Role of Sphingosine 1-Phosphate Receptor Type 1 in Lymphocyte Egress from Secondary Lymphoid Tissues and Thymus

Circulation of mature lymphocytes between blood and secondary lymphoid tissues plays a central role in the immune system. Homing of lymphocytes from blood into secondary lymphoid tissues beyond high endothelial venules is highly dependent on the interaction between the chemokines CCL19, CCL21, CXCL12, and CXCL13, and their receptors CCR7, CXCR4 and CXCR5. However, the molecular mechanism（s） of lymphocyte egress from secondary lymphoid tissues to lymph remained unclear. We have found a new class of immunomodulator, FTY720 by chemical modification of vegetative wasp-derived natural product, ISP-I （myriocin）. FTY720 has been shown to be highly effective in experimental allograft and autoimmune disease models. A striking feature of FTY720 is the induction of a marked decrease in peripheral blood lymphocytes at doses that show immunomodulating activity in these models. The reduction of circulating lymphocytes by FTY720 is caused by sequestration of lymphocytes into secondary lymphoid tissues and thymus. FTY720 is rapidly converted to （S）-enantiomer of FTY720-phosphate [（S）-FTY720-P] by sphingosine kinase 2 in vivo. （S）-FTY720-P acting as a potent agonist of S1P receptor type 1 （S1P1）, induces long-term down-regulation of S1P1 on lymphocytes, and thereby inhibits the migration of lymphocytes toward S1P. Thus, it is presumed that FTY720-induced lymphocyte sequestration is due to the inhibition of S1P/S1P1-dependent lymphocyte egress from secondary lymphoid tissues and thymus by its active metabolite （S）-FTY720-P. Throughout the analysis of the mechanism of action of FTY720, it is clarified that S1P/S1P1 interaction plays an important role for lymphocyte egress from secondary lymphoid tissues and thymus.

Involvement of Sphingosine 1-Phosphate (S1P) Receptor Type 1 and Type 4 in Migratory Response of Mouse T Cells toward S1P

Sphingosine 1-phosphate （S1P）, a pleiotropic lysophospholipid, regulates signal transduction pathway via Gprotein-coupled receptors termed S1P1-5 in several types of the cells including lymphocytes. Higher levels of S1P4 mRNA as well as S1P1 mRNA are expressed in lymphoid tissues such as the spleen, thymus, lymph nodes, and Payer＇s patches. In contrast to S1P1 that plays an essential role in lymphocyte egress, little is known about the role of S1P4 in immune system. In this study, we found that S1P at 10 to 100 nM significantly induced the cell migration and the significant levels of S1P1 and S1P4 mRNA were expressed in mouse CD4 T cells, D10.G4.1 mouse Th2 cells, and ***-2 mouse thymoma cells. In D10.G4.1 and ***-2 cells, S1P-induced migration was almost completely inhibited by pretreatment with pertussis toxin, Clostoridium difficile toxin B, and （S）-enantiomer of FTY720-phosphate, a potent agonist at S1P1 and S1P4. The members of the Rho family small GTPase, Cdc42 and Rac were activated by S1P stimulation in these cells. The transfection with dominant negative or constitutively active forms of Cdc42 and Rac revealed that the activation of both Cdc42 and Rac is essential for S1P-induced migration of these cells. The immunoprecipitation assays using CHO cells co-expressing both S1P4 and S1P1 receptors indicated that S1P4 and S1P1 are associated on the cell surface. These results suggest that the association of S1P4 and S1PI plays an important role in migratory response of mouse T cells toward S1P.

Controlling Li ion transport in glasses at atomic and molecular levels is key to realizing all-solid-state batteries,a promising technology for electric *** this context,Li3PS4glass,a promising solid electrolyte candidate,exhibits dynamic coupling between the Li+cation mobility and the PS43-anion libration,which is commonly referred to as the paddlewheel *** addition,it exhibits a concerted cation diffusion effect （i.e.,a cation-cation interaction）,which is regarded as the essence of high Li ion ***,the correlation between the Li+ions within the glass structure can only be vaguely determined,due to the limited experimental information that can be ***,this study reports that the Li ions present in glasses can be classified by evaluating their valence oscillations via Bader analysis to topologically analyze the chemical *** is found that three types of Li ions are present in Li3PS4glass,and that the more mobile Li ions （i.e.,the Li3-type ions） exhibit a characteristic correlation at relatively long distances of 4.0-5.0?.Furthermore,reverse Monte Carlo simulations combined with deep learning potentials that reproduce X-ray,neutron,and electron diffraction pair distribution functions showed an increase in the number of Li3-type ions for partially crystallized glass structures with improved Li ion transport *** results show order within the disorder of the Li ion distribution in the glass by a topological analysis of their ***,considering the molecular vibrations in the glass during the evaluation of the Li ion valences is expected to lead to the development of new solid electrolytes.

