Effect of Different Cultivation Systems on the Accumulation of Nutrients and Phytochemicals in Ligularia fischeri

A hydroponic cultivation system was established to improve the nutraceutical properties of Ligularia fischeri(Ledeb.) Turcz, during which nutrient uptake by the plant from nutrient solution was measured using inductive coupled plasma-atomic emission spectroscopy(ICP-AES).Based on the obtained data, the uptake of macro and micro elements per gram of fresh weight was calculated. The uptake of macro-elements of NH_4^+-N,NO_3^--N, K, S, P, Ca,and Mg were 1,62,4.27, 8.41,1.19, 2.59, 2,79, and 0.84 mg·g^(-1) FW and micro-elements of B, Fe, Mn, Mo, Cu, and Zn were 9.91,22.31,25.73,2.51,2.91, and 5.07 μg·g^(-1) FW. Moreover, the effects of cultivation systems on growth and phytochemical composition of *** were compared. The greatest biomass was observed in the hydroponic cultivation system with continuous circulation nutrient solution compared to natural soil and Tosilee media based culture systems. The recirculated hydroponic system significantly increased the total phenol contents of the leaf, petiole, and root extracts by 17.6%, 30.60%, and 20.9% more compared to the soil grown. The recirculated hydroponic system treatment significantly increased the total antioxidant capacity of root extracts by 55.9% more compared to the soil treatment. Based on the contents of elements, total phenolic and flavonoid, it was concluded that hydroponic cultivation system is the optimal method to enhance medicinal value.

Physiological and Biochemical Modulations upon Root Induction in Rose Cuttings as Affected by Growing Medium

Two cut rose(Rosa hybrida) cultivars ‘Sherbet'(easy-to-root) and ‘Alibaba'(difficult-to-root) were propagated by single nodal cuttings on preformed media. Four commercial media such as Tosilee, Ellepot, rockwool and phenolic foam RC, and four newly-developed foam media namely Oasis PU 14-S1, Oasis TP-S1, Oasis PU-7 B and Oasis PU 15-S1 were employed in this study. All media were manufactured to fit into50-cell plug trays. The experiment was laid out in a complete randomized block design with three replications on a propagation bench with a plastic tunnel installed in a glasshouse, with(98 ± 2)% relative humidity and over 50% shading of the natural sunlight. After four weeks of experiment, growth parameters, localization of superoxide radicals, and activities of rooting biochemical markers were measured. The results suggested that the newly-developed foam media Oasis TP-S1 and Oasis PU-7 B enhanced the growth, density of leaf stomata, and consisted of lesser harmful superoxide radicals in similar manner with the existing commercial media. Moreover, the Oasis TP-S1 and Oasis PU-7 B media significantly enhanced the total phenol content and the activities of important enzymes involved in the phenol metabolism which plays a vital role in the formation of roots. Thus, the understanding of both physiological and biochemical changes implicated by the growing medium during the rooting of cuttings would aid in the selection of optimal growing medium for the propagation of rose cuttings.

Reefs in the Early Paleozoic Taebaek Group,Korea:A Review

Various early Paleozoic （Cambrian Series 3-Middle Ordovician） reefs are found in the Taebaek Group, eastern Korea, located in the eastern margin of the Sino-Korean Block. They occur in every carbonate-dominant lithostratigraphic unit of the group, but their morphology and composition differ markedly. The Daegi Formation （middle Cambrian： Cambrian Series 3） contains siliceous sponge-Epiphyton reefs formed in a shallow subtidal environment, which is one of the earliest metazoan-bearing microbial reefs after the archaeocyath extinction. The Hwajeol Formation （upper Cambrian： Furongian） encloses sporadic dendrolites consisting of Angulocellularia, which developed in a relatively deep subtidal environment, representing a rare deeper water example. The onset of the Ordovician radiation resulted in the formation of microbialite-Archaeoscyphia-calathiid patch reefs in shallow subtidal deposits of the Lower Ordovician Dumugol Formation. Subsequent late Early Ordovician relative sea-level fall established extensive peritidal environments, forming microbial mats and stromatolites of the Lower-Middle Ordovician Makgol Formation. Ensuing Ordovician radiation resulted in one of the earliest metazoan skeletal reefs of the Middle Ordovician Duwibong Formation, constructed by stromatoporoid Cystostroma and bryozoan Nicholsonella, and developed around shallow shoals. These reefs reflect ongoing evolution and sea-level change during the early Paleozoic, and exemplify a rare glimpse of peri-Gondwanan records of reef evolution, which warrant detailed investigations and comparison with their counterparts in other regions.

