Shikonin alleviates collagen-induced arthritis mice by inhibiting M1 macrophage polarization

OBJECTIVE: To investigate the effects of shikonin(SKN) on M1 and M2 polarization of macrophages both in vitro and in vivo. METHODS: Collagen-induced arthritis(CIA) in male DBA/1 mice were treated with a dose of 4 mg/kg/day of SKN for 23 d(n = 6/group). The histopathology of inflamed joints in CIA mice was evaluated to test the antiarthritic effect of SKN. M1/M2 polarization of macrophages induced by lipopolysaccharide(LPS) and interferon(IFN)-γ or interleukin(IL)-4 and IL-13, were used to assess the effect of SKN(0.05, 0.1, and 0.2 μM). The effect of SKN on the protein expression of nitric oxide synthase, arginase, CD68, and CD206 was evaluated using western blot analysis. RESULTS: The results of this study revealed that SKN delayed the arthritis feet symptom score, reduced the incidence rate of arthritis, and relieved the inflammation of joints in CIA mice. SKN inhibited M1 macrophage polarization but did not affect M2 macrophage polarization in the joints of CIA mice. Moreover, SKN inhibited M1 polarization induced by LPS and IFN-γ, but did not affect M2 polarization induced by IL-4 and IL-13. CONCLUSION: These findings suggest that SKN alleviated CIA through inhibiting M1 macrophage polarization and has great potential as a new drug for RA treatment.

Anaerobic digestion(AD)operating under conditions of organic overload stress typically exacerbates the potential for process instability,thereby resulting in significant economic and ecological ramifications.In this investigation,an augmented substrate-to-inoculum ratio(S/I)along with varying total solid content(TS)levels was employed to replicate diverse organic loadings,utilizing maize straw and cattle manure.The findings reveal that a moderate augmentation in S/I and TS proves advantageous in augmenting methane yield,while an excessive substrate loading diminishes methane yield,hampers the kinetics of methane production,and even induces severe process instability.Kinetic study also displayed the variation of the model parameters for the first-order model,the modified Gompertze model,and the transfer function model.Both the modified Gompertze model and transfer function model exhibited the same environmental stress trend.Thus,both the increase in particulate content and the increase in S/I had a substantial effect on the substrate conversion rate to methane.Microbial analysis demonstrates the dominant influence of Firmicutes and Methanosarcina under different organic loading stresses.From both a kinetic and a microbiological point of view,this work provides novel insights into the fundamental processes that regulate anaerobic digestion(AD)under varying loading stress.Furthermore,it has significant implications for improving the operating efficiency of AD,which is a significant benefit.

In this paper,to present a lightweight-developed front underrun protection device(FUPD)for heavy-duty trucks,plain weave carbon fiber reinforced plastic(CFRP)is used instead of the original high-strength steel.First,the mechanical and structural properties of plain carbon fiber composite anti-collision beams are comparatively analyzed from a multi-scale perspective.For studying the design capability of carbon fiber composite materials,we investigate the effects of TC-33 carbon fiber diameter(D),fiber yarn width(W)and height(H),and fiber yarn density(N)on the front underrun protective beam of carbon fiber compositematerials.Based on the investigation,a material-structure matching strategy suitable for the front underrun protective beam of heavy-duty trucks is proposed.Next,the composite material structure is optimized by applying size optimization and stack sequence optimization methods to obtain the higher performance carbon fiber composite front underrun protection beam of commercial vehicles.The results show that the fiber yarn height(H)has the greatest influence on the protective beam,and theH1matching scheme for the front underrun protective beamwith a carbon fiber composite structure exhibits superior performance.The proposed method achieves a weight reduction of 55.21% while still meeting regulatory requirements,which demonstrates its remarkable weight reduction effect.

Tailoring grain size can improve the strength of polycrystals by regulating the proportion of grains to grain boundaries and the interaction area.As the grain size decreases to the nanoscale,the deformation mechanism in polycrystals shifts from being primarily mediated by dislocations to deformation occurring within the grains and grain boundaries.However,the mechanism responsible for fine-grain strengthening in ferroelectric materials remains unclear,primarily due to the complex multi-field coupling effect arising from spontaneous polarization.Through molecular dynamics simulations,we investigate the strengthening mechanism of barium titanate(BaTiO3),with extremely fine-grain sizes.This material exhibits an inverse Hall–Petch relationship between grain size and strength,rooting in the inhomogeneous concentration of atomic strain and grain rotation.Furthermore,we present a theoretical model to predict the transition from the inverse Hall–Petch stage to the Hall–Petch stage based on strength variations with size,which aligns well with the simulation results.It has been found that the piezoelectric properties of the BaTiO3 are affected by polarization domain switching at various grain sizes.This study enhances our understanding of the atomic-scale mechanisms that contribute to the performance evolution of fine-grain nano-ferroelectric materials.It also provides valuable insights into the design of extremely small-scale ferroelectric components.

