While stock markets and economies are smoldering all over the world, stem cell science is on fire. The promise of curing many untreatable forms of human male infertility is now one small step closer based on a recent paper in Cell. In this work, scientists from Kyoto University in japan created mature, fertile sperm from embryonic and induced pluripotent stem cells by grabbing the primitive germ cells called primordial germ cells just as they were being made from their pluripotent precursors in vitro and transplanting them back into genetically sterile mice.

As the field of assisted reproduction has advanced, many previously untreatable men are now biological fathers. Although finding sperm in men with obstructive azoospermia is not difficult, locating and retrieving spermatozoa in men with non-obstructive azoospermia remains a clinical challenge, largely because sperm production in these men can be patchy or focal in nature. In response to this challenge, strategies such as fine-needle aspiration （FNA） mapping have been developed to find spermatozoa. This review discusses the history, evolution and current clinical utility and findings with FNA mapping for male infertility）. Review of the current literature in the English language on FNA （diagnostic or therapeutic） with a keyword focuses on sperm detection, retrieval, safety and complications. FNA was described in human medicine over 100 years ago. Testis FNA was described 45 years ago and FNA ＇mapping＇ of spermatozoa was described in 1997. This comparative review of the literature on sperm detection and complication rates with FNA and open testis biopsy or microdissection procedures suggests that FNA is highly informative, minimally invasive and is associated with fewer complications than other commonly used approaches to sperm detection in non-obstructive azoospermic patients. FNA mapping has gained considerable traction as an informative, ＇testis sparing＇ technique for sperm detection in non-obstructive azoospermia. With knowledge of sperm presence and location prior to sperm retrieval, FNA maps can help clinicians tailor sperm retrieval to optimize time, effort and extent of procedures needed to procure spermatozoa in these difficult cases.

Meat is a rich nutrient matrix that provides a suitable environment for proliferation of meat spoilage microbiota and common food-bom pathogens, therefore adequate preservation has to be applied in order to preserve its safety and qualib：. Lantibiotic nisin is not conunonly used in the meat industry, in spite of its beneficial effect in e.g. dairy industry. Theretbre, here, the effect of nisin was searched in traditional fermented sausage Gombasek experimentally contaminated with Listeria innocua Lil strata Ihe antimicrobial effect of nisin was demonstrated by the Lil strain decrease at day 2 （after contamination） with the difference 1.79 logarithmic （log） cycles comparing with the control sausage （CLi）. At week l, the difference 4.14 log cycles was noted in the experimental sausages comparing with CLi sausages with prolonging inhibitory effect up to week 2; although the difference was diminished （1.82 log cycles）; but higher than at day 2, The pH values as well as lactic acid values were not influenced by the nisin treatment （pH value in the reference sausage was 5.0, in CLi 5.7 and in the experimental sausage E, 5.1; amount of lactic acid in t： was 0.099 mmol L^-1： and in CLi 0.107 mmol L^-1）.

To determine whether center to center discrepancies in the ability to locate sperm in infertile testes with abnormal histology stems in part from inconsistencies in pathologists’ readings of testis biopsies. Design: Prospective cohort study. Setting: Academic male infertility practice. Patient(s): Consecutive series of azoospermic men referred with testis biopsy slides between 1998 and 2003. Intervention(s): Testis biopsy histologies on azoospermic patients referred for infertility care were re-reviewed by a single pathologist blinded to the original reading. Subsequent infertility care was guided by the findings from the second histologic reading. Main Outcome Measure(s): Agreement between the outside and in-house review of testis biopsy readings was assessed with the kappa statistic. Pregnancy outcomes that resulted from clinical decisions informed by the second histologic readings were also assessed. Result(s): Among 113 histologic specimens, re-review was complicated by fixation artifacts in 18 cases (16% ) and insufficient biopsy sample size in 13 cases (12% ). The kappa score for interobserver agreement in readings was 0.43 (95% CI 0.32- 0.54). Mixed histology patterns in particular were underappreciated by outside pathologists (13% of cases on original reading, 36% of cases on review). In 27% of all cases, the differences in biopsy readings had a significant impact on clinical management. Conclusion(s): A correlation between independent testis histology readings in azoospermic men demonstrates frequent inconsistencies. These differences contribute to inaccurate phenotyping of male infertility and can significantly impact the direction of infertility care. These findings highlight the need for a standardized approach to testis histologic review.

Objective: To conduct clinical, genetic, and molecular diagnostics of two sisters with typical symptoms of complete androgen insensitivity syndrome. Design: Case report. Setting: Research laboratory at a university of medical science. Patient(s): Two patients with 46,XY karyotype and a female phenotype were diagnosed because of primary amenorrhea. Their sister with 46,XX karyotype, her daughter, and five other family members including their mother also were examined. Intervention(s): Orchiectomy, estrogen substitution therapy. Main Outcome Measure(s): Cancer prophylaxis. Result(s): Multiple-temperature single-stranded conformation polymorphism and sequence analyses of the androgen receptor gene(AR) revealed a c.C2812T transition in exon 7 in the two sisters. Their mother and the third sister(46,XX) were carriers of the same mutation. This mutation, which previously had never been reported, resulted in Pro817Leu substitution in the ligand-binding domain of the androgen. Computer simulation of structural changes generated by Pro817Leu substitution revealed appreciable conformational changes in the region responsible for dimerization of the receptor. Conclusion(s): The novel c.C2812T transition that might impair dimerization of the receptor is responsible for the clinical symptoms of complete androgen insensitivity syndrome in the affected individuals. Molecular analysis of AR proved to be very useful for genetic counseling of the unaffected sister, who was a carrier of the same mutation.
