Tumor-protein 53(p53)is a transcription factor(TF)encoded by TP53(Trp53 in mice)and is a master regulator of tumor-suppressive programs'.Importantly,TP53 is the most frequently mutated gene in human cancer,and its loss of function contributes to tumor development,exemplified by variable p53 mutations detected in 50%-75%of pancreatic cancers.2 From these perceptions,a number of targets transcribed by p53 that confer tumor suppression have been ***,our understanding of how p53 prevents normal cells from initiating tumorigenesis is still *** complement this limitation,herein,we established a new approach to identify underlying effectors transcribed by p53 in normal physiology.

Convenience rice has become widely popular due to its easy availability for cooking. This study investigated the starch structure and composition of leachate and the microstructure of reheated convenience rice using novel processing technologies: super-heated steaming(SHS), auto-electric cooking(AEC), and pressurized-steam cooking(PSC). Additionally, the effect of two different target water contents(58% and 63%) was also evaluated. The PSC_63% sample had the highest total solids and amylopectin amount in the leachate. The amylopectin amount in the leachate differed significantly based on the targeted water content. Morphological characterization revealed that the swelling of starch and the coated layer on the surface of rice grains were most pronounced in the PSC_63% sample due to the pressure processing. The textural hardness of the AEC_58% sample was much higher than that of the other samples. The PSC_63% sample had the highest textural adhesiveness value, which can be attributed to the highest amylopectin amount in the leachate. Sensory characterization showed that the PSC_63% sample had the highest glossiness, whiteness, moistness, and overall acceptability. The principal component analysis score plots presented substantial differences in the leachate and textural and sensory characteristics of reheated convenience rice among the different processing technologies.

Biliary tract cancers(BTCs)refer to invasive adenocarcinomas arising from the bile duct(intrahepatic cholangiocarcinoma and extrahepatic cholangiocarcinoma)and gallbladder(gallbladder cancer).Patients with BTCs typically present at an advanced stage due to a lack of early detection methods and non-specific symptoms;systemic chemotherapy is the cornerstone of treatment for these advanced *** plus cisplatin(GC)has been the standard treatment since the pivotal ABC-02 randomized phase 3 trial conducted in 2010(1).Moving beyond the era of GC,several triplet combination regimens have been proposed in recent years(Table 1).The combination of nab-paclitaxel(nab-P)and GC showed promising survival outcomes in a phase 2 trial(2),although it eventually failed to meet the primary endpoint of overall survival(OS)in a randomized phase 3 trial(3).Around the same time,TOPAZ-1(5)demonstrated that the programmed death ligand 1(PD-L1)inhibitor durvalumab combined with GC significantly improved the median OS[11.5 vs.12.8 months;hazard ratio(HR),0.80;95%confidence interval(CI):0.66–0.97;P=0.021]and median progression-free survival(PFS)(5.7 vs.7.2 months;HR,0.75;95%CI:0.63–0.89;P=0.001),with an objective response rate(ORR)of 26.7%.Moreover,KEYNOTE-966(6)found that the addition of programmed death-1(PD-1)inhibitor pembrolizumab also improved median OS(10.9 vs.12.7 months;HR,0.83;95%CI:0.72–0.95;P=0.0034),whereas the median PFS only showed numerical improvements(5.6 vs.6.5 months;HR,0.86;95%CI:0.75–1.00;P=0.023),with an ORR of 29%.Consequently,the combination of PD-1/PD-L1 inhibitors and GC has become the new standard of care for first-line treatment of patients with BTCs,establishing the new era of triplet combinations.

