Thioredoxin Reductase Type C （NTRC） Orchestrates enhanced Thermotolerance to Arabidopsis by Its Redox-Dependent holdase Chaperone Function

Genevestigator analysis has indicated heat shock induction of transcripts for NADPh-thioredoxin reduc-tase, type C （NTRC） in the light. here we show overexpression of NTRC in Arabidopsis （NTRC°e） resulting in enhanced tolerance to heat shock, whereas NTRC knockout mutant plants （ntrcl） exhibit a temperature sensitive phenotype. To investigate the underlying mechanism of this phenotype, we analyzed the protein＇s biochemical properties and protein structure. NTRC assembles into homopolymeric structures of varying complexity with functions as a disulfide reductase, a foldase chaperone, and as a holdase chaperone. The multiple functions of NTRC are closely correlated with protein structure.. Complexes of higher molecular weight （hMW） showed stronger activity as a holdase chaperone, while low molecular weight （LMW） species exhibited weaker holdase chaperone activity but stronger disulfide reductase and fol-dase chaperone activities. heat shock converted LMW proteins into hMW complexes. Mutations of the two active site Cys residues of NTRC into Ser （C217/454S-NTRC） led to a complete inactivation of its disulfide reductase and foldase chaperone functions, but conferred only a slight decrease in its holdase chaperone function. The overexpression of the mutated C217/454S-NTRC provided Arabidopsis with a similar degree of thermotolerance compared with that of NTRC°e plants. however, after prolonged incubation under heat shock, NTRC°e plants tolerated the stress to a higher degree than C217/454S-NTRC°e plants. The results suggest that the heat shock-mediated holdase chaperone function of NTRC is responsible for the increased thermotolerance of Arabidopsis and the activity is significantly supported by NADPh.

A case of hypereosinophilic syndrome presenting with intractable gastric ulcers

We report a rare case of hypereosinophilic syndrome(heS)presenting with intractable gastric ulcers.A 71-year-old man was admitted with.epigastric *** endoscopic findings revealed multiple,active gastric ulcers in the gastric *** underwent helicobacter pylori(h pylori)eradication therapy followed by proton pump inhibitor(PPI)***,follow-up endoscopy at 4,6,10 and 14 mo revealed persistent multiple gastric ulcers without significant *** proportion of his eosinophil count increased to 43%(total count:7903/mm 3).Abdominal-pelvic and chest computed tomography scans showed multiple small nodules in the liver and both *** endoscopic biopsy specimen taken from the gastric antrum revealed prominent eosinophilic infiltration,and the liver biopsy specimen also showed eosinophilic infiltration in the portal tract and sinusoid.A bone marrow biopsy disclosed eosinophilic hyperplasia as well as increased cellularity of 70%.The patient was finally diagnosed with heS involving the stomach,liver,lung,and bone *** gastric ulcers do not improve despite h pylori eradication and prolonged PPI therapy,infiltrative gastric disorders such as heS should be considered.

evaluation of characteristics on titanium surface treatment for absorption of functional groups

Background In order to bind or fix bioactive materials directly to the surface of a Ti implant, the prior binding process of functional groups （FGs, -COoh and -oh） to the implant surface is necessary. Conventional binding processes are so high-cost and complex, so it is essential to find a simple and effective procedure for Ti-FG *** Various electrolyte compositions and electrochemical processing were adopted in this study to develop a relatively simple and effective Ti-FG binding process. The ability of Ti-FG binding and calcium （Ca）/phosphorous （P）absorption and corrosion resistance were evaluated according to various titanium surface treatment in electrolyte involving -COoh and -oh ion by using X ray photoelectron spectroscopy （XPS）, field emission scanning electron microscope （Fe-SeM） and potentiodynamic scan method *** In cases of -COoh, the anodic oxidation process （AN） showed an effective binding ability between -COoh and Ti surface. On the other hand, in cases of -oh, there were no significant differences in the result between the conditions used. In regard to the absorption of Ca and P on Ti surface, there was a minimal amount of Ca absorbed but no P was absorbed. The anodic oxidation series showed homogenous corrosion, whereas the electrolyte immersion （eL）series showed unstable corrosion. Although.eL-oh showed a novel corrosion potential, the eL-COoh series showed good corrosion resistance over the anodic potential *** The ability of binding between FG and the Ti surface and Ca/P absorption were strongly associated with the surface potential （ξ, potential）, which was dependent on the ph of the electrolyte. Accordingly, in order to ach.eve the effective absorption of various FGs on the Ti surface, it is needed to develop the combination process in addition to the electric affinity, relation with the ξ, potential.

