Background:Hepatitis C virus(HCV)persistence is associated with chronic necroinflammatory liver disease with dysfunctional antiviral T cells,induction of immune inhibitory pathways and the emergence of T cell escape *** Aims:Our aim was to develop a convenient co-culture system of HCV-specific T cells and HCV-replicating hepatocytesto examine potential host-virus interactions(***,PD-1)relevant in HCV persistence and disease ***:HCV-permissive Huh7.5 cells were lentivirally transduced to express HLA-A2,electroporated or infected with full length HCV genotype 1a(H77s)and 2a(JFH1-derived)clones and confirmed for HLA-A2 and HCV expressions by flow cytometry using anti-HLA-A2 and anti-HCV NS5A ***-term T cell lines specific for HLA-A2-restricted HCV genotype 1a-derived NS3 and NS5B CD8 T cell epitopes were expanded from 2 HLA-A2+spontaneous HCV ***1-derived HCV clone was modified to express genotype 1a-derived NS3 1073 orNS5B 2594 *** vectors encoding T cell receptors(TCR)specific for NS3 1073 and NS5B 2594 were developed from TCRs isolated from short-term T cell ***-specificity was confirmed by pMHC tetramer ***-specific effector function was examined by intracellular cytokine staining upon co-culture with HCV-replicating or peptide-pulsed HLA-A2+Huh7.5 *** and conclusions:HCV-specific CD8 T cells recognized cognate epitopes presented by HLA-A2Huh7.5 cells by exogenous peptide pulse and endogenous processing via full-length HCV *** TCR-transduced CD8 T cells showed dose-dependent antigen-specific effector function that resembled HCV-specific CD8 T cells directly expanded from patients and suppressed HCV replication when co-cultured with HCV-replicating HLA-A2Huh7.5 cell.

Further Studies of the Cap Pushing Response in Honey Bees (Apis mellifera)

The Cap Pushing Response (CPR) is a free-flying technique used to study learning and memory in honey bees (Apis mellifera). The series of experiments outlined in this paper aimed to test whether honey bees exhibit the cognitive concept of “expectancy” utilizing the CPR in a weight differentiation paradigm. Five previous experiments in our laboratory have explored whether the concept of expectancy can account for honey bee performance and have all failed to support the cognitive interpretation. The first experiment examined if bees could differentiate between the two caps in the amount of force they used to push the cap and the distance the cap was pushed when the caps were presented one at a time. The second experiment explored cap weight preference by presenting bees with a choice between the two caps. The third and fourth experiments tested the bee’s ability to expect reward or punishment based on cap weight. Results revealed that bees were found to have a strong preference for the light cap and therefore were not able to expect reward or punishment based on cap weight. These experiments contribute to the debate on whether bees have “cognitive” representations and continue to support the behaviorist interpretation.

Fat injection to correct contour deformities of the reconstructed breast:a single surgeon experience

Aim:Autologous fat grafting has gained acceptance as a technique to improve aesthetic outcomes in breast *** purpose of this study was to share our clinical experience using autologous fat injection to correct contour deformities during breast ***:A single surgeon,prospectively maintained database of patients who underwent autologous fat injection during breast reconstruction from January 2008 to November 2013 at McGill University Health Center was *** characteristics,breast history,type of breast reconstruction,volume of fat injected,and complications were ***:One hundred and twenty-four patients benefited from autologous fat injection from January 2008 to November 2013,for a total of 187 treated *** patients were on average 49.3 years old(±8.9 years).Fat was harvested from the medial thighs(20.5%),flanks(39.1%),medial thighs and flanks(2.9%),trochanters(13.3%),medial knees(2.7%),and abdomen(21.9%).An average of 49.25 mL of fat was injected into each reconstructed breast.A total of 187 breasts in 124 patients were lipo-infiltrated during the second stage of breast *** breasts(in 12 separate patients)were injected several years after having undergone lumpectomy and *** the 187 treated breasts,118 were reconstructed with expanders to implants,45 with deep inferior epigastric perforator flaps,9 with latissimus dorsi flaps with implants,4 with transverse rectus abdominis myocutaneous flaps,and 13 had previously undergone lumpectomy and *** complications were noted in the entire series,for a rate of 3.2%.All were in previously radiated *** follow-up time was 12 months(range:2-36 months).Conclusion:Fat injection continues to grow in popularity as an adjunct to breast *** experience demonstrates a low complication rate as compared to most surgical interventions of the breast and further supports its safety in breast ***
