Aicardi-Goutieres syndrome(AGS)is a systemic inflammatory disorder caused by mutations in any one of the nine different genes,whose deficiency provokes a type I(interferon)IFN response probably central to pathogenesis.^(1) ADAR1,one of the genes mutated in AGS(AGS6),encodes for an enzyme that belongs to the ADAR family(ADAR1,ADAR2,and ADAR3)that catalyzes the conversion of adenosine to inosine within double-stranded RNAs(dsRNAs)(RNA editing A-to-1).

目的:探讨高强度间歇训练(high-intensity interval training,HIIT)对高脂喂养的载脂蛋白E(ApoE)敲除(Apo^(E-/-))小鼠骨骼肌氧化应激的改善作用以及肌肉因子β-氨基异丁酸(β-aminoisobutyic acid,BAIBA)在其中的可能调节机制。方法:9周龄健康雄性c57BL/6J小鼠和Apo^(E-/-)小鼠分别喂食标准和高脂饲料,其中c57BL/6J小鼠为基线对照组(WT,n=6),Apo^(E-/-)小鼠经高脂饲料喂养12周后随机分为Apo^(E-/-)安静组(Apo^(E-/-),n=10)和Apo^(E-/-)高强度间歇训练组(Apo^(E-/-)HIIT,n=10),Apo^(E-/-)HIIT组进行6周HIIT,WT和Apo^(E-/-)组在笼中自由活动。最后一次运动后48 h取小鼠两侧腿部腓肠肌进行氧化损伤、抗氧化及BAIBA生成相关指标的检测。随后采用L-BAIBA预孵育c2c12细胞,后用过氧化氢(H_(2)O_(2))诱导氧化应激,结合干预条件随机分为control、H_(2)O_(2)和L-BAIBA+H_(2)O_(2)组。相差显微镜观察细胞形态,并检测细胞ROS水平和Nrf2介导的抗氧化通路的相关指标。结果:1)与WT组相比,Apo^(E-/-)组小鼠骨骼肌氧化损伤相关的4-HNE修饰蛋白、蛋白质羰基含量和ROS水平显著升高,而抗氧化相关的p-Nrf2蛋白表达和GSH含量显著降低;而与Apo^(E-/-)组相比,Apo^(E-/-)HIIT组小鼠骨骼肌4-HNE修饰蛋白和蛋白质羰基含量明显降低,p-Nrf2蛋白表达和GSH含量明显升高。2)Apo^(E-/-)HIIT组小鼠骨骼肌生成BAIBA的相关酶基因Acads和Hadha mRNA的表达水平显著高于Apo^(E-/-)组。3)与control组相比,H_(2)O_(2)干预后c2c12细胞明显皱缩,活细胞数量降低,ROS水平显著增加;而与H_(2)O_(2)组相比,L-BAIBA+H_(2)O_(2)组活细胞数量增加,形态较饱满,ROS水平明显降低。4)与control组相比,H_(2)O_(2)显著降低了细胞Nrf2、p-Nrf2及其介导的抗氧化酶SOD1、NQO1和cAT蛋白以及合成GSH的相关酶基因Gss、Gsr和Gclm mRNA的表达水平,并显著降低了细胞GSH含量;而L-BAIBA+H_(2)O_(2)组较H_(2)O_(2)组则显著增加了p-Nrf2、SOD1和NQO1蛋白以及Gss、Gsr、Gclc和Gclm mRNA的表达量,同时细胞GSH含量也显著增加。结论:6周HIIT降低了高脂喂养的Apo^(E-/-)小鼠骨骼肌的氧化损伤,其中肌肉因子BAIBA对Nrf2介导的抗氧化信号的激活可能起着一定的作用。

Environmental toxicants are ubiquitous,and many are known to cause harmful health ***,much of what we know or think we know concerning the targets and long-term effects of exposure to environmental stressors is sadly *** exposure may have health effects that are currently mischaracterized or at least mechanistically incompletely *** much of the recent excitement about stem cells(Scs)focuses on their potential as therapeutic agents,they also offer a valuable resource to give us insight into the mechanisms and risks of toxicant *** only as a response to the increasing ethical pressure to reduce animal testing,Sc studies allow us valuable insight into the true effects of human exposure to environmental stressors under controlled *** present a review of the history of publications on the effects of environmental stressors on Scs,followed by a consolidation of the literature over the past five years on a subset of key environmental stressors of importance to human health and their effects on both embryonic and tissue *** review will make constructive suggestions as to areas of toxicant research where further studies are needed,as well as making indications of the potential utility for advancing knowledge and directing research on environmental toxicology.

