Mismatched effects of receptor interacting protein kinase-3 on hepatic steatosis and inflammation in nonalcoholic fatty liver disease

AIM To validate the effects of receptor interacting protein kinase-3(RIP3) deletion in non-alcoholic fatty liver disease(NAFLD) and to clarify the mechanism of *** Wild-type(WT) and RIP3 knockout(KO) mice werefed normal chow and high fat(HF) diets for 12 wk. The body weight was assessed once weekly. After 12 wk, the liver and serum samples were extracted. The liver tissue expression levels of RIP3, microsomal triglyceride transfer protein, protein disulfide isomerase, apolipoprotein-B, X-box binding protein-1, sterol regulatory element-binding protein-1c, fatty acid synthase, cluster of differentiation-36, diglyceride acyltransferase, peroxisome proliferator-activated receptor alpha, tumor necrosis factor-alpha(TNF-α), and interleukin-6 were assessed. Oleic acid treated primary hepatocytes from WT and RIP3 KO mice were stained with Nile red. The expression of inflammatory cytokines, including chemokine(C-X-C motif) ligand(CXCL) 1, CXCL2, and TNF-α, in monocytes was *** RIP3 KO HF diet fed mice showed a significant gain in body weight, and liver weight, liver to body weight ratio, and liver triglycerides were increased in HF diet fed RIP3 KO mice compared to HF diet fed WT mice. RIP3 KO primary hepatocytes also had increased intracellular fat droplets compared to WT primary hepatocytes after oleic acid treatment. RIP3 overexpression decreased hepatic fat content. Quantitative real-time polymerase chain reaction analysis showed that the expression of very-low-density lipoproteins secretion markers(microsomal triglyceride transfer protein, protein disulfide isomerase, and apolipoprotein-B) was significantly suppressed in RIP3 KO mice. The overall NAFLD Activity Score was the same between WT and RIP3 KO mice; however, RIP3 KO mice had increased fatty change and decreased lobular inflammation compared to WT mice. Urban rail transit (URT) disruptions present considerable challenges due to several factors: i) a high probability of occurrence, arising from facility failures, disasters, and vandalism;ii) substantial negative effects, notably the delay of numerous passengers;iii) an escalating frequency, attributable to the gradual aging of facilities;and iv) severe penalties, including substantial fines for abnormal operation. This article systematically reviews URT disruption management literature from the past decade, categorizing it into pre-disruption and post-disruption measures. The pre-disruption research focuses on reducing the effects of disruptions through network analysis, passenger behavior analysis, resource allocation for protection and backup, and enhancing system resilience. Conversely, post-disruption research concentrates on restoring normal operations through train rescheduling and bus bridging services. The review reveals that while post-disruption strategies are thoroughly explored, pre-disruption research is predominantly analytical, with a scarcity of practical pre-emptive solutions. Moreover, future research should focus more on increasing the interchangeability of transport modes, reinforcing redundancy relationships between URT lines, and innovating post-disruption strategies.

Plants exhibit an astonishing ability to regulate organ regeneration upon *** of leaf explants promotes the biosynthesis of indole-3-acetic acid(IAA),which is polar-transported to excised regions,where cell fate transition leads to root founder cell specification to induce de novo root *** regeneration capacity of plants has been utilized to develop in vitro tissue culture ***,we report that IAA accumulation near the wounded site of leaf explants is essential for callus formation on 2,4-dichlorophenoxyacetic acid(2,4-D)-rich callus-inducing medium(CIM).Notably,a high concentration of 2,4-D does not compensate for the action of IAA because of its limited efflux;rather,it lowers IAA biosynthesis via a negative feedback mechanism at an early stage of in vitro tissue culture,delaying callus *** auxin negative feedback loop in CIM-cultured leaf explants is mediated by an auxin-inducible APETALA2 transcription factor,ENHANCER OF SHOOT REGENERATION 2(ESR2),along with its interacting partner HISTONE DEACETYLASE 6(HDA6).The ESR2–HDA6 complex binds directly to,and removes the H3ac mark from,the YUCCA1(YUC1),YUC7,and YUC9 loci,consequently repressing auxin biosynthesis and inhibiting cell fate transition on 2,4-D-rich *** findings indicate that negative feedback regulation of auxin biosynthesis by ESR2 and HDA6 interferes with proper cell fate transition and callus initiation.

The origin of agriculture in the farming-pastoral zone of northern China remains in *** central region of the Inner Mongolia Plateau is located in the core area of the farming-pastoral zone;thus,it is a critical region for exploring the origin of the dryland farming system in northern *** study selected the Yumin Site and Banan Site,which belong to the Yumin Culture-the beginning of Neolithic culture in Inner Mongolia-as the research *** on the quartz optically stimulated luminescence(OSL)dating on the sedimentary sections from the Yumin site(YM)and Banan site(BN1 and BN2),the Holocene chronology framework of each section was *** that,by identifying carbonized grains in the Yumin site and the multi-proxy analysis of each section,we investigated the relationship between the origin of agriculture and climate change in this *** results revealed that the timing of the origin of agriculture recorded in the Yumin site lagged behind the timing of a significant increase of precipitation during the early Holocene but coincided with the timing of a significant increase of vegetation around 8.4 *** phenomenon was further confirmed by the published high-resolution paleoenvironmental records from the surrounding area of the Yumin *** propose that with the gradual amelioration of hydrothermal conditions since the beginning of the Holocene,the regional ecosystem had been improved,resulting in the gradual conversion of the land surface from infertile sand to organic-rich soil,providing an appropriate environmental foundation for the origin of dryland farming in northern China around 8.4 *** study highlighted that the“accumulative environmental effects”during the early Holocene played a vital role in the origin of agriculture in northern China and provided a reference for agricultural management in the context of future climate change.

Environmental toxicity assessment of chromium (III) oxide nanoparticles using a phytotoxic, cytotoxic, and genotoxic approach

Chromium(III)oxide nanoparticles(NPs)have a wide range of industrial applications,particularly in the production of lithium-ion batteries;however,the toxicity of these NPs has not been studied *** phytotoxicity and genotoxicity of chromium(III)oxide NPs have not been assessed,and only one study reported their cytotoxicity,underscoring the need to perform comprehensive toxicity studies on these ***,we showed that chromium(III)oxide NPs had no effect on germination rates,and only decreased root elongation in *** as determined by root elongation and biomass ***(III)oxide NPs had no phytotoxic effects under environmental realistic conditions,whereas they had cytotoxic effects against HK-2 and Raw 264.7 cells in ***,the cytotoxicity of chromium(III)oxide NPs did not occur under environmentally realistic *** genotoxicity of chromium(III)oxide NPs was not significant in HK-2 cells,suggesting that the potential environmental cytotoxicity of chromium(III)oxide NPs is *** study proposes comprehensive phytotoxicity,cytotoxicity,and genotoxicity testing tools that would be easy,safe,and effective for evaluating the toxicity of *** integrated testing system could be an accurate and stable method for evaluating the toxicity of other nanomaterials.
