Changes of the gastric endocrine cells in the C57BL/6 mouse after implantation of murine lung carcinoma: An immunohistochemical quantitative study

AIM: The regional distributions and relative frequencies of some gastric endocrine cells of C57BL/6 mice were studied by immunohistochemical method using seven types of specific antisera against chromogranin A (CGA), serotonin, somatostatin, gastrin, cholecystokinin (CCK)-8, glucagon and human pancreatic polypeptide (HPP) after subcutaneous implantation of murine lung carcinoma (3LL) cells. METHODS: The experimental animals were divided into two groups, one is non-implanted sham and the other is 3LL-implanted group. Samples were collected from the two regions of stomach (fundus and pylorus) at 28 d after implantation of 3LL cells (1x105 cell/mouse). RESULTS: In this study, all the seven types of immunoreactive (IR) cells were identified except for HPP. Most of these IR cells in the gastric portion were generally spherical or spindle in shape (open-type cell) while cells showing round in shape (closed-type cell) were found occasionally. The regional distributions of gastric endocrine cells in the 3LL-implanted group were similar to those of non-implanted sham. However, significant decreases of some types of IR cells were detected in 3LL-implanted group compared to those of non-implanted sham. In addition, the IR cells showing degranulation were numerously detected in 3LL-implanted group. CGA-, serotonin- and somatostatin-IR cells in the fundus and pylorus regions, and gastrin-IR cells in the pylorus regions of 3LL-implanted groups significantly decreased compared to those of non-implanted sham. However, no changes on frequencies of CCK-8- and glucagon-IR cells were demonstrated between 3LL-implanted and non-implanted groups. CONCLUSION: Endocrine cells are the anatomical units responsible for the production of gut hormones, and the change in their density would reflect a change in the capacity of producing these hormones. BACKGROUND Enterotoxigenic Bacteroides fragilis(ETBF)causes colitis and diarrhea,and is considered a candidate pathogen in inflammatory bowel diseases as well as colorectal *** diseases are dependent on ETBF-secreted toxin(BFT).Dendritic cells(DCs)play an important role in directing the nature of adaptive immune responses to bacterial infection and heme oxygenase-1(HO-1)is involved in the regulation of DC *** To investigate the role of BFT in HO-1 expression in *** Murine DCs were generated from specific pathogen-free C57BL/6 and Nrf2−/−knockout *** were exposed to BFT,after which HO-1 expression and the related signaling factor activation were measured by quantitative RT-PCR,EMSA,fluorescent microscopy,immunoblot,and *** HO-1 expression was upregulated in DCs stimulated with *** BFT activated transcription factors such as NF-κB,AP-1,and Nrf2,activation of NF-κB and AP-1 was not involved in the induction of HO-1 expression in BFT-exposed ***,upregulation of HO-1 expression was dependent on Nrf2 activation in ***,HO-1 expression via Nrf2 in DCs was regulated by mitogenactivated protein kinases such as ERK and ***,BFT enhanced the production of reactive oxygen species(ROS)and inhibition of ROS production resulted in a significant decrease of phospho-ERK,phospho-p38,Nrf2,and HO-1 CONCLUSION These results suggest that signaling pathways involving ROS-mediated ERK and p38 mitogen-activated protein kinases-Nrf2 activation in DCs are required for HO-1 induction during exposure to ETBF-produced BFT.

Mechanical alloying of platinum with 5% ZrO_2 nanoparticles for glass making tools

Synthesis and characterization of mechanically alloyed Pt-5%ZrO2(volume fraction) for structural components in the glass industry were described. Zirconia(ZrO2) nanoparticles(<100 nm) were produced by the electrical explosion of zirconium(Zr) wires, and blended with platinum(Pt) powders(<44 ?m) for 2-72 h in ambient atmosphere. The Pt particle size followed the typical decreasing trend of the normal ball milling process up to 48 h, but particle agglomeration was observed at 72 h. The grain size evolution was similar to that of the particle size, dropping down to around 50 nm at 48 h. The root mean square strain of the Pt crystallites showed the opposite behavior, maximizing at 48 h with a subsequent relaxation process. For the 48 h ball milled powders, spark plasma sintering was carried out to form a bulk disk. The measured mass loss of the sintered bulk sample shows a decent thermal stability despite its relatively low density.