Can early precut reduce post-endoscopic retrograde cholangiopancreatography pancreatitis in patients with difficult bile duct cannulation?

BACKGROUND Endoscopic retrograde cholangiopancreatography(ERCP)is associated with a variety of adverse events(AEs).One of the most important AEs is post-ERCP pancreatitis(PEP),which is most common in cases of difficult biliary *** the precut technique has been reported as a PEP risk factor,recent studies indicate that early precut could reduce PEP,and that precut itself is not a risk *** To evaluate the safety of the precut technique,especially in terms of *** We conducted a retrospective study,spanning the period from November 2011 through December *** included 1556 patients,aged≥20 years,who underwent their initial ERCP attempt for biliary disease with a naïve papilla at the Kawasaki University General Medical *** compared the PEP risk between the early precut and the delayed precut *** The PEP incidence rate did not significantly differ between the precut and nonprecut ***,the PEP incidence was significantly lower in the early precut group than the delayed precut group(3.5%vs 10.5%;P=0.02).The PEP incidence in the delayed precut group without pancreatic stent insertion(17.3%)was significantly higher compared to other cases(P<0.01).CONCLUSION Our findings indicate that early precut may reduce PEP *** the precut decision is delayed,a pancreatic stent should be inserted to prevent PEP.

Fundamentals in CO2 capture of Na2CO3 under a moist condition

Capacity and kinetics of COcapture of NaCOwere studied to determine the mechanism for COsequestration under ambient conditions. Bicarbonate formation of NaCOwas examined by a thermogravimetric analysis（TGA） under various COand water vapor concentrations and the accompanying structural changes of NaCOwere demonstrated by X-ray diffraction（XRD）. Morphological variations were observed during the reaction of COcapture through scanning electron microscope（SEM）. Structural changes and morphological variations, which occurred during the course of the reaction, were then connected to the kinetic and exothermic properties of the COcapture process from the XRD and SEM measurements. The results showed that the bicarbonate formation of NaCOhas two different *** higher COand HO concentrations, the bicarbonate formation proceeded effectively. However, for lower COand HO concentrations, the reactions were more complicated. The formation of NaCO·HO from NaCOas the first step, followed by the subsequent formation of NaH（CO）, and then the bicarbonate formation proceeds. To understand such fundamental properties in COcapture of NaCOis very important for utilization of NaCOas a sorbent for COcapture.

Power generation using small temperature difference such as ocean thermal energy conversion(OTEC)and discharged thermal energy conversion(DTEC)is expected to be the countermeasures against global warming *** ammonia and ammonia/water are used in evaporators for OTEC and DTEC as working fluids,the research of their local boiling heat transfer is important for improvement of the power generation *** of local boiling heat transfer coefficients were performed for ammonia/water mixture(z=0.9-1)on a vertical flat plate heat exchanger in a range of mass flux(7.5-15 kg/m2s),heat flux(15-23 kW/m 2),and pressure(0.7-0.9 MPa).The result shows that in the case of ammonia/water mixture,the local heat transfer coefficients increase with an increase of mass flux and composition of ammonia,and decrease with an increase of heat flux.

The AgCl/Al2O3 catalyst has potential for use in the selective catalytic reduction（SCR） of NOx.A compound hydrocarbon,following oxygenation is used as a type of reducing *** this experiment,the AgCl/Al2O3 catalyst was produced by four different methods,and the differences among their reduction catalysis of NOx were *** was used as a type of reducing agent.X-ray diffraction analysis was performed to study the crystalline structure and scanning electron microscope and transmission electron microscope（TEM） were applied to determine the *** results indicated that,in the range of 350-400℃,there was no significant difference on the NOx reduction rate;however,there was dispersion at high and low temperature *** size of the AgCl particles was about 20-100 nm.
