BACKGROUND Inflammatory bowel diseases(IBD)have been associated with a low quality of life(QoL)and a negative impact on work productivity compared to the general *** about disease control,patient-reported outcomes(PROs),treatment patterns and use of healthcare resources is relevant to optimizing IBD *** To describe QoL and work productivity and activity impairment(WPAI),treatment patterns and use of healthcare resources among IBD patients in *** A multicenter cross-sectional study included adult outpatients who were previously diagnosed with moderate to severe Crohn’s disease(CD)or ulcerative colitis(UC).At enrolment,active CD and UC were defined as having a Harvey Bradshaw Index≥8 or a CD Activity Index≥220 or calprotectin>200μg/g or previous colonoscopy results suggestive of inadequate control(per investigator criteria)and a 9-point partial mayo score≥5,*** PRO assessment included the QoL questionnaires SF-36 and EQ-5D-5L,the Inflammatory Bowel Disease Questionnaire(IBDQ),and the WPAI *** about healthcare resources and treatment during the previous 3 years was collected from medical ***-square,Fisher’s exact and Student’s t-/mann-Whitney U tests were used to compare PROs,treatment patterns and the use of healthcare resources by disease activity(α=0.05).RESULTS Of the 407 patients in this study(CD/UC:64.9%/35.1%,mean age 42.9/45.9 years,54.2%/56.6%female,38.3%/37.1%employed),44.7%/25.2%presented moderate-to-severe CD/UC activity,respectively,at *** in median values for CD/UC,respectively,the SF-36 physical component was 46.6/44.7 and the mental component was 45.2/44.2,the EQ-visual analog scale score was 80.0/70.0,and the IBDQ overall score was 164.0/*** to severe activity,female gender,being unemployed,a lower educational level and lower income were associated with lower QoL(Pmedian work productivity impairment was 20%and 5%for CD and UC patients,re

Electrooxidation of sulfanilamide and its voltammetric determination in pharmaceutical formulation,human urine and serum on glassy carbon electrode

For the first time, sulfanilamide(SFD) was determined in otologic solution, human urine and serum by electroanalytical techniques on glassy carbon electrode(GCE). The cyclic voltammetry(CV) experiments showed an irreversible oxidation peak at t 1.06 V in 0.1 mol/L BRBS(p H ? 2.0) at 50 m V/s. Different voltammetric scan rates(from 10 to 250 m V/s) suggested that the oxidation of SFD on the GCE was a diffusioncontrolled process. Square-wave voltammetry(SWV) method under optimized conditions showed a linear response to SFD from 5.0 to 74.7 μmol/L(R ? 0.999) with detection and quantification limits of 0.92 and3.10 μmol/L, respectively. The developed SWV method showed better results for detection limit and linear range than the chronoamperometry method. It has been successfully applied to determine SFD concentration in pharmaceutical formulation, human urine and serum samples with recovery close to 100%.

Regularized quantile regression for SNP marker estimation of pig growth curves

Background: Genomic growth curves are general y defined only in terms of population mean; an alternative approach that has not yet been exploited in genomic analyses of growth curves is the Quantile Regression(QR). This methodology allows for the estimation of marker effects at different levels of the variable of interest. We aimed to propose and evaluate a regularized quantile regression for SNP marker effect estimation of pig growth curves, as well as to identify the chromosome regions of the most relevant markers and to estimate the genetic individual weight trajectory over time(genomic growth curve) under different quantiles(levels).Results: The regularized quantile regression(RQR) enabled the discovery, at different levels of interest(quantiles), of the most relevant markers al owing for the identification of QTL regions. We found the same relevant markers simultaneously affecting different growth curve parameters(mature weight and maturity rate): two(ALGA0096701 and ALGA0029483)for RQR(0.2), one(ALGA0096701) for RQR(0.5), and one(ALGA0003761) for RQR(0.8). Three average genomic growth curves were obtained and the behavior was explained by the curve in quantile 0.2, which differed from the ***: RQR allowed for the construction of genomic growth curves, which is the key to identifying and selecting the most desirable animals for breeding purposes. Furthermore, the proposed model enabled us to find, at different levels of interest(quantiles), the most relevant markers for each trait(growth curve parameter estimates) and their respective chromosomal positions(identification of new QTL regions for growth curves in pigs). These markers can be exploited under the context of marker assisted selection while aiming to change the shape of pig growth curves.

