Cimicifuga heracleifolia Komarov(CH) is used traditionally as a herbal remedy for many ***,published literature suggests that large doses of cimicifuga racemosa might be associated with ***,its long-term toxicity was not evaluated *** study was performed to evaluate 14-day repeated dose toxicity(RDT) and genotoxicity of CH extract according to the USNTP protocol and GLP guideline. To determine the appropriate dosage of CH extract administration,14-days RDT study were performed.14-days repeated dose study was performed to dosage levels of 0.125,0.25,0.5,1 and 2g/kg/day in F344/N *** test substance-related effect were observed in some criteria included mortality;clinical signs;body weight changes;urinalysis,organ weight,gross findings and histopathological findings. In reverse mutation assay of CH extract using Salmonella typhimurium TA98,TA100,TA102,TA1535 and TA1537,test substance was diluted to 5 dosage levels of 312.5,625,1,250,2,500 and 5,000μg/plate with D.D.W..Each tester strain was exposed to the test item with and without metabolic activation *** extract might not have reverse mutagenicity under the condition of this study. Chromosome aberration test of CH extract was performed to evaluate its clastogenic potential in Chinese Hamster Lung fibroblast(CHL) cell *** test substance was diluted to 3 dosage levels of 2,500,1,250,625fig/*** were exposed to test items for 6 or 24 hours in the absence of metabolic activation system,or for 6 hours in the presence of metabolic activation *** was no statistically significant increase in the number of structural or numeric aberrant metaphases at any dose both in the presence and absence of metabolic activation. Adenomyoma is a term generally applied to nodular lesions showing proliferation of both epithelial and smooth muscle components. Despite its benign nature,ampullary adenomyoma is usually presented as biliary obstruction. Most cases are misdiagnosed as carcinoma or adenoma by preoperative endoscopic or radiologic procedure. Therefore,it is frequently treated with extensive surgery. To our knowledge,this is the first reported case in English literature of adenomyoma located in the peripancreatic orifice resulting in intermittent pancreatic duct obstruction and recurrent pancreatitis diagnosed by the endoscopic piecemeal resection.

Objective The Mediodorsal thalamus(MD) is a part of the limbic system and connects reciprocally with the prefrontal cortex,the amygdala and the hippocampus,which comprise a network of structures involved in fear memory. However,the contribution of the MD to fear behavior is poorly *** C(PLC) b4 is expressed intensely in the MD but very lightly in the prefrontal cortex and the amygdala,the two key regions known to regulate fear *** Here,we examined a role of the MD using the PLCb4 knock-out(PLCb4) mice in the formation and extinction of auditory-conditioned *** The PLCb4 mice showed normal acquisition and long-term memory of auditory-conditioned ***,the mutant was severely impaired in fear *** clamp recording on the mutant MD neurons in thalamic slices showed an increase of low threshold Ca currents mediated via T-type Ca channels compared to the wild *** infusion in vivo of T-type channel blocker into the MD ameliorated the impaired extinction phenotype in PLCb4 and facilitated the extinction in wild type *** These results suggest that the mediodorsal thalamus is involved in the control of fear extinction and T-type Ca channels play an inhibitory role in this process.

Currently, sixty-five original sprinkle drug products are available in various dosage forms including tablets, powders, granules, immediate-release capsules, extended-release capsules, delayed-release capsules, and multiparticulate drug delivery systems. By sprinkling on soft food vehicles, these products provide dosing flexibility and convenience of administration, which potentially improve the compliance of patients with *** to these advantages, the growth of sprinkle products picked up since the 1990s, and several regulatory issues regarding this dosage form have been raised and *** this article, the types of sprinkle formulations were discussed by dividing them into seven categories, and the commercial products were summarized in terms of the drug substance, pharmaceutical excipients, storage conditions and administration methods. In addition, several US Food and Drug Administration guidelines related to the regulatory issues of sprinkle formulations were reviewed, which led to the conclusion that the future development of this promising dosage form demands integrated guidance for industry rather than scattered information in various documents.

Dominant intermediate charcot-Marie-Tooth disease type C（DI-CMTC） is a dominantly inherited neuropathy that has been classified primarily based on motor conduction velocity tests but is now known to involve axonal and demyelination ***-CMTC is linked to tyrosyl-t RNA synthetase（YARS）-associated neuropathies,which are caused by E196 K and G41 R missense mutations and a single de novo deletion（153-156 del VKQV）.It is well-established that these YARS mutations induce neuronal dysfunction,morphological symptoms involving axonal degeneration,and impaired motor *** present study is the first to describe a novel mouse model of YARS-mutation-induced neuropathy involving a neuron-specific promoter with a deleted mitochondrial targeting sequence that inhibits the expression of YARS protein in the *** adenovirus vector system and in vivo techniques were utilized to express YARS fusion proteins with a Flag-tag in the spinal cord,peripheral axons,and dorsal root *** transfection of YARS-expressing viruses,the distributions of wild-type（WT） YARS and E196 K mutant proteins were compared in all expressed regions; G41 R was not *** proportion of Flag/green fluorescent protein（GFP） double-positive signaling in the E196 K mutant-type mice did not significantly differ from that of WT mice in dorsal root ganglion *** adenovirus genes,and even the empty vector without the YARS gene,exhibited GFP-positive signaling in the ventral horn of the spinal cord because GFP in an adenovirus vector is driven by a cytomegalovirus *** present study demonstrated that anatomical differences in tissue can lead to dissimilar expressions of YARS ***,use of this novel animal model will provide data regarding distributional defects between mutant and WT genes in neurons,the DICMTC phenotype,and potential treatment approaches for this disease.

我们提出了运动不耐受(motion sickness,MS),视觉诱发运动不耐受(visually induced motion sickness,VIMS),运动病(motion sickness disorder,MSD)和视觉诱发运动病(visually induced motion sickness disorder,VIMSD)的诊断标准,这些标准将被纳入国际前庭疾病分类。MS和VIMS是一种正常生理反应,几乎所有人都可能出现,但在某些情况下,个体对这种反应的易感性和严重性过高则被认为是一种疾病。本文提供了对由躯体运动或视觉运动引起的症状和体征的评价标准及个体反应严重程度是否可构成疾病诊断的指南。MS和VIMS的诊断标准包括由于暴露于躯体运动或视觉运动所引发的不良反应,通常导致在以下方面出现严重程度大于最低限度的症状或体征:恶心和/或胃肠道功能紊乱、体温调节功能紊乱、觉醒水平改变、头晕和/或眩晕、头痛和/或眼疲劳。这些症状/体征在暴露于运动状态时出现,随着暴露时间延长而加重,最终随运动结束而消失。当相同或相似的刺激反复诱发MS或VIMS发作,其严重程度在反复暴露后没有明显减轻,且因症状/体征而调整活动状态、避免参与诱发不耐受的运动刺激或负性情绪反应时,即可诊断为MSD和VIMSD。MS/MSD和VIMS/VIMSD可以单独出现,也可同时出现。由躯体运动或视觉运动刺激诱发的症状严重程度个体差异较大,并且同一个体也会因年龄、适应能力及合并疾病的变化而发生改变。本文探讨了评估MS症状的主要方法,易出现MS和VIMS相关情境以及导致易感性增加相关的个体特征。对上述环境和个人因素特征的准确评估及理解,将有助于MSD和VIMSD的诊断优化。