Totally laparoscopic total gastrectomy using the modified overlap method and conventional open total gastrectomy:A comparative study

AIM: To investigate whether, or how, DA-9601, which is a new gastroprotective agent, inhibits TNF-α-induced inflammatory signals in gastric epithelial AGS cells. METHODS: Cell viability was determined by MTT assay. IL-8 and CCL20 promoter activities were determined by a luciferease reporter gene assay. NF-κB-dependent transcriptional activity was determined by I-κBαdegradation, NF-κB p65 nuclear translocation and a luciferase activity assay. IL-8 and CCL20 gene expression and protein secretion were determined by RT-PCR and an enzymelinked immunosorbent assay (ELISA). Total and phos-phorylated forms of mitogen-activated protein kinases (MAPKs) were determined by Western blot. RESULTS: Treatment of AGS cells with DA-9601 reduced TNF-α-induced IL-8 and CCL20 promoter activities, as well as their gene expression and protein release. TNF-αalso induced NF-κB-dependent transcriptional activity in AGS cells. In contrast, in cells treated with DA-9601, TNF-α-induced NF-κB activity was significantly blocked. Although all three MAP kinase family members were phosphorylated in response to TNF-α, a selective inhibitor of p38 kinase SB203580 only could inhibit both NF-κB-dependent transcriptional activity and IL-8 and CCL20 production, suggesting a potential link between p38 kinase and NF-κB-dependent pathways in AGS cells. Interestingly, DA-9601 also selectively inhibited p38 kinase phosphorylation induced by TNF-α. CONCLUSION: DA-9601 blocked TNF-α-mediated inflammatory signals by potentially modulating the p38 kinase pathway and/or a signal leading to NF-κBdependent pathways in gastric epithelial cells.

Effects of ranitidine for exercise induced gastric mucosal changes and bleeding

AIM： To evaluate the effect of ranitidine on gastric mucosal changes and on GI bleeding in long distance runhers. METHODS： Twenty-four long distance runners （M： 16, F： 8, age： 18.2＋ 1.5 years） participated in this study. A symptom questionnaire, stool hemoccult test, and upper gastrointestinal （GI） endoscopy were performed on the subjects prior to the study. The subjects took oral ranitidine （150 mg, b.i.d.） for two weeks. The upper GI endoscopy and stool Hemoccult tests were repeated after the treatment. RESULTS： Twenty-two of the 24 runners had at least one upper G1 mucosal lesion before the medication. The Endoscopic improvements were seen in eleven of the 14 cases of erosive gastritis and four of the 5 cases of esophagitis. Six subjects were Heine occult positive prior to the study, but only one was positive after the medication. CONCLUSION： Gastric mucosal lesions and GI bleeding in long distance runners seem to be associated to acidrelated factors mediated by the high level of regular running. Ranitidine seems to be and effective prophylaxis to prevent gastric mucosal lesions and GI bleeding.

AIM: To study the mechanism of Je-Chun-Jun (JCJ)-inducing the apoptosis of the human cervical carcinoma, HeLa cells. METHODS: The cell viability was assessed using MTT assay. The optical density was measured at 570 nm. The caspase activity was measured using 50 mmol/L of fluorogenic substrate, AC-DEVD-AMC (caspase-3), AC-VEID-AMC (caspase-8) or AC-LEHD-AFC (caspase-9). To confirm the expression of proteins, Western blotting was performed. To detect the characteristic of apoptosis chromatin condensation, HeLa cells were stained with Hoechst 33258 in the presence of JCJ. For the cell cycle analysis, HeLa cells were incubated with Propidium iodide (PI) solution. Fluorescence intensity of cell cycle was measured using flow cytometry system. RESULTS: The loss of viability occurred following the exposure of 10 g/L JCJ. Cells treated with 10 g/L JCJ for 3 d exhibited the apoptotic morphology (brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining) and the reduction of cell volume. Cells incubated