Anxiety-Behavior modulated by Ventral medial Prefrontal Cortex of Rats Submitted to the Vogel Conflict Test Involves a Local NmDA Receptor and Nitric Oxide

It was demonstrated in the Vogel conflict test (VCT) that the ventral portion of medial prefrontal cortex (vmPFC) of rats is involved with anxiety behavior. moreover, the vmPFC local glutamatergic and nitrergic system interaction is involved in modulation of fear conditioning, a model of anxiety. To better understand the role of the mPFC-glutamatergic and nitrergic system on the VTC behavior response, male Wistar rats (250 g) were water deprived for 48 h before the VCT. After 24 h of water deprivation, they were subjected to an initial 3-min non-punished (pre-test) drinking session. Twenty-four hours later bilateral microinjections of NmDA-antagonist LY235959 (4 nmol/200 nL), the specific nNOS inhibitor N-Propyl-L-arginine (N-Propyl –0.08 nmol/200 nL), the NO scavenger Carboxi-PTIO (C-PTIO, 2 nmol/200 nL) or 200nL of vehicle were applied in the vmPFC. After 10 min, the animals were submitted to 3-min punished-licking session. LY235959 increased the number of punished licks. Similar to LY235959, both N-Propyl and C-PTIO also increased the number of punished licks. No changes were observed when LY235959, N-Propyl and C-PTIO were micro- injected into vmPFC surrounding structures such as the cingulate cortex area 1, the corpus callosum and the tenia tecta. In control experiments these drugs did not change neither the number of unpunished licks nor had any effect in the tail-flick test. The results show that NO signaling in the vmPFC can modulate anxiety-behavior in the VCT by control punished behavior. moreover, this NO modulation could be associated with local glutamatergic activation through NmDA receptors.

This study aimed to understand bark thickness variations of Araucaria angustifolia(Bertol.)Kuntze trees growing in natural forest remnants in southern Brazil,and their relationship with quantitative and qualitative *** thickness must be accurately estimated in order to determine timber volume *** is an important variable for the sustainable management and conservation of araucaria *** spite of its importance and visibility,bark thickness variations have not been evaluated for this key species in southern Brazil.A total of 104 trees were selected,and their qualitative and quantitative attributes such as diameter at breast height(D_(BH)),height(H),crown base height(C_(BH)),crown length(C_(L)),social position(S_(P)),stoniness(S_(T)),position on the relief(P_(R)),vitality(V_T)and branch arrangement(B_(A))were *** trees were categorized into two groups:red bark or gray *** analysis and artificial neural networks(ANN)were used for modelling bark *** results indicate that:(1)bark thickness showed good correlation to D_(BH),with 0.76 as coefficient of determination(RS_P),0.540 as mean Absolute Error(m_(AE))and 22.4 root-meansquare error in percentage(R_(mSE%));(2)the trend changed according to bark colour,with significant differences for the intersection(_0–Pr>F:p=0.0124)and slope(β_(1)–Pr>F:p=0.0126)of bark thickness curves between groups;(3)the highest correlation of bark thickness was found with:D_(BH)(ρ=0.88),H(ρ=0.58),C_(BH)(ρ=0.46),S_(P)(ρ=-0.52),and B_(A)(ρ=-0.32);(4)modelling with ANN confirmed high adjustment(R^(2)=0.99)and accuracy(R_(mSE%)=3.0)of the *** is an efficient and robust technique for the modelling of various qualitative and quantitative attributes commonly used in forest *** effective use of ANN to estimate araucaria bark in natural forests reinforces its potential,besides the possibility of application for other forest species.

Determination and Removal of Endocrine Disruptors in Wastewater by Activated Carbon

This study aimed to evaluate the EDC （endocrine disruptors compounds） in the city of Sao Paulo＇s water sources, from samples collected at predetermined sampling points and to evaluate the adsorptive capacity of these compounds in different types of activated carbon. The effects of these EDC on humans are not well established due to the necessary large exposure time for the effect＇s manifestation. After tests using powdered and granular activated carbon, all samples were filtered under vacuum using cellulose acetate membrane （0.45 μm） to remove eventual impurities, and posteriorly carded out the solid-phase extraction SPE （solid-phase extraction） and chromatographic analysis. The results lead to the conclusion that both powdered activated carbon have removal effectiveness of these compounds by adsorption. Furthermore, great amount of endocrine disruptors were found at several sampling points in river and city＇s water reservoirs, which shows different levels of pollution of water sources, some of them responsible for the watersupply of the city of Silo Paulo, Brazil.

Optimization of the Chitinase Production by Different metarhizium anisopliae Strains under Solid-State Fermentation with Silkworm Chrysalis as Substrate Using CCRD

Entomopathogenic fungi, such as metarhizium anisopliae, are able to control various insect pests. These fungi attack the integument of the host using an enzymatic complex. Among the enzymes found in this complex, chitinase is an important component. However, the relation between the chitinase production and the virulence from different m. anisopliae strains has not been analyzed. In this manuscript it is presented the chitinase production by four m. anisopliae strains with different potential of virulence in Solid-State Fermentation using silkworm chrysalis as substrate. The higher chitinase level was obtained with the strain IBCB 360 (7.14 U/g of substrate) with potential virulence of 68% on Diatrea saccharalis. The enzyme production was optimized for all strains using a factorial planning (CCRD) considering the cultivation time and medium humidity as independent variables. The maximal production of chitinase was obtained at a range from 8 to 12-days old cultures and from 45% to 62% of moisture according to the surface response plot, with high R2 value. The enzyme production by the strain IBCB 167 was increased two-folds under optimized conditions, while for the strains IBCB 360 and 425 the chitinase production was increased four-folds and nine-folds for the strain IBCB 384.