An experiment was conducted to determine how an e. coil challenge and dietary clays affect the intestinal barrier of pigs. Two groups of 32 pigs （initial BW： 6.9 ± 1.0 kg） were distributed in a 2 x 4 factorial arrangement of a randomized complete block design （2 challenge treatments： sham or e. coil, and 4 dietary treatments： control, 0.3% smectite A, 0.3% smectite B and 0.3% zeolite）, with 8 replicates total. Diarrhea score, growth performance, goblet cell size and number, bacterial translocation from intestinal lumen to lymph nodes, intestinal morphology, and relative amounts of sulfo and sialo mucins were measured. The e. coli challenge reduced performance, increased goblet cell size and number in the ileum, increased bacterial translocation from the intestinal lumen to the lymph nodes, and increased ileal crypt depth. One of the clays （smectite A） tended to increase goblet cell size in ileum, which may indicate enhanced protection. In conclusion, e. coli infection degrades intestinal barrier integrity but smectite A may enhance it.

Berberine inhibits inflammatory activation of rat brain microglia

Chronic activation of microglial cells endangers neuronal survival through the release of various proinflammatory and neurotoxic factors. Berberine, the effective ingredient of Coptidis Rhizoma and Cortex Phellodendti, has a wide range of pharmacological functions, including anti-inflammatory, anti-atherosclerotic and anti-cancer effects. The neuroprotective potential of berberine has previously been demonstrated. The present study aimed to examine whether berberine could repress microglial activation and can be considered a potential therapeutic candidate to target neurodegenerative diseases. Primary microglial cells and BV2 microglial cells were cultured and stimulated with bacterial lipopolysaccharide （LPS）. Berberine chloride was treated prior to LPS or simultaneously with LPS stimulation. Results revealed that berberine was effective at inhibiting nitric oxide release from primary microglial cells when cells were exposed to the compound prior to LPS or simultaneously with LPS. It also reduced the LPS-stimulated production of tumor necrosis factor-α, interleukin-1β, prostaglandin e2, and intracellular reactive oxygen species and nuclear factor-kappa activation. Additionally, berberine reduced nitric oxide release from microglia stimulated with interferon-γ and amyloid β. These results suggest that berberine provides neuroprotection by reducing the production of various neurotoxic molecules from activated microglia.

A flexible electromagnetic interference(eMI)sh.elding film was innovatively fabricated using both low melting temperature solder arrays and amorphous soft magnet particulates consolidated on a polyethylene terephthalate(PeT)***,the IneSneBi solder arrays presented a low melting point of 99.4C,which.enabled attach.ent to the heat-sensitive plastic substrate without any thermal damage,and a low electrical resistivity of 14.1 mU cm,making them very effective at interrupting eM ***,the solder arrays with a high thermal conductivity of 61.2 W/(m$K)at 298.15 K were also useful as a thermal conductor for a heat ***,the solder arrays provide efficient electrical and thermal channels for electron transport induced by abruptly or consistently created eM waves due to suddenly turning on and the long term operation of electronics,***,the Fe-based magnetic particulates were added and resulted in effective saturation magnetization of 161.1 emu/g,remanence of 2.9 emu/g,and coercivity of 12.5 G,thus were able to disturb the eM pathway because of their soft magnetic ***,the hybrid eMI sh.elding film with.exceptionally high.electrical and thermal conductivity and superior soft magnetic properties provided remarkable sh.elding effectiveness of 102.5 dB at 10.2 Ghz.

Apocrine carcinoma is a rare malignant adnexal neoplasm. The differential diagnosis between apocrine carcinoma and cutaneous metastasis is often difficult. here, we report a case of locally recurrent penile apocrine carcinoma initially diagnosed as metastatic adenocarcinoma of the colon. A 75-year-old man with a history of surgical resection due to sigmoid colon cancer and penile metastasis two years prior to this study presented with a nodule at the left penile base. he underwent a wide local resection of the penile mass under a suggested preoperative diagnosis of extra-mammary Paget＇s disease （eMPD） associated with previous sigmoid colon cancer. however, the previously and currently resected penile masses were identified as primary apocrine carcinoma upon hematoxylin and eosin （h.e） staining and immunohistochemical staining. Although the incidence is extremely rare, both clinicians and pathologists should be alert to the possibility of synchronous double primary apocrine carcinoma in cancer patients with malignant cutaneous lesions.

The identification of jets originating from quarks and gluons,often referred to as quark/gluon tagging,plays an important role in various analyses performed at the Large hadron Collider,as Standard Model measurements and searches for new particles decaying to quarks often rely on suppressing a large gluon-induced *** paper describes the measurement of the efficiencies of quark/gluon taggers developed within the ATLAS Collaboration,using √s=13 TeV proton–proton collision data with an integrated luminosity of 140 fb^(-1) collected by the ATLAS *** taggers with high performances in rejecting jets from gluon over jets from quarks are studied:one tagger is based on requirements on the number of inner-detector tracks associated with the jet,and the other combines several jet substructure observables using a boosted decision tree.A method is established to determine the quark/gluon fraction in data,by using quark/gluon-enriched subsamples defined by the jet *** in tagging efficiency between data and simulation are provided for jets with transverse momentum between 500 GeV and 2 TeV and for multiple tagger working points.
