Unlike mammals with adaptive immunity,plants rely on their innate immunity based on pattern-triggered immunity(PTI)and effector-triggered immunity(ETI)for pathogen *** oxygen species,known to play crucial roles in PTI and ETI,can perturb cellular redox homeostasis and lead to changes of redox-sensitive proteins through modification of cysteine sulfhydryl *** redox regulation of protein functions has emerged as an important mechanism in several biological processes,little is known about redox proteins and how they function in PTI and *** this study,cysTMT proteomics technology was used to identify similarities and differences of protein redox modifications in tomato resistant(PtoR)and susceptible(prf3)genotypes in response to Pseudomonas syringae pv tomato(Pst)*** addition,the results of the redox changes were compared and corrected with the protein level changes.A total of 90 potential redox-regulated proteins were identified with functions in carbohydrate and energy metabolism,biosynthesis of cysteine,sucrose and brassinosteroid,cell wall biogenesis,polysaccharide/starch biosynthesis,cuticle development,lipid metabolism,proteolysis,tricarboxylic acid cycle,protein targeting to vacuole,and oxidation–*** inventory of previously unknown protein redox switches in tomato pathogen defense lays a foundation for future research toward understanding the biological significance of protein redox modifications in plant defense responses.

Regulation of D-galacturonate metabolism in <i>Caulobacter crescentus</i>by HumR, a LacI-family transcriptional repressor

The oligotrophic freshwater bacterium Caulobacter crescentus encodes a cluster of genes (CC_1487 to CC_1495) shown here to be necessary for metabolism of D-galacturonate, the primary constituent of pectin, a major plant polymer. Sequence analysis suggests that these genes encode a version of the bacterial hexuronate isomerase pathway. A conserved 14 bp sequence motif is associated with promoter regions of three operons within this cluster, and is conserved in homologous gene clusters in related alpha-Proteobacteria. Embedded in the hexuronate gene cluster is a gene (CC_1489) encoding a member of the LacI family of bacterial transcription factors. This gene product, designated here as HumR (hexuronate metabolism regulator), represses expression of the uxaA and uxaC operon promoters by binding to the conserved operator sequence. Repression is relieved in the presence of galacturonate or, to a lesser extent, by glucuronate. Other genes potentially involved in pectin degradation and hexuronate transport are also under the control of HumR. Adoption of a LacI-type repressor to control hexuronate metabolism parallels the regulation of xylose, glucose, and maltose utilization in C. crescentus, but is distinct from the use of GntR-type repressors to control pectin and hexuronate utilization in gamma-Proteobacteria such as Escherichia coli.

Objective To determine the prevalence and associations of general practice registrars’performing absolute cardio-vascular risk(ACVR)assessment(ACVRa).Design A cross-sectional study employing data(2017–2018)from the Registrar Clinical Encounters in Training project,an ongoing inception cohort study of Australian GP *** outcome measure was whether an ACVRa was *** employed univariable and multivariable *** was conducted for all patient problems/diagnoses,then for an‘at-risk’population(specific problems/diagnoses for which ACVRa is indicated).Setting Three GP regional training organisations(RTOs)across three Australian *** GP registrars training within participating *** 1003 registrars(response rate 96.8%)recorded details of 69105 problems either with Aboriginal and/or Torres Strait patients aged 35 years and older or with non-Indigenous patients aged 45 years and *** these problems/diagnoses,1721(2.5%(95%CI 2.4%to 2.6%))involved an *** ACVRa was‘plausibly indicated’in 10384 problems/*** these,1228(11.8%(95%CI 11.2%to 12.4%))involved ***‘all problems/diagnoses’,on multivariable analysis female gender was associated with reduced odds of ACVRa(OR 0.61(95%CI 0.54 to 0.68)).There was some evidence for Aboriginal and/or Torres Strait Islander people being more likely to receive ACVRa(OR 1.40(95%CI 0.94 to 2.08),p=0.10).There were associations with variables related to continuity of care,with reduced odds of ACVRa:if the patient was new to the registrar(OR 0.65(95%CI 0.57 to 0.75)),new to the practice(OR 0.24(95%CI 0.15 to 0.38))or the problem was new(OR 0.68(95%CI 0.59 to 0.78));and increased odds if personal follow-up was organised(OR 1.43(95%CI 1.24 to 1.66)).For‘ACVRa indicated’problems/diagnoses,findings were similar to those for‘all problems/diagnoses’.Association with Aboriginal and/or Torres Strait Islander status,however,was significant at p<0.05(OR 1.60(95%CI 1.04 to 2.46))a

