Telomeric DnA breaks in human induced pluripotent stem cells trigger ATR-mediated arrest and telomerase-independent telomere damage repair

Although mechanisms of telomere protection are well-defined in differentiated cells,how stem cells sense and respond to telomere dysfunction,in particular telomeric double-strand breaks(DSBs),is poorly ***,we report the DnA damage signaling,cell cycle,and transcriptome changes in human induced pluripotent stem cells(iPSCs)in response to telomere-internal *** engineer human iPSCs with an inducible TRF1-FokI fusion protein to acutely induce DSBs at *** this model,we demonstrate that TRF1-FokI DSBs activate an ATR-dependent DnA damage response,which leads to p53-independent cell cycle arrest in *** CRISPR–Cas9 to cripple the catalytic domain of telomerase reverse transcriptase,we show that telomerase is largely dispensable for survival and lengthening of TRF1-FokI-cleaved telomeres,which instead are effectively repaired by robust homologous recombination(HR).In contrast to HR-based telomere maintenance in mouse embryonic stem cells,where HR causes ZSCAn4-dependent extension of telomeres beyond their initial lengths,HR-based repair of telomeric breaks is sufficient to maintain iPSC telomeres at a normal length,which is compatible with sustained survival of the cells over several days of TRF1-FokI *** findings suggest a previously unappreciated role for HR in telomere maintenance in telomerase-positive iPSCs and reveal distinct iPSC-specific responses to targeted telomeric DnA damage.

Phase composition,conductivity,and sensor properties of cerium-doped indium oxide

The hydrothermal synthesis of In_(2)O_(3)and CeO_(2)–In_(2)O_(3)is investigated as well as the properties of sensor layers based on these *** the synthesis of In_(2)O_(3),intermediate products In(OH)_(3)and InOOH are formed,which are the precursors of stable cubic(c-In_(2)O_(3))and metastable rhombohedral(rh-In_(2)O_(3))phases,respectively.A transition from c-In_(2)O_(3)to rh-In_(2)O_(3)is observed with the addition of CeO_(2).The introduction of cerium into rh-In_(2)O_(3)results in a decrease in the sensor response to hydrogen,while it increases in composites based on c-In_(2)O_(3).The data on the sensor activity of the composites correlate with XPS results in which CeO_(2)causes a decrease in the concentrations of chemisorbed oxygen and oxygen vacancies in rh-In_(2)O_(3).The reverse situation is observed in composites based on c-In_(2)O_(3).Compared to In_(2)O_(3)and CeO_(2)–In_(2)O_(3)obtained by other methods,the synthesized composites demonstrate maximum response to H_(2)at low temperatures by 70–100℃,and have short response time(0.2–0.5 s),short recovery time(6–7 s),and long-term stability.A model is proposed for the dependence of sensitivity on the direction of electron transfer between In_(2)O_(3)and CeO_(2).

The common ways to activate a chemical reaction are by heat,electric current,or ***,mechanochemistry,where the chemical reaction is activated by applied mechanical force,is less common and only poorly understood at the atomic *** we report a tip-induced activation of chemical reaction of carbon monoxide to dioxide on oxidized rutile TiO_(2)(110)*** activation is studied by atomic force microscopy,Kelvin probe force microscopy under ultrahigh-vacuum and liquid nitrogen temperature conditions,and density functional theory(DFT)*** reaction is inferred from hysteretic behavior of frequency shift signal further supported by vector force mapping of vertical and lateral forces needed to trigger the chemical reaction with torque motion of carbon monoxide towards an oxygen *** reaction is found to proceed stochastically at very small tip-sample ***,the local contact potential difference reveals the atomic-scale charge redistribution in the reactants required to unlock the *** results open up new insights into the mechanochemistry on metal oxide surfaces at the atomic scale.

Maximizing aged photovoltaic array power:a computer modelling study

Photovoltaic(PV)modules age with time for various reasons such as corroded joints and terminals and glass coating defects,and their ageing degrades the PV array *** the help of the PV array numerical model,this paper explores the effects of PV module ageing on the PV array power,and the power gains and costs of rearranging and recabling aged PV modules in a PV *** numerical PV array model is first revised to account for module ageing,rearrangement and recabling,with the relevant equations presented *** updated numerical model is then used to obtain the array powers for seven different PV *** power results are then analysed in view of the attributes of the seven PV array examples.A guiding method to recommend recabling after rearranging aged modules is then proposed,leading to further significant power gains,while eliminating intra-row *** certain conditions are met,it was shown that recabling PV modules after rearranging them may lead to further significant power gains,reaching 57%and 98%in two considered PV array *** gains are possible in other arrays.A cost-benefit analysis weighing annual power gains versus estimated recabling costs is also given for the seven considered PV array examples to guide recabling decisions based on technical and economic *** the considered examples,recabling costs can be recovered inns due to our proposed rearranging and recabling the PV arrays ranged between 73%and 131%in the considered examples—well over the gains reported in the ***,the cost of our static module rearrangement and recabling method outshines the costs of dynamic reconfiguration methods recently published in the literature.

