Dexmedetomidine reduces hippocampal microglia inflammatory response induced by surgical injury through inhibiting NLRP3

Purpose: To investigate whether dexmedetomidine (Dex) can reduce the production of inflammatory factor IL-1b by inhibiting the activation of NLRP3 inflammasome in hippocampal microglia, thereby alleviating the inflammatory response of the central nervous system induced by surgical injury. Methods: Exploratory laparotomy was used in experimental models in this study. Totally 48 Sprague Dawley male rats were randomly divided into 4 groups (n = 12 for each), respectively sham control (group A), laparotomy only (group B);and Dex treatment with different doses of 5 mg/kg (group D1) or 10 mg/kg (group D2). Rats in groups D1 and D2 were intraperitoneally injected with corresponding doses of Dex every 6 h. The rats were sacrificed 12 h after operation;the hippocampus tissues were isolated, and frozen sections were made. The microglia activation was estimated by immunohistochemistry. The protein expression of NLRP3, caspase-1, ASC and IL-1b were detected by immunoblotting. All data were presented as mean ± standard deviation, and independent sample t test was used to analyze the statistical difference between groups. Results: The activated microglia in the hippocampus of the rats significantly increased after laparotomy (group B vs. sham control, p < 0.01). After Dex treatment, the number was decreased in a dosedependent way (group D1 vs. D2, p < 0.05), however the activated microglia in both groups were still higher than that of sham controls (both p < 0.05). Further Western blot analysis showed that the protein expression levels of NLRP3, caspase-1, ASC and downstream cytokine IL-1b in the hippocampus from the laparotomy group were significantly higher than those of the sham control group (all p < 0.01). The elevated expression of these proteins was relieved after Dex treatment, also in a dose-dependent way (D2 vs. D1 group, p < 0.05). Synergistic Effects of Activin A and Fibroblast Growth Factor 2 in the Modulation of Insulin Expression

Diabetes is the most prevalent and serious metabolic disease, and the number of diabetic patients worldwide is increasing. The reduction of insulin biosynthes is in pancreatic E-cells is closely associated with the onset and progression of diabetes, therefore, it is important to search for ways to induce insulin-producing cells in non-E-cells. In the present study, it has been reported that activin A and a basic fibroblast growth factor 2 （ FGF2）, can synergistically increase the insulin mRNA level, in both mouse El4 striatal primary cell cultures and the hippocampal neuronal cell line HT22. Activin A and FGF2 can jointly stimulate the nuclear translocation of Smad3 and specifically activate ERK1/2. It is interesting to note that a specific inhibitor for MEK, U0126, can efficiently block the induction of an insulin promoter activity by activin A and FGF2. This indicates that activin A collaborates with FGF2, giving a signal to induce the insulin gene through selective activation of the ERK-type MAP kinase and Smad3 in mouse striatal and HT22 cells. These data suggest that activin A may act in concert with FGF2 for the development of insulin -positive neurons

电力高压试验设备运行环境往往涉及高电压、高电流等危险因素,对设备状态的检测造成了较大的困难,干扰因素较多,使得检测的结果精度不高。为了解决这一问题,提出了基于多特征融合的电力高压试验设备状态检测方法。计算电力高压试验设备的最大截流过电压,分析其对应的电压波形特征。利用小波分解去噪处理设备的超声波信号后得到相应的信号特征。分别提取设备在紫外脉冲环境下的场强特征及紫外辐射量特征,对于提取到的多种特征进行融合处理后进行电力高压试验设备的状态检测,得到了相关的检测结果。经过测试可知,该方法在电力高压试验设备的状态检测实践中表现出了较高的精度,能够满足电力高压试验设备安全运维工作的实际需求。

Exploratory COVID-19 death risk score based on basic laboratory tests and physiological clinical measurements

The efficacy of pelvic radiation in the management of locally advanced stage rectal cancer has come under scrutiny in the context of modern precision medicine and systemic therapy as evidenced by recent clinical trials such as FOWARC(J Clin Oncol 2019;37:3223-3233),NCT04165772(N Engl J Med 2022;386:2363-2376),and PROSPECT(N Engl J Med 2023;389:322-334).In this review,we comprehensively assess these pivotal trials and offer additional insights into the evolving role of pelvic radiation in contemporary oncology.

Risk factors affecting the mortality of HIVinfected patients with pulmonary tuberculosis in the cART era:a retrospective cohort study in China

Background:Tuberculosis infection still places a great burden on HIV-infected individuals in China and other developing *** of the survival of HIV-infected patients with pulmonary tuberculosis(PTB)would provide important insights for the clinical management of this population,which remains to be well described in current ***:HIV-infected patients with PTB admitted to Shanghai Public Health Clinical Center from January 2011 to December 2015 were retrospectively *** this cohort,the survival prognosis was estimated by the Kaplan-Meier method,while univariate and multivariate Cox proportional hazards models were used to determine the risk factors affecting ***:After reviewing 4914 admitted patients with HIV infection,359 PTB cases were *** the time of PTB diagnosis,the patients’median CD4+T cell count was 51/mm3(IQR:23-116),and 27.30%of patients(98/359)were on combination antiretroviral therapy(cART).For the 333 cases included in the survival analysis,the overall mortality was 15.92%(53/333)during a median 27-month *** risk factors,including age older than 60 years(HR:3.18;95%CI:1.66-6.10),complication with bacterial pneumonia(HR:2.64;95%CI:1.30-5.35),diagnosis delay(HR:2.60;95%CI:1.42-4.78),CD4+T cell count less than 50/mm3(HR:2.38;95%CI:1.27-4.43)and pulmonary atelectasis(HR:2.20;95%CI:1.05-4.60),might independently contribute to poor *** patients without cART before anti-TB treatment,the later initiation of cART(more than 8 weeks after starting anti-TB treatment)was found to increase the mortality rate(OR:4.33;95%CI:1.22-15.36),while the initiation of cART within 4-8 weeks after starting anti-TB treatment was associated with the fewest deaths(0/14).Conclusions:The subjects in this study conducted in the cART era were still characterized by depressed immunological competence and low rates of cART administration,revealing possible intervention targets for preventing TB reactivation in HIV-in

芬兰中文教育历经传统汉学、新汉学、新汉学与中文教学双轨并行发展三个阶段。双边关系、经贸合作、语言政策、华人人口数量是影响芬兰中文教育发展的重要因素。当前芬兰中文教育发展呈现教学机构数量逐年增多,学历教育层次多样化趋势。面临基础教育阶段、高中阶段教学机构体量较小、学校分布不平衡;高等教育阶段中文课程以选修课为主,中文类专业课程相对较少;华文教育以家庭教育为主,华校发展不足;本土化教学资源不足,本土师资力量薄弱等问题,提出以需求为导向,加强顶层设计;树立良好的舆论导向,培养高层次本土中文人才;发展虚拟空间中文教育;加速本土化、数字化教学资源开发;加强高校中文学科建设,建立学科辐射中心等策略。