Severe thrombocytopenia before liver transplantation is associated with delayed recovery of thrombocytopenia regardless of donor type

AIM： To compare the recovery of thrombocytopenia and splenomegaly during long-term follow-up after liver transplantation in patients receiving a living donor transplant or a cadaveric donor transplant. METHODS： This was a retrospective cohort study of 216 consecutive liver transplant patients who survived for 〉 6 mo after transplantation; 169 received a liver transplant from a living donor and 47 from a cadaveric donor. The platelet counts or spleen volumes were examined before transplant, i, 6, and 12 mo after transplant, and then annually until 5 years after transplant. RESULTS： The mean follow-up period was 49 mo （range, 21-66）. Platelet counts increased continuously for 5 years after orthotopic liver transplant. The restoration of platelet counts after transplant was significantly slower in patients with severe pretransplant thrombocytopenia （〈 50000/μL） until 4 years after transplant （P = 0.005）. Donor type did not significantlyaffect the recovery of platelet count and spleen volume in either patient group. In multivariate analysis, pretransplant severe thrombocytopenia （〈 50000/μL） was an independent factor associated with sustained thrombocytopenia （P 〈 0.001, odds ratio 6.314; confidence interval, 2.828-14.095）. Thrombocytopenia reappeared after transplant in seven patients with portal flow disturbance near the anastomosis site. CONCLUSION： Our study suggests that severe thrombocytopenia before transplant is closely associated with delayed recovery of platelet count after transplant and donor type did not affect the recovery of thrombocytopenia. The reappearance of thrombocytopenia after transplant should be considered a possible indicator of flow disturbance in the portal vein.

In recent years,pure iron(Fe)has attracted significant attention as a promising biodegradable orthopedic implant material due to its excellent mechanical and biological ***,in physiological conditions,Fe has an extremely slow degradation rate with localized and irregular degradation,which is problematic for practical *** this study,we developed a novel combination of a nanostructured surface topography and galvanic reaction to achieve uniform and accelerated degradation of an Fe *** target-ion induced plasma sputtering(TIPS)technique was applied on the Fe implant to introduce biologically compatible and electrochemically noble tantalum(Ta)onto its surface and develop surface nano-galvanic *** tests revealed that the uniformly distributed nano-galvanic corrosion cells of the TIPS-treated sample(nano Ta-Fe)led to relatively uniform and accelerated surface degradation compared to that of bare ***,the mechanical properties of nano Ta-Fe remained almost constant during a long-term in vitro immersion test(~40 weeks).Biocompatibility was also assessed on surfaces of bare Fe and nano Ta-Fe using in vitro osteoblast responses through direct and indirect contact assays and an in vivo rabbit femur medullary cavity implantation *** results revealed that nano Ta-Fe not only enhanced cell adhesion and spreading on its surface,but also exhibited no signs of cellular or tissue *** results demonstrate the immense potential of Ta-implanted surface nanostructures as an effective solution for the practical application of Fe-based orthopedic implants,ensuring long-term biosafety and clinical efficacy.

Shockwave intravascular lithotripsy and drug-coated balloon angioplasty in calcified coronary arteries:preliminary experience in two cases

Heavily calcified coronary arteries are extremely difficult to treat as calcium deposits "hardens" the arteries and alter vascular compliance. Intravascular lithotripsy (IVL)is a new technology consisting of a balloon-based system with miniaturized lithotripsy emitters that create a localised field effect to crack calcium in the arterial wall. Drug-coated balloon (DCB) angioplasty is an alternative treatment option in percutaneous coronary intervention (PCI) with an attractive strategy of leaving nothing behind combined with local drug delivery. In this brief clinical report, we described our early experience of using a novel combination of IVL and DCB angioplasty in two elderly patients with good angiographic results.

