（enlarged cell bodies with short and thicken processes） in all layers 2 days postCA. Meanwhile, GFAP-immunoreactive astrocytes did not change significantly until 2 days post-CA. TNF-α immunoreactivity decreased significantly in neurons of the stratum pyramidale in the CA1 region 6 hours post-CA, decreased gradually until 1 day post-CA, and increased significantly again 2 days post-CA. These findings suggest that low survival rate of normothermic rats in the early period of asphyxia-induced CA is related to increased TNF-α immunoreactivity, but not to neuronal damage in the hippocampal CA1 region.

早期应用参附注射液预防脓毒性心肌病的探索性随机对照研究

目的观察早期应用参附注射液预防脓毒症患者脓毒性心肌病(SIC)的临床疗效。方法2022年6月至2023年1月急诊ICU收治的未发生SIC的脓毒症或脓毒性休克患者,按1∶1随机被分为治疗组和对照组。治疗组予参附注射液50 mL静脉泵入,每12 h 1次,疗程5 d;对照组生理盐水50 mL静脉泵入,每12 h 1次,疗程5 d。主要观察终点为前5 d SIC发生率。次要观察终点为血管活性药物应用时间、前一周液体平衡、ICU住院时间、总通气时间及28 d病死率。结果112例患者进入随机分组,治疗组7例被二次排除,纳入49例进行分析,对照组二次排除6例纳入50例进行分析。治疗组5 d内SIC总发生率为42.9%,低于对照组的64.0%(P=0.035)。其中左室收缩功能障碍治疗组低于对照组(24.5%vs.52.0%,P=0.005),左室舒张功能障碍发生率两组差异无统计学意义,对照组右室功能障碍发生率28.0%,高于治疗组10.2%(P=0.025)。治疗组缩血管药物应用时长75(48,97)h,低于对照组的97(66,128)h(P=0.039);强心药物应用时长治疗组为32(18,49)h,短于对照组的44(25,61)h(P=0.046)。对照组第一周液体平衡为(1260±850)mL,显著高于治疗组的(450±520)mL(P=0.008)。ICU住院时间、总通气时间及28 d病死率两组间均差异无统计学意义(均P>0.05)。结论早期应用参附注射液可以显著降低SIC的发生率,伴有更少的血管活性药物应用和液体正平衡,具有良好的临床应用前景。

目的观察化湿降浊饮对脾虚湿阻型痛风老年患者体质的影响。方法选取2021年1月-2022年1月期间郑州市中医院门诊及住院收治的痛风老年患者80例,按照随机数字表法分为对照组和治疗组,每组各40例。对照组给予非布司他片,治疗组给予化湿降浊饮,两组患者疗程均为4周、8周、12周。观察比较两组患者临床疗效、痛风总发作率、安全性,治疗前后疼痛评分(Visualanaloguescale,VAS)、中医体质变化、中医证候积分、肾功[血尿酸(Blooduricacid,BUA)、血清肌酐(Serum creatinine concentration,SCR)、血尿素氮(Blood urea nitrogen,BUN)]、肝功[天冬氨酸氨基转移酶(Aspartate aminotransferase,AST)、丙氨酸氨基转移酶(Alanine aminotransferase,ALT)]。结果治疗4、8、12周后两组患者VAS评分均较治疗前降低,且治疗12周后治疗组VAS评分明显低于对照组,差异有统计学意义(P湿热质>气虚质为多。治疗后治疗组较治疗前痰湿、湿热质明显减少,平和质增多;对照组较治疗前阳虚及瘀血质、平和质增多,湿热质、气虚质较前有所下降。治疗4周、8周、12周后两组患者中医证候积分均较治疗前降低,差异有统计学意义(P0.05)。治疗期间,两组患者胃肠道不适各2例,对照组尿常规异常1例,谷丙转氨酶异常对照组1例,治疗组2例,尿素氮异常治疗组1例,对照组2例,两组患者出现不良反应较轻,均未做相应处理。结论化湿降浊饮治疗脾虚湿阻型痛风能明显改善中医体质和总体疗效;其降尿酸作用较非布司他片更为平稳,可有效减少痛风发作和非甾体类抗炎药的使用,临床疗效可靠,使用安全。