BAcKGROUND Prevalence of hepatocellular carcinoma(Hcc)is increasing,especially in patients with metabolic dysfunctionassociated steatotic liver disease(MASLD).AIM To investigate rifaximin(RIF)effects on epigenetic/autophagy markers in *** Adult Sprague-Dawley rats were randomly assigned(n=8,each)and treated from 5-16 wk:control[standard diet,water plus gavage with vehicle(Veh)],Hcc[high-fat choline deficient diet(HFcD),diethylnitrosamine(DEN)in drinking water and Veh gavage],and RIF[HFcD,DEN and RIF(50 mg/kg/d)gavage].Gene expression of epigenetic/autophagy markers and circulating miRNAs were *** All Hcc and RIF animals developed metabolic-dysfunction associated steatohepatitis fibrosis,and cirrhosis,but three RIF-group did not develop *** animals who developed Hcc with those who did not,miR-122,miR-34a,tubulin alpha-1c(Tuba-1c),metalloproteinases-2(Mmp2),and metalloproteinases-9(Mmp9)were significantly higher in the *** opposite occurred with Becn1,coactivator associated arginine methyltransferase-1(carm1),enhancer of zeste homolog-2(Ezh2),autophagy-related factor Lc3A/B(Map1 Lc3b),and p62/sequestosome-1(p62/SQSTM1)-*** with controls,Map1 Lc3b,Becn1 and Ezh2 were lower in Hcc and RIF-groups(Pcarm1 was lower in Hcc compared to RIF(Pc expression of Mmp9 was higher in Hcc in relation to the control;the opposite was observed for p62/Sqstm1(Pcompared to the control(P=0.024).There was no difference among groups for Tuba-1c,Aldolase-B,alpha-fetoprotein,and Mmp2(P>0.05).miR-122 was higher in Hcc,and miR-34a in RIF compared to controls(Pcc compared to RIF,and the inverse was observed for miR-224(Pce among groups regarding miR-33a,miR-143,miR-155,miR-375 and miR-21(P>0.05).cONcLUSION RIF might have a possible beneficial effect on preventing/delaying liver carcinogenesis throu

Background: Harvesting of forest products is a widespread driver of disturbance in developing nations, where policies are increasingly aimed at managing natural forests for sustainable use. There is thus need for research aimed at understanding the impact of resource use on forest habitats and concomitant effects on *** forests in the Eastern cape, South Africa are harvested informally for poles and medicinal bark and occur along elevational gradients of 800–1600 m above sea level. Patterns of spatial diversity and human disturbance are expected to be affected by elevation. Furthermore, species’ responses to disturbance are expected to vary depending on their level of habitat specialisation. Understanding harvest impacts on forest biodiversity thus requires disentangling the separate effects of elevation and disturbance, and considering forest-specialist and forest-generalist species separately. This study comprises two components. First, harvest activities, resultant harvestmediated habitat heterogeneity, and avifaunal species richness, composition and beta-diversity were compared across two elevational zones in a harvested forest. Second, the role of harvest-mediated habitat heterogeneity in driving patterns of avifaunal diversity were assessed, while controlling for elevation, and considering forest-specialist and forest-generalist species ***: Harvest rates were higher, and activities more varied in the lower elevation zone, with significant impacts of harvesting on habitat features resulting in higher harvest-mediated habitat heterogeneity at lower ***-mediated increases in habitat heterogeneity positively affected forest-generalist species richness, while forest-specialist richness was negatively affected. While species composition of both groups differed across elevational zones, variation in harvest-mediated habitat heterogeneity did not fully account for this, suggesting that factors other than disturbance