Predicting uniaxial compressive strength of tuff after accelerated freeze-thaw testing: Comparative analysis of regression models and artificial neural networks

Ignimbrites have been widely used as building materials in many historical and touristic structures in the Kayseri region of Türkiye. Their diverse colours and textures make them a popular choice for modern construction as well. However, ignimbrites are particularly vulnerable to atmospheric conditions, such as freeze-thaw cycles, due to their high porosity, which is a result of their formation process. When water enters the pores of the ignimbrites, it can freeze during cold weather. As the water freezes and expands, it generates internal stress within the stone, causing micro-cracks to develop. Over time, repeated freeze-thaw (F-T) cycles lead to the growth of these micro-cracks into larger cracks, compromising the structural integrity of the ignimbrites and eventually making them unsuitable for use as building materials. The determination of the long-term F-T performance of ignimbrites can be established after long F-T experimental processes. Determining the long-term F-T performance of ignimbrites typically requires extensive experimental testing over prolonged freeze-thaw cycles. To streamline this process, developing accurate predictive equations becomes crucial. In this study, such equations were formulated using classical regression analyses and artificial neural networks (Ann) based on data obtained from these experiments, allowing for the prediction of the F-T performance of ignimbrites and other similar building stones without the need for lengthy testing. In this study, uniaxial compressive strength, ultrasonic propagation velocity, apparent porosity and mass loss of ignimbrites after long-term F-T were determined. Following the F-T cycles, the disintegration rate was evaluated using decay function approaches, while uniaxial compressive strength (UCS) values were predicted with minimal input parameters through both regression and Ann analyses. Association of initial assessment variables and mortality in severe pediatric traumatic brain injury

Background Predictive scales have been used to prognosticate long-term outcomes of traumatic brain injury(TBI),but gaps remain in predicting mortality using initial trauma resuscitation *** sought to evaluate the association of clinical variables collected during the initial resuscitation of intubated pediatric severe patients with TBI with in-hospital *** Intubated pediatric trauma patientsjanuary 2011 to December 2020 were *** between initial trauma resuscitation variables(temperature,pulse,mean arterial blood pressure,GCS score,hemoglobin,international normalized ratio(InR),platelet count,oxygen saturation,end tidal carbon dioxide,blood glucose and pupillary response)and mortality were evaluated with multivariable logistic *** Among 314 patients,median age was 5.5 years(interquartile range(IQR):2.2-12.8),GCS score was 3(IQR:3-6),Head Abbreviated Injury Score(hAIS)was 4(IQR:3-5),and most had a severe(25-49)Injury Severity Score(ISS)(48.7%,153/314).Overall mortality was 26.8%.GCS score,hAIS,ISS,InR,platelet count,and blood glucose were associated with in-hospital mortality(all pnd GCS score increased,the odds of mortality *** 1-point increase in GCS score was associated with a 35%decrease in odds of *** hAIS,InR,and blood glucose increased,the odds of mortality *** each 1.0 unit increase in InR,the odds of mortality increased by 1427%.Conclusions Pediatric patients with severe TBI are at substantial risk for in-hospital *** are needed to examine whether earlier interventions targeting specific parameters of InR and blood glucose impact mortality.

Measurements based on optical microscopy can be severely impaired if the access exhibits variations of the refractive *** the case of fluctuating liquid-gas boundaries,refraction introduces dynamical aberrations that increase the measurement *** is prevalent at multiphase flows(***,film flows)that occur in many technical applications as for example in coating and cleaning processes and the water management in fuel *** this paper,we present a novel approach based on adaptive optics for correcting the dynamical aberrations in real time and thus reducing the measurement *** shape of the fluctuating water-air interface is sampled with a reflecting light beam(Fresnel Guide Star)and a Hartmann-Shack sensor which makes it possible to correct its influence with a deformable mirror in a closed ***-dimensional flow measurements are achieved by using a double-helix point spread *** measure the flow inside a sessile,oscillating 50-μl droplet on an opaque gas diffusion layer for fuel cells and show that the temporally varying refraction at the droplet surface causes a systematic underestimation of the flow field magnitude corresponding to the first droplet eigenmode which plays a major role in their detachment *** demonstrate that the adaptive optics correction is able to reduce this systematic ***,the adaptive optics system can pave the way to a deeper understanding of water droplet formation and detachment which can help to improve the efficiency of fuels cells.

Antibiotic ciprofloxacin removal from aqueous solutions by electrochemically activated persulfate process:Optimization,degradation pathways,and toxicology assessment

Ciprofloxacin(CIP)is a commonly used antibiotic in the fluoroquinolone group and is widely used in medical and veterinary medicine disciplines to treat bacterial *** CIP is discharged into the sewage system,it cannot be removed by a conventional wastewater treatment plant because of its recalcitrant *** this study,boron-doped diamond anode and persulfate were used to degrade CIP in an aquatic solution by creating an electrochemically activated persulfate(EAP)*** added to the system as a coactivator and the process was called EAP+*** effects of independent variables,including pH,Fe^(2+),persulfate concentration,and electrolysis time on the systemwere optimized using the response surface *** results showed that the EAP+Fe process removed 94%of CIP under the following optimum conditions:A pH of 3,persulfate/Fe^(2+)concentration of 0.4 mmol/L,initial CIP concentration 30 mg/L,and electrolysis time of 12.64 *** removal efficiency was increased from 65.10%to 94.35%by adding Fe^(2+)as a transition *** degradation products,7 pathways,and 78 intermediates of CIP were studied,and three of those intermediates(m/z 298,498,and 505)were *** toxicological analysis based on toxicity estimation software results indicated that some degradation products of CIP were toxic to targeted animals,including fathead minnow,Daphnia magna,Tetrahymena pyriformis,and *** optimumoperation costswere similar in EAP and EAP+Fe processes,approximately 0.54€/m^(3).