AIM To increase evidence-based pain prevention strategy use during routine vaccinations in a pediatric primary care clinic using quality improvement *** Specific intervention strategies(i.e.,comfort positioning,nonnutritive sucking and sucrose analgesia,distraction) were identified,selected and introduced in three waves,using a Plan-Do-Study-Act ***-wide change was measured from baseline to post-intervention by:(1) percent of vaccination visits during which an evidence-based pain prevention strategy was reported as being used; and(2) caregiver satisfaction ratings following the ***,self-reported staff and caregiver attitudes and beliefs about pain prevention were measured at baseline and 1-year post-intervention to assess for possible long-term cultural *** Significant improvements were noted *** of at least one pain prevention strategy was documented at 99% of patient visits and 94% of caregivers were satisfied or very satisfied with the pain prevention care ***/caregivers reported greater satisfaction with the specific pain prevention strategy used [t(143) = 2.50,P ≤ 0.05],as well as greater agreement that the pain prevention strategies used helped their children's pain [t(180) = 2.17,P ≤ 0.05] and that they would be willing to use the same strategy again in the future [t(179) = 3.26,P ≤ 0.001] as compared to *** and caregivers also demonstrated a shift in attitudes from baseline to 1-year ***,staff reported greater agreement that the pain felt from vaccinations can result in harmful effects [2.47 vs 3.10; t(70) =-2.11,P ≤ 0.05],less agreement that pain from vaccinations is "just part of the process" [3.94 vs 3.23; t(70) = 2.61,P ≤ 0.05],and less agreement that parents expect their children to experience pain during vaccinations [4.81 vs 4.38; t(69) = 2.24,P ≤ 0.05].Parents/caregivers reported more favorable attitudes about pain prevention st

[背景]越来越多的证据显示,母亲空气污染暴露与对胎儿生长的不良影响之间存在关联;但是,现有的文献并不一致。[目的]研究9个国家14个中心的数据,量化母亲颗粒物空气污染暴露与足月出生体重和低出生体重(LBW)之间的关联,并探讨此关联中地点特征和暴露评估方法对中心之间异质性的影响。[方法]空气污染与妊娠结局国际合作(ICAPPO)的各中心采用共同的分析方案,对PM10和PM2.5(颗粒物≤10和2.5μm)与足月LBW和连续出生体重进行效应评估。采用meta分析将各个中心(~300万例出生)的效应评估结合起来,并采用meta回归分析在报告的效应评估中,中心特征和暴露评估方法对中心之间异质性的影响。[结果]随机效应meta分析显示,调整母亲的社会经济地位后,在整个妊娠期间,足月LBW与PM10[比值比(OR)=1.03;95%CI:1.01~1.05]和PM2.5[OR=1.10;95%CI:1.03~1.18]每增加10μg/m3呈正相关。在充分调整随机效应的meta分析中,作为一个连续性的结果,PM10暴露每增加10μg/m3与足月出生体重呈负相关(-8.9g;95%CI:-13.2^-4.6 g)。meta回归分析显示,具有较高平均PM2.5水平和PM2.5∶PM10比率的中心,以及采用时间暴露评估(与时空相比)的中心,所报告的关联更强。[结论]在研究人群中,母亲颗粒物污染暴露与足月LBW相关联。有3个地点特征和暴露评估方法学方面的不同导致各个中心报告的关联的差异。

The Great Plains region of the United States is susceptible to drought of all kinds including meteorological/climatological, agricultural, hydrological, and socioeconomic. Drought conditions in the region span varying spatial and temporal scales and the causes include: 1) certain synoptic conditions that favor drought such as mid-tropospheric ridging over the drought-affected area and a weak low-level jet;2) sea surface temperature anomalies and associated teleconnections;3) land-atmosphere coupling;and 4) anthropogenic effects. While drought can span as few as a couple of months, the most severe droughts can occur at the decadal scale such as the 1930s Dust Bowl, the worst drought in recent history from a societal standpoint. Such droughts in the Great Plains have widespread impacts on agriculture, water resources, human health, and the economy.