Background Rosemary extract(RE)has been reported to exert antioxidant property.However,the application of RE in late-phase laying hens on egg quality,intestinal barrier and microbiota,and oviductal function has not been systematically studied.This study was investigated to detect the potential effects of RE on performance,egg quality,serum parameters,intestinal heath,cecal microbiota and metabolism,and oviductal gene expressions in late-phase laying hens.A total of 21065-week-old“Jing Tint 6”laying hens were randomly allocated into five treatments with six replicates and seven birds per replicate and fed basal diet(CON)or basal diet supplemented with chlortetracycline at 50 mg/kg(CTC)or RE at 50 mg/kg(RE50),100 mg/kg(RE100),and 200 mg/kg(RE200).Results Our results showed that RE200 improved(Phe biomarkers of cecal microbiota and metabolite induced by RE 200,including Firmicutes,Eisenbergiella,Paraprevotella,Papillibacter,and butyrate,were closely associated with Haugh unit,n-6/n-3,SOD,IL-6,and TC.PICRUSt2 analysis indicated that RE altered carbohydrate and amino acid metabolism of cecal microbiota and increased butyrate synthesizing enzymes,including 3-oxoacid Co A-transferase and butyrate-acetoacetate Co A-transferase.Moreover,transcriptomic analysis revealed that RE200 improved gene expressions and functional pathways related to immunity and albumen formation in the oviductal magnum.Conclusions Dietary supplementation with 200 mg/kg RE could increase egg quality of late-p

工程实践证明软土地基加固处理后桩基水平承载力提高显著,但如何在工程前期合理预估其提高值仍需深入研究;基于此,文中参考港口工程桩基规范建立了简化p-y法模型,推导了桩侧土极限水平土抗力P_(u)=m(x+x_(0))^(n)幂函数关系曲线,建立了桩身平衡微分方程,并利用常微分方程的级数解理论建立了水平受荷桩解析解,解决了港口工程桩基规范中p-y法计算困难的问题,且便于工程师实际应用。最后通过依托工程现场试桩试验,利用新推求的桩基水平承载力计算公式,分别对软土地基真空预压处理前、后的桩基水平承载力对比分析,认为软土地基真空预压处理后桩基水平承载力由51.6 kN提高至121 kN,并通过现场桩基检测成果对比验证其合理性,为类似工程提供技术参考。

水-温循环作用下千枚岩的动态拉伸特性

为研究水-温耦合作用下0°层理倾角千枚岩的动态拉伸特性变化规律,分别对3组试样进行0、1、3、5、7、8、11次温度循环自然降温、温度循环冷水降温、干湿循环后,采用霍普金森杆试验装置对0°层理倾角千枚岩试样开展动态巴西劈裂试验,从动态拉伸应变曲线、动态峰值抗拉强度、动态弹性模量、能量分析与宏观破坏5个角度研究水、温劣化条件下千枚岩的动态拉伸特性。结果表明:千枚岩应力应变曲线包括极速弹性变形阶段、屈服变形阶段、破坏变形阶段;随着水-温循环次数的增加,千枚岩应力-应变曲线极速弹性变形阶段逐渐缩短,屈服变形阶段的应变增长率不断增大;千枚岩动态峰值抗拉强度呈负指数函数关系变化,耗散能比不断减小;水-温耦合条件下,千枚岩峰值抗拉强度、耗散能比普遍小于温度循环自然降温时;动态冲击下,千枚岩发生贯穿层理的张拉破坏,主要破碎为2块;随着水-温循环次数的增加,千枚岩主碎块发生沿层理面的张拉与穿层理面的剪切复合破坏,千枚岩碎块的平均尺寸不断减小;温度循环冷水降温条件下,千枚岩碎块的平均尺寸更小,且降幅最为显著。

人诺如病毒(Human norovirus, HuNoV)属于杯状病毒科诺如病毒属,为无包膜单股正链RNA病毒,易发生重组,变异快,传染性强,致病剂量低;人群普遍易感,是全球急性胃肠炎(Acute gastroenteritis, AGE)的主要病因之一。HuNoV感染后,≤5岁儿童和>65岁老年人群中少数人会因大量失水导致电解质紊乱等重症,甚至死亡。HuNoV还易在封闭和/或半封闭人群拥挤的场所引起AGE暴发;新的变异株或/和型别出现,会导致暴发事件大幅升高,甚至出现大流行。因此,HuNoV导致了沉重的疾病负担,是全球性的公共卫生问题。本文综述了全球HuNoV AGE的流行病学研究进展,还从宿主、病毒和环境等多个方面综述了当前影响HuNoV AGE流行特征的主要影响因素,为HuNoV AGE的防控提供支撑。
