Objective:Multiple myeloma(MM)remains incurable with high rates of *** therapeutic drugs are therefore urgently needed to improve the ***(JA),a natural product isolated from Inula japonica Thunb,has shown good anti-MM potential.A comprehensive study should therefore be conducted to identify both the in vitro and in vivo mechanisms of the anti-MM effects of ***:CCK8 assays and flow cytometry were used to detect the proliferation,apoptosis,and cell cycle of MM cell lines when treated with *** vivo experiments were conducted using subcutaneous xenograft mouse *** also identified possible targets and the mechanism of JA using RNA-seq and c-Map databases,and identified the specific targets of JA in bortezomib-sensitive and-resistant MM cell lines using CETSA,DARTS,and rescue ***,JA and bortezomib were used separately or together to characterize their possible synergistic ***:In vitro,JA inhibited proliferation,and induced apoptosis and G2/M phase arrest in MM cell lines,and selectively killed primary CD138+MM *** vivo,JA also demonstrated a strong anti-tumor effect with no observable *** addition,JA showed synergetic effects in combination with bortezomib,and enhanced the anti-tumor effect of bortezomib in bortezomibresistant *** and DARTS confirmed direct binding of JA to NF-κB inhibitor kinase beta(IKKβ),and overexpression of IKKβor knockdown of IκBαpartially rescued the apoptosis induced by ***:JA exhibited strong anti-tumor effects in *** sensitized myeloma cells to bortezomib and overcame NF-κB-induced drug resistance by inhibiting IKKβ,providing a new treatment strategy for MM patients.

Objective:To isolate and identify the undescribed compounds from the fruits of Cinnamomum migao and evaluate its nitric oxide inhibition ***:The chromatographic techniques of silica gel,Sephadex,and HPLC were used for isolation and purification of the compounds,while HR-ESI-MS,1D NMR,2D NMR,ECD,and X-ray diffraction techniques were used to characterize and confirm the isolated ***,the antiinflammatory activity of the isolated compounds was carried out to check inhibitory potential against the production of nitric oxide with RAW264.7 cells stimulated by ***:Camganoid A(1),a novel sesquiterpene possessing an unprecedented skeleton,and camganoid B(2),containing a unique eight-membered sesquiterpene moiety with a new carbon skeleton,were isolated and identified from the fruits of *** absolute configurations of 1 and 2 were confirmed by single crystal X-ray diffraction and electronic circular dichroism(ECD)*** these compounds,compound 1 exhibited potent inhibitory activity against the production of nitric oxide with IC50value of 4.59μmol/L in RAW264.7 cells stimulated by ***:The isolation of two new skeletons from the fruits part of *** possessed unique skeletons which have not been reported before.

Objective:This study aimed to elucidate the possible mechanism of Gest-Aid Plus Oral Liquid(GAP)on healthcare ***:Ultrahigh-performance liquid chromatography-quadrupole time-of-flight mass spectrometry-based metabolomics and 16S rDNA sequencing of gut microbiota were performed on serum and fecal samples of GAP and control ***,short-chain fatty acids(SCFAs)and inflammatory cytokines in fecal samples were determined through gas chromatography-mass spectrometry and enzyme-linked immunosorbent assay ***:Metabolomics discovered 41 metabolites,which mainly involved amino acid metabolism,lipid metabolism,coenzyme factors,and vitamin *** of GAP increased abundance of Prevotella_9,Alloprevotella,Blautia,Phascolarctobacterium,Parabacteroides,and Fusicatenibacter,and six SCFAs were increased in the GAP *** of inflammatory cytokines showed that GAP had an anti-inflammatory effect in ***:Administration of GAP greatly affects the aspartate metabolism and microecology of rats,enhances intestinal motility and gut barrier integrity and *** findings not only have possible implications for further application of GAP,but also provide a link between the gut microbiome,SCFAs,inflammation and serum metabolites in rats.

Two new phenolic amides from Allium chinense

Objective: To isolate the phenolic amides from the dried bulbs of Allium chinense and investigate their myocardium protective ***: The chemical constituents were isolated and purified by combining with silica gel column,Sephadex LH-20 column, HPLC and other chromatography techniques. Their structures were elucidated by NMR techniques and mass spectrometry. The isolated compounds were evaluated to determine their protective effect for myocardium cells in ***: Two new phenolic amides, namely, alichinemide Ⅰ(1) and alichinemide Ⅱ(2), and six konwn amides were isolated from the dried bulbs of A. chinense. The structures of compounds 3–8 were identified as 3-indolcarbaldehyde(3), 1-(2-aminophenyl)urea(4), 2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole-3-carboxylic acid(5), N-trans-feruloyltyramine(6), N-trans-p-coumaroyltyramine(7), and N-(3,4-dimethoxyphenethyl) acetamide(8). Compound 3(50 μmol/L) showed significant inhibitory effect on the damage of H9c2 myocardial cells induced by H2O2in ***: Compounds 1 and 2 were new phenolic amides. Compound 3 could be one of the potential myocardium protective constituents of A. chinense.