Herein,we examine the performance of dye-sensitized solar cells containing five D-π-A organic dyes designed by systematic modification of π-bridge size and geometric *** dye has a simple push-pull structure with a triarylamino group as an electron donor,bithiophene-4,4-dimethyl-4 H-cyclopenta 1,2-b:5,4-b’]dithiophene(M11),4,4-dimethyl-4 H-cyclopenta[1,2-b:5,4-b’]dithiophenethiophene(M12),thiophene-4,4-dimethyl-4 H-cyclopenta[1,2-b:5,4-b’]dithiophene(M13),4,4-dimethyl-4 H-cyclopenta[1,2-b:5,4-b’]dithiophene-benzene(M14),and 4,4-dimethyl-4 H-cyclopenta[1,2-b:5,4-b’]dithiophene(M15)units asπ-bridges,and cyanoacrylic acid as an electron acceptor/*** extension of theπ-bridge linkage favors wide-range absorption but,because of the concomitant molecular volume increase,hinders the efficient adsorption of dyes on the TiO_(2) film ***,higher loadings are achieved for smaller dye molecules,resulting in(i)a shift of the TiO_(2) conduction band edge to more negative values,(ii)a greater photocurrent,and(iii)suppressed charge recombination between the photoanode and the redox couple in the ***,under one-sun equivalent illumination(AM 1.5 G,100 mW/cm^(2)),the highest photovoltage,photocurrent,and conversion efficiency(η=7.19%)are observed for M15,which has the smallest molecular volume among M series dyes.

Influence of protein characteristics and the proportion of gluten on end-use quality in korean wheat cultivars

The effects of protein characteristics and the proportion of gluten on end-use quality in 13 korean wheat cultivars for three years were verified in this study. Year, cultivar, and the interaction between the year and the cultivar influenced protein characteristics, the proportion of gluten except for γ-and ω-gliadin using RP-HPLC(reversed-phase high-performance liquid chromatography), and end-use quality. Protein characteristics and the proportion of gluten in korean wheat cultivars were between those of Australian standard white(ASW) and hard wheat(AH). korean wheat cultivars exhibited a higher average α+β gliadin proportion than imported wheat, a γ-gliadin proportion similar to that of dark northern spring wheat, and the same ω-gliadin proportion as AH. They showed a bread-loaf volume intermediate between those of ASW and AH and a texture of cooked noodles similar to that of soft white wheat, but less springiness than imported wheat. The cookie diameter of korean wheat cultivars was similar to that of hard red winter wheat. There was a correlation between bread-loaf volume and protein characteristics, except for the protein content in korean wheat cultivars. Springiness and cohesiveness of cooked noodles were not correlated with protein characteristics, while hardness was correlated with the protein content and water absorption of a mixograph. Cookie diameter was negatively correlated with the sodium dodecyl sulfate(SDS) sedimentation volume and water absorption of a mixograph. The end-use quality was not correlated with any proportion of gluten composition. Principal component analysis(PCA) showed that the proportion of gluten was not related to the quality of the bread(both PCs, 81.3%), noodle(77.7%), and cookie(82.4%). PCA explained that Keumkang is suitable for superior bread, while Uri is good for cooked noodles and cookies.

Thermal stability and mechanical properties of thick thermal barrier coatings with vertical type cracks

The thermal stability and failure mechanism of thick thermal barrier coatings(TBCs) with and without vertical type cracks were investigated through the cyclic thermal exposure and thermal-shock tests. The TBC systems with thickness of about 2000 μm in the top coat were prepared by an air plasma spray(APS) on the bond coat of about 150 μm in thickness prepared by APS. The adhesive strength values of the as-prepared TBCs with and without vertical type cracks were determined to be 24.7 and 11.0 MPa, respectively, indicating the better interface stability in the TBC with vertical type cracks. The TBC with vertical type cracks shows a better thermal durability than that without vertical type cracks in the thermal cyclic exposure and thermal-shock tests. The hardness values of the as-prepared TBCs with and without vertical type cracks were found to be 6.6 and 5.3 GPa, respectively, which were increased to 9.5 and 5.5 GPa, respectively, after the cyclic thermal exposure tests. These results indicate that the vertical type cracks developed in the top coat are important in improving the lifetime performance of thick TBC in high temperature environment.
