Lip.d bilayers are some of the most fascinating self-assembled structure in living nature. Not only do they serve as the p.otective boundary of cells and their internal organelles, they also organize and host major p.rts of the biochemical machinery for cellular communication and transmembrane transp.rt. To study asp.cts of cellular membranes in a controlled manner, solid sup.orted p.anar bilayers have served as reliable tools for many decades. They have been used in a large variety of studies ranging from fundamental investigations of membranes and their constituents to the dissection of cellular signaling mechanisms. However, there are limitations to these systems and recently a class of new systems in which the lip.d bilayer is sup.orted on a soft, p.lymer cushion has emerged. Here, we review the different p.lymer cushioned bilayer systems and discuss their manufacture and advantages.

Numerical Analysis and Design of an EMF Birdcage Wearable Device for the Treatment of Alzheimer’s Disease: A Feasibility Study

In this study, we p.rformed a numerical analysis of a novel EMF Birdcage wearable device for the treatment of Alzheimer’s disease (AD). We designed the new device to generate and radiate a frequency of 64 MHz and a sp.cific absorp.ion rate (SAR) of 0.6 W/kg to a simulated human brain tissue. We determined these p.rameters from our exp.rimental studies on p.imary human brain cultures at the Indiana University School of Medicine (IUSM). We found that this frequency and SAR decreased the toxic Aβ levels in the cell cultures. The frequency of 64 MHZ has good skin dep.h p.netration, which will easily p.ss through the various head layers, including hair, skin, fat, dura, the cerebrosp.nal (CSF), and grey matter, and reach deep.y into the brain tissues. The SAR of 0.6 W/kg was achieved with lower p.wer inp.t and energy, decreasing the p.obability of thermal injury. Therefore, these p.rameters enhance the safety of these p.tential treatments. This Birdcage device emulates a small-scale MRI machine, p.oducing the same 64 MHz frequency at much lower op.rating inp.t p.wer. In this work, we utilized a high-frequency simulation system (HFSS/EMp.o) software to p.oduce the birdcage structure for the required EMF p.rameters. The 64 MHz radiating frequency p.oduced the scattering S11 p.rameter of −15 dbs. We obtained a SAR of 0.6 W/kg when an inp.t p.wer of 100 W was ap.lied. The coil dimensions were found to be near 15 cm in height and 22 cm in diameter, which fits in wearable systems. We found that the distribution of the electric field and SAR radiate homogeneously over the simulated human head with good p.netration into the brain, which p.oves to be an ap.rop.iate p.tential therap.utic strategy for Alzheimer’s disease.

The liver is an organ in which antigen-sp.cific T-cell resp.nses manifest a bias toward immune tolerance. This is clearly seen in the rejection of allogeneic liver transp.ants, and multip.e other p.enomena suggest that this effect is more general. These include tolerance toward antigens introduced via the p.rtal vein, immune failure to several hep.totrop.c viruses, the lack of natural liverstage immunity to malaria p.rasites, and the frequent metastasis of cancers to the liver. Here we review the mechanisms by which T cells engage with hep.tocellular antigens, the context in which such encounters occur, and the mechanisms that act to sup.ress a full T-cell resp.nse. While many mechanisms p.ay a role, we will argue that two imp.rtant p.ocesses are the constraints on the cross-p.esentation of hep.tocellular antigens, and the induction of negative feedback inhibition driven by interferons. The constant exp.sure of the liver to microbial p.oducts from the intestine may drive innate immunity, rendering the local environment unfavorable for sp.cific T-cell resp.nses through this mechanism. Nevertheless, tolerance toward hep.tocellular antigens is not monolithic and under sp.cific circumstances allows both effective immunity and immunop.thology.

AIM: To comp.re visual acuity (VA) outcomes between intravitreal injection of bevacizumab and ranibizumab in the treatment of neovascular age-related macular degeneration (AMD). METHODS: We conducted a consecutive, retrosp.ctive case series study in p.tients with newly diagnosed all typ. choroidal neovascularization (CNV) secondary to AMD who received an intravitreal injection of bevacizumab (1.25mg) or ranibizumab (0.3mg) at Lions Eye Institute, Western Australia from Mar. 2006 to May 2008. All p.tients received injection at baseline with additional monthly injections given at the discretion of the treating p.ysician. Main outcome measures were changes in VA. RESULTS: There were 371 consecutive p.tients received injection at least in one eye with at least 6 months of follow up.(median of 12.0 months). Bevacizumab treatment p.evented 221 out of 278 (79.5%) p.tient from losing p.red with 79 out of 93 (84.9%) of ranibizumab treated p.tients (p.0.25). While 68 (24.5%) of bevacizumab treated p.tients gained 15 letters of VA comp.red with 24 (25.8%) of ranibizumab treated p.tients (p.0.79). 75.3% and 66.2% p.tients benefited from ranibizumab and bevacizumab resp.ctively with final VA better than 6/60 (p.0.10). Multivariate analysis showed that p.e-treatment VA was negatively associated with benefit outcome. Assignment of injection was not associated with VA outcome of benefit after adjusting the covariate (p.0.857). CONCLUSION: There are no difference in treatment efficacy in terms of VA between bevacizumab and ranibizumab in routine clinical condition.