Most microfluidic separation techniques rely largely on object size as a separation *** ability to separate micro-objects based on their shape is crucial in various biomedical and chemical ***,we develop an ondemand,label-free acoustofluidic method to separate prolate ellipsoids from spherical microparticles based on traveling surface acoustic wave-induced acoustic radiation force and *** freely rotating non-spherical microobjects were aligned under the progressive acoustic field by the counterrotating radiation torque,and the major axis of the prolate ellipsoids was parallel to the progressive wave *** specific alignment of the ellipsoidal particles resulted in a reduction in the cross-sectional area perpendicular to the wave *** a consequence,the acoustic backscattering decreased,resulting in a decreased magnitude of the radiation *** the variation in radiation force,which depended on the micro-object morphology enabled the acoustofluidic shape-based *** conducted numerical simulations for the wave scattering of spherical and prolate objects to elucidate the working mechanism underlying the proposed method.A series of experiments with polystyrene microspheres,prolate ellipsoids,and peanut-shaped microparticles were performed for *** quantitative analysis of the separation efficiency,we confirmed the high purity and high recovery rate of the proposed acoustofluidic shapebased separation of *** a bioparticle,we utilize Thalassiosira eccentrica to perform shape-based separation,as the species has a variety of potential applications in drug delivery,biosensing,nanofabrication,bioencapsulation and immunoisolation.

The Weather Research and Forecasting model coupled with Chemistry(WRF-Chem),a type of online coupled chemistry-meteorology model(CCMM),considers the interaction between air quality and meteorology to improve air quality *** data assimilation(DA)can be used to reduce uncertainty in meteorological field,which is one factor causing prediction uncertainty in the *** this study,WRF-Chem and three-dimensional variational DA were used to examine the impact of meteorological DA on air quality and meteorological forecasts over the Korean *** nesting model domains were configured over East Asia(outer domain)and the Korean Peninsula(inner domain).Three experiments were conducted by using different DA domains to determine the optimal model domain for the meteorological *** the meteorological DA was performed in the outer domain or both the outer and inner domains,the root-mean-square error(RMSE),bias of the predicted particulate matter(PM)concentrations,and the RMSE of predicted meteorological variables against the observations were smaller than those in the experiment where the meteorological DA was performed only in the inner *** indicates that the improvement of the synoptic meteorological fields by DA in the outer domain enhanced the meteorological initial and boundary conditions for the inner domain,subsequently improving air quality and meteorological *** to the experiment without meteorological DA,the RMSE and bias of the meteorological and PM variables were smaller in the experiments with *** effect of meteorological DA on the improvement of PM predictions lasted for approximately 58-66 h,depending on the ***,the uncertainty reduction in the meteorological initial condition by the meteorological DA contributed to a reduction of the forecast errors of both meteorology and air quality.

BACKGROUND Stem cell transplantation is a promising therapeutic option for curing perianal fistula in Crohn’s disease(CD).Anti-tumor necrotic factor(TNF)therapy combined with drainage procedure is effective as ***,previous studies are limited to proving whether the combination treatment of biologics and stem cell transplantation improves the effect of fistula *** This study aimed to evaluate the long-term outcomes of stem cell transplantation and compare Crohn’s perianal fistula(CPF)closure rates after stem cell transplantation with and without anti-TNF therapy,and to identify the factors affecting CPF closure and *** The patients with CD who underwent stem cell transplantation for treating perianal fistula in our institution between Jun 2014 and December 2022 were *** data were compared according to anti-TNF therapy and CPF *** A total of 65 patients were *** median age of females was 26 years(range:21-31)and that of males was 29(44.6%).The mean follow-up duration was 65.88±32.65 months,and complete closure was observed in 50(76.9%)*** closure rates were similar after stem cell transplantation with and without anti-TNF therapy(66.7%vs 81.6%at 3 year,P=0.098).The patients with fistula closure had short fistulous tract and infrequent proctitis and anorectal stricture(P=0.027,0.002,and 0.008,respectively).Clinical factors such as complexity,number of fistulas,presence of concurrent abscess,and medication were not significant for *** cumulative 1-,2-,and 3-year closure rates were 66.2%,73.8%,and 75.4%,*** Anti-TNF therapy does not increase CPF closure rates in patients with stem cell ***,both refractory and non-refractory CPF have similar closure rates after additional anti-TNF *** tract length,proctitis,and anal stricture are risk factors for non-closure in patients with CPF after stem cell transplantation.
