Electroosmotic Water Vapor Transport across Novel, Smart, Functionalized Conducting Polymer Microporous Membranes in Active Mode at Very High Rates, with Concomitant Chemical Warfare (CW) Agent Blocking

Electroosmotic water vapor transport (WVT) across very thin, flexible, functionalized conducting polymer (CP) microporous (μP) membranes at a very high rate is reported. Both passive and active (6 VDC applied) WVT are reported, the latter for the first time ever. WVT occurs with concomitant, effective blocking of chemical warfare (CW) agents, again demonstrated for the first time ever. Initial active liquid||membrane||liquid interface studies demonstrated WVT rates of >1.7 × 10-5g .mm-2s-1, >3 × the highest prior reported values of 5 × ***-2s-1. Subsequent vapor||membrane|| vapor interface studies using industry-standard methods (including ASTM E96B Upright Cup (“WVT”), ASTM F2298 (“Dynamic Moisture Permeation Cell”) and ASTM F1868 (“Sweating Guard Hotplate”) were done at independent, external labs for independent corroboration. These yielded, e.g., WVT values of2564.4 g.m2.d-1 (passive) and3706.7 g.m2d-1 (active), to be compared with the highest (passive) value ever reported previously,984.8 g.m2.d-1 for a μP-Nylon membrane. More than 15 different membrane configurations, porosities and types were studied, including membranes with CP + organophosphate hydrolase (OPH), an enzyme reactive to CW agents. Efficient blocking of the actual CW agents GB, HD, VX is also reported, using the TOP-8-2-501standard. Membranes also passed all Industry-standard durability tests, e.g. ASTM D2261 (Tearing), ASTM D5034 (Breaking), ASTM D3886 (Abrasion), ASTM F392 (Gelbo Flex). Incorporation into smart soldiers’ garments was demonstrated;power consumption was -2. Mechanisms of enhanced WVT and CW agent blocking in the membranes are discussed.

Addition of Thiourea Host Monomer to Polymer Flocculants to Improve Selectivity of Phosphate Sorption

Inorganic phosphate is a common nutrient that is applied as a fertilizer to both agricultural fields as well as urban settings such as private yards, public parks and other urban landscaping. While phosphate typically binds tightly to soil, movement of phosphate off of application sites can occur through soil erosion. The soil and its bound phosphate can then end up in surface waters such as rivers and lakes. Phosphate found in surface water bodies exists both as bound to the suspended clay as well as that free in solution. Elevated phosphate concentration in surface waters can lead to algal blooms and eutrophication. While the phosphate bound to clay in suspension in surface water bodies can be removed by commercially available polymer flocculants, the phosphate that is free in solution is more challenging as it is usually found in low concentrations and other anionic salts are generally present in higher concentrations. To remove phosphate from contaminated water systems, where other anions exist at higher concentrations, it is favorable to have a method of removal that is selective for phosphate. As a proof of principle, thiourea derivatized polymer flocculants were examined for the selective removal of phosphate in the presence of competing anions. The polymer flocculants exhibited selectivity for phosphate through hydrogen bonding and were effective at removing up to 43% of phosphate from simulated wastewater. Computational studies and 1H NMR were used to investigate the selectivity of the thiourea monomer for phosphate over competing anions such as chloride and sulfate.

Siah proteins, the mammalian homologs of fly seven-in-absentia protein, have been implicated as tumor suppressors that may have pro-apoptotic activity. Prior work has also characterized Siah family's E3 ligase activity and potential substrates, however, the mechanisms by which Siah induces apoptotic death have not been clear. In our study, we identify SIAH1 as a binding partner for POSH, a scaffold protein that we previously showed to be involved in apoptotic activation of the jNK pathway (EMBO j;Xu et al., 2003). We show that SIAH1 promotes cell death by activating the jNK pathway and the E3 ligase activity of SIAH1 is essential for SIAH1-evoked death. Moreover, we demonstrate that SIAH1 plays a required role in death evoked by DNA damage or trophic factor deprivation. We further find that apoptotic stimuli markedly elevate endogenous SIAH1 protein by a mechanism due to SIAH1 stabilization and that such stabilization requires activation of the jNK pathway components including MLKs, MKKs and jNKs. These findings thus reveal a feed-forward loop by which apoptotic stimuli recruit SIAH1 to participate in the jNK pathway and thereby, in promotion of death. We have provided a substantial amount of data on endogenous proteins as well as a mechanism by which the jNK pathway induces SIAH1A stabilization. We find POSH is essential for SIAH1 stabilization induced by activation of the jNK pathway. In addition, apoptotic stimuli and jNK pathway activation lead to phosphorylation of SIAH1 on tyrosines 100 and 126 and that this enhances its interaction with POSH. The enhanced interaction with POSH, in turn, promotes SIAH1 stabilization.

Benzodiazepines and other benzodiazepine receptor agonists, such as the “Z” drugs, are widely prescribed medications mainly used for treating anxiety and seizures, and for inducing sedation. Unfortunately, despite their popularity, benzodiazepine prescribing often exceeds recommendations and the consequences can be severe. On September 23, 2020, the United States FDA announced a new requirement for a Boxed Warning for benzodiazepines prescribing. Along with this announcement, the FDA stated that relevant information regarding the initiation, continuation, and discontinuation of benzodiazepines is lacking. Here, we describe initial pilot studies intended to investigate the questions 1) can animal models be developed that demonstrate benzodiazepine physical dependence and/or withdrawal symptoms, and 2) determine whether translocator protein (TSPO) plays a role in benzodiazepine dependence and/or withdrawal processes. The former was demonstrated, methodological limitations prevented the latter.

Metabolic engineering strategies have been successfully implemented to improve the production of isobutanol,a next-generation biofuel,in Saccharomyces ***,we explore how two of these strategies,pathway re-localization and redox cofactor-balancing,affect the performance and physiology of isobutanol producing *** equipped yeast with isobutanol cassettes which had either a mitochondrial or cytosolic localized isobutanol pathway and used either a redox-imbalanced(NADPH-dependent)or redox-balanced(NADH-dependent)ketol-acid reductoisomerase *** then conducted transcriptomic,proteomic and metabolomic analyses to elucidate molecular differences between the engineered *** localization had a large effect on isobutanol production with the strain expressing the mitochondrial-localized enzymes producing 3.8-fold more isobutanol than strains expressing the cytosolic ***-balancing did not improve isobutanol titers and instead the strain with the redox-imbalanced pathway produced 1.5-fold more isobutanol than the balanced version,albeit at low overall pathway *** genomic analyses suggested that the poor performances of the cytosolic pathway strains were in part due to a shortage in cytosolic Fe-S clusters,which are required cofactors for the dihydroxyacid dehydratase *** then demonstrated that this cofactor limitation may be partially recovered by disrupting iron homeostasis with a fra2 mutation,thereby increasing cellular iron *** resulting isobutanol titer of the fra2 null strain harboring a cytosolic-localized isobutanol pathway outperformed the strain with the mitochondrial-localized pathway by 1.3-fold,demonstrating that both localizations can support flux to isobutanol.
