Interactions among deep-sea mussels and their epibiotic and endosymbiotic chemoautotrophic bacteria: Insights from multi-omics analysis

Endosymbiosis with Gammaproteobacteria is fundamental for the success of bathymodioline mussels in deep-sea chemosynthesis-based ecosystems. however, the recent discovery of Campylobacteria on the gill surfaces of these mussels suggests that these host-bacterial relationships may be more complex than previously thought. Using the cold-seep mussel(Gigantidas haimaensis) as a model, we explored this hostbacterial system by assembling the host transcriptome and genomes of its epibiotic Campylobacteria and endosymbiotic Gammaproteobacteria and quantifying their gene and protein expression levels. We found that the epibiont applies a sulfur oxidizing(SOX)multienzyme complex with the acquisition of sox B from Gammaproteobacteria for energy production and switched from a reductive tricarboxylic acid (rTCA) cycle to a Calvin-Benson-Bassham(CBB)cycle for carbon assimilation. The host provides metabolic intermediates, inorganic carbon, and thiosulfate to satisfy the materials and energy requirements of the epibiont, but whether the epibiont benefits the host is unclear. The endosymbiont adopts methane oxidation and the ribulose monophosphate pathway(Ru MP) for energy production, providing the major source of energy for itself and the host. The host obtains most of its nutrients, such as lysine, glutamine, valine,isoleucine, leucine, histidine, and folate, from the endosymbiont. In addition, host pattern recognition receptors, including toll-like receptors, peptidoglycan recognition proteins, and C-type lectins, may participate in bacterial infection, maintenance, and population regulation. Overall, this study provides insights into the complex host-bacterial relationships that have enabled mussels and bacteria to thrive in deep-sea chemosynthetic ecosystems.

DEAR EDITOR,Ancient DNA(a DNA) from mollusc shells is considered a potential archive of historical biodiversity and ***, such information is currently lacking for mollusc shells from the deep ocean, especially those from acidic chemosynthetic environments theoretically unsuitable for longterm DNA preservation. Here, we report on the recovery of mitochondrial and nuclear gene markers by Illumina sequencing of a DNA from three shells of Archivesica nanshaensis – a hydrocarbon-seep vesicomyid clam previously known only from a pair of empty shells collected at a depth of 2626 m in the South china Sea.

Background and purpose Brainomix e-Stroke is an artificial intelligence-based decision support tool that aids the interpretation of CT imaging in the context of acute *** e-Stroke has the potential to improve the speed and accuracy of diagnosis,real-world validation is *** aim of this study was to prospectively evaluate the performance of Brainomix e-Stroke in an unselected cohort of patients with suspected acute ischaemic *** The study cohort included all patients admitted to the University College London hospital Hyperacute Stroke Unit between October 2021 and April *** e-ASPECTS and e-CTA,the ground truth was determined by a neuroradiologist with access to all clinical and imaging *** e-CTP,the values of the core infarct and ischaemic penumbra were compared with those derived from ***,an alternate software used at our *** 1163 studies were performed in 551 patients admitted during the study *** these,1130(97.2%)were successfully processed by e-Stroke in an average of *** identifying acute middle cerebral artery territory ischaemia,e-ASPECTS had an accuracy of 77.0%and was more specific(83.5%)than sensitive(58.6%).The accuracy for identifying hyperdense thrombus was lower(69.1%),which was mainly due to many false positives(positive predictive value of 22.9%).Identification of acute haemorrhage was highly accurate(97.8%)with a sensitivity of 100%and a specificity of 97.6%;false positives were typically caused by areas of *** accuracy of e-CTA for large vessel occlusions was 91.5%.The core infarct and ischaemic penumbra volumes provided by e-CTP strongly correlated with those provided by ***(ρ=0.804—0.979).Conclusion Brainomix e-Stroke software provides rapid and reliable analysis of CT imaging in the acute stroke setting although,in line with the manufacturer’s guidance,it should be used as an adjunct to expert interpretation rather than a standalone decision-making tool.

The Basidiomycota constitutes a major phylum of the kingdom Fungi and is second in species numbers to the *** present work provides an overview of all validly published,currently used basidiomycete genera to date in a single *** outline of all genera of Basidiomycota is provided,which includes 1928 currently used genera names,with 1263 synonyms,which are distributed in 241 families,68 orders,18 classes and four *** provide brief notes for each accepted genus including information on classification,number of accepted species,type species,life mode,habitat,distribution,and sequence ***,three phylogenetic analyses with combined LSU,SSU,5.8s,rpb1,rpb2,and ef1 datasets for the subphyla Agaricomycotina,Pucciniomycotina and Ustilaginomycotina are conducted,*** time estimates are provided to the family level with 632 species from 62 orders,168 families and 605 *** study indicates that the divergence times of the subphyla in Basidiomycota are 406-430 Mya,classes are 211-383 Mya,and orders are 99-323 Mya,which are largely consistent with previous *** this study,all phylogenetically supported families were dated,with the families of Agaricomycotina diverging from 27-178 Mya,Pucciniomycotina from 85-222 Mya,and Ustilaginomycotina from 79-177 *** times as additional criterion in ranking provide additional evidence to resolve taxonomic problems in the Basidiomycota taxonomic system,and also provide a better understanding of their phylogeny and evolution.