Fragrant rice has a high market value,and it is a popular rice type among consumers owing to its pleasant *** methods,nitrogen(N)fertilizers,and silicon(Si)fertilizers can affect the grain yield and fragrance of fragrant ***,the core commercial rice production attributes,namely the head rice yield(HRY)and 2-acetyl-1-pyrroline(2-AP)content of fragrant rice,under various nitrogen and silicon(N-Si)fertilization levels and different plantation methods remain *** field experiment in this study was performed in the early seasons of 2018 and 2019 with two popular indica fragrant rice cultivars(Yuxiangyouzhan and Xiangyaxiangzhan).They were grown under six N-Si fertilization treatments(combinations of two levels of Si fertilizer,0 kg Si ha^(−1)(Si0)and 150 kg Si ha^(−1)(Si1),and three levels of N fertilizer,0 kg N ha^(−1)(N0),150 kg N ha^(−1)(N1),and 220 kg N ha^(−1)(N2))and three plantation methods(artificial transplanting(AT),mechanical transplanting(MT),and mechanical direct-seeding(MD)).The results showed that the N-Si fertilization treatments and all the plantation methods significantly affected the HRY and 2-AP content and related parameters of the two different fragrant rice *** with the Si0N0 treatment,the N-Si fertilization treatments resulted in higher HRY and 2-AP *** rates of brown rice,milled rice,head rice,and chalky rice of the fragrant rice also improved with the N-Si fertilization *** N-Si fertilization treatments increased the activities of N metabolism enzymes and the accumulation of N and Si in various parts of the fragrant rice,and affected their antioxidant response *** key parameters for the HRY and 2-AP content were assessed by redundancy ***,the structural equation model revealed that the Si and N accumulation levels indirectly affected the HRY by affecting the N metabolism enzyme activity,N use efficiency,and grain quality of fragrant ***,high N

Metastatic pancreatic cancer(mPC)has a dismal ***,we conducted a prospective,multicentre,single-arm,phase II trial evaluating the efficacy and safety of penpulimab and anlotinib in combination with nab-paclitaxel/gemcitabine(PAAG)in patients with first-line mPC(NCT05493995).The primary endpoints included the objective response rate(ORR)and disease control rate(DCR),while secondary endpoints encompassed progression-free survival(PFS),overall survival(OS),and *** 66 patients analysed for efficacy,the best response,indicated by the ORR,was recorded at 50.0%(33/66)(95%CI,37.4–62.6%),with 33 patients achieving partial response(PR).Notably,the DCR was 95.5%(63/66,95%CI,87.3–99.1%).The median PFS(mPFS)and OS(mOS)were 8.8(95%CI,8.1–11.6),and 13.7(95%CI,12.4 to not reached)months,*** 3/4 treatment-related adverse events(TRAEs)were reported in 39.4%of patients(26/66).In prespecified exploratory analysis,patients with altered SWI/SNF complex had a poorer ***,low serum CA724 level,high T-cell recruitment,low Th17 cell recruitment,and high NK CD56dim cell scores at baseline were potential predicative biomarkers for more favourable *** conclusion,PAAG as a first-line therapy demonstrated tolerability with promising clinical efficacy for *** biomolecular findings identified in this study possess the potential to guide the precise clinical application of the triple-combo regimen.

以民为本、民贵君轻、与民同乐一直被视为孟子仁政思想的重要内容,但要注意分辨孟子在何种层面上主张民本、民贵,又在何种意义上提倡与民同乐。在孟子思想中,民心与民意存在差别:民心具有同然的理义,包含大共理,是纯善的;但民意会受到偏私意见的影响而有善有不善。政权的基础和依据应是民心,而非民意。国家不仅要通过行仁政而获得民心、滋养民心,还要警惕民意的偏私不公,因而要以善政结合善教来引导民意。厘清民心与民意的区别与联系,是理解孟子仁政思想的关键。

目的:研究水朝阳旋覆花Inula helianthus-aquatica地上部分中的倍半萜内酯类化学成分。方法:应用硅胶柱色谱,Sephadex LH-20柱色谱,以及高效液相制备色谱等方法分离和纯化化合物,再通过运用光谱学方法,结合化合物的理化性质来鉴定结构。结果:从水朝阳旋覆花的地上部分分离得到7个倍半萜内酯类和4个其他类的化合物,并且依次鉴定为2-desoxy-4-epi-pulchellin(1),6-acetoxy-4-hydroxy-1,10H-pseudoguaia-11(13)-en-12,8-olide(2),4-acetoxy-6-hydroxy-1,10H-pseud-oguaia-11(13)-en-12,8-olide(3),8-epi-inuviscolide(4),2,3,11,13-tetrahydroaromaticin(5),11,13-dihydro-ergolide(6),4-epip-ulchellin-2-O-acetate(7),7-差向黑麦草内酯(8),黑麦草内酯(9),β-谷甾醇(10),胡萝卜苷(11)。结论:所有化合物均为首次从该植物中分离得到。

[目的/意义]短视频作为新兴的互联网信息表达方式,已逐渐成为政策传播的重要工具。探索短视频平台在公共政策信息传播中的效力以及受众采纳意愿,有助于提升公共政策传播效果。[研究设计/方法]以“双减”政策为例,根据框架效应理论和信息采纳理论,通过内容分析法归纳总结短视频平台中“双减”政策信息框架的组织方式,同时分析不同政策信息框架对传播效果的影响;此外,采用在线控制实验法验证了不同政策信息框架对受众采纳意愿的影响。[结论/发现]在短视频平台中有近四成的信息使用了目标框架,其中损失框架居多,多数采用集体主义视角和非叙事陈述方式;目标框架、文化框架、叙事框架对受众的评论、转发、点赞意愿并未产生显著影响;双减政策信息中,损失、收益框架,集体、个人主义价值观以及叙事和非叙事要素都能增强受众的采纳意愿,并且目标框架与文化框架之间存在交互效应。[创新/价值]研究结论对于政策宣传部门及相关政府机构优化政策传播策略,提高传播效果具有启示作用。
