A comparison of three methods of decellularization of pig corneas to reduce immunogenicity

·AIM: To investigate whether decellularization using different techniques can reduce immunogenicity of the cornea, and to explore the decellularized cornea as a scaffold for cultured corneal endothelial cells(CECs).Transplantation of decellularized porcine corneas increases graft transparency and survival for longer periods compared with fresh grafts.·METHODS: Six-month-old wild-type pig corneas were cut into 100-200 μm thickness, and then decellularized by three different methods: 1) 0.1% sodium dodecyl sulfate(SDS); 2) hypoxic nitrogen(N2); and 3) hypertonic NaCl. Thickness and transparency were assessed visually. Fresh and decellularized corneas were stained with hematoxylin/eosin(H&E), and for the presence of galactose-α1,3-galactose(Gal) and N-glycolylneuraminic acid(NeuGc, a nonGal antigen). Also, a human IgM/IgG binding assay was performed. Cultured porcine CECs were seeded on the surface of the decellularized cornea and examined after H&E staining.· RESULTS: All three methods of decellularization reduced the number of keratocytes in the stromal tissue by >80% while the collagen structure remained preserved. No remaining nuclei stained positive for Gal or NeuGc, and expression of these oligosaccharides on collagen was also greatly decreased compared to expression on fresh corneas. Human IgM/IgG binding to decellularized corneal tissue was considerably reduced compared to fresh corneal tissue. The cultured CECs formed a confluent monolayer on the surface of decellularized tissue.· CONCLUSION: Though incomplete, the significant reduction in the cellular component of the decellularized cornea should be associated with a significantly reduced in vivo immune response compared to fresh corneas.

Selective synthesis of carbon monoxide via formates in reverse water–gas shift reaction over alumina-supported gold catalyst

Thermal decomposition of formic acid on SiO2, CeO2 and γ-Al2O3 was studied as an elementary step of reverse water–gas shit reaction（RWGS） over supported Au catalysts. γ-Al2O3 showed the highest CO selectivity among the tested oxides in the decomposition of formic acid. Infrared spectroscopy showed the formation of four formate species on γ-Al2O3： three η~1-type and one μ~2-type species, and these formates decomposed to CO at 473 k or higher. Au-loaded γ-Al2O3 samples were prepared by a depositionprecipitation method and used as catalysts for RWGS. The supported Au catalyst gave CO with high selectivity over 99% from CO2 and H2, which is attributed to the formation of formates on Au and subsequent decomposition to CO on γ-Al2O3.

A lensless Vanderlugt optical correlator using two phase-only spatial light modulators （SLMs） is proposed. The SLMs are used for displaying input and filter patterns respectively. The SLMs are also used as programmable lenses in order to realize the lensless construction. This lensless system is simple and its alignment adjustment is easy. The performance of the SLMs as programmable lenses is also described.

We present and analyze an unsupervised method for Word Sense Disambiguation（WSD）.Our work is based on the method presented by McCarthy et *** 2004 for finding the predominant sense of each word in the entire *** maximization algorithm allows weighted terms（similar words） from a distributional thesaurus to accumulate a score for each ambiguous word sense,i.e.,the sense with the highest score is chosen based on votes from a weighted list of terms related to the ambiguous *** list is obtained using the distributional similarity method proposed by Lin Dekang to obtain a *** the method of McCarthy et al.,every occurrence of the ambiguous word uses the same thesaurus,regardless of the context where the ambiguous word *** method accounts for the context of a word when determining the sense of an ambiguous word by building the list of distributed similar words based on the syntactic context of the ambiguous *** obtain a top precision of 77.54%of accuracy versus 67.10%of the original method tested on *** also analyze the effect of the number of weighted terms in the tasks of finding the Most Precuent Sense（MFS） and WSD,and experiment with several corpora for building the Word Space Model.

The glucuronide metabolites of benproperine were synthesized from mono-hydroxylate metabolites of benproperine that were treated with methyl （2,3,4-tri-O-acetyl-1-O-tfichloroacetimidoyl ） -α-D-glucopyranuronate with BF3 · Eh O as the promoter followed by basic hydrolyzation with Na2 CO3. The form of basic acceptors, the order of addition, and the promoter are all important variables in this glucuronidation. The salt form of the basic acceptor was found to be better than its free form for glucuronidation with a Lewis acid as the promoter. Two mono-hydroxylated benproperines were synthesized from 2-benzylphenol in three steps.

Chronic heat stress induces renal fibrosis and mitochondrial dysfunction in laying hens

Background Heat stress in laying hens negatively affects egg production and shell quality by disrupting the homeo-stasis of plasma calcium and phosphorus *** the kidney plays an important role in calcium and phos-phorus homeostasis,evidence regarding the effect of heat stress on renal injury in laying hens is yet to be ***,the aim of this study was to evaluate the effects of chronic heat stress on renal damage in hens during laying *** A total of 16 white-leghorn laying hens(32 weeks old)were randomly assigned to two groups(n=8).One group was exposed to chronic heat stress(33°C for 4 weeks),whereas the other group was maintained at 24°*** Chronic heat exposure significantly increased plasma creatinine and decreased plasma albumin levels(Pk mtDNAs into the cytosol and mtDNA leakage may activate the cyclic GMP-AMP synthase(cGAS)stimulator of interferon genes(STING)signaling *** results showed that chronic heat exposure activated the cGAS-STING pathway as indicated by increased expression of MDA5,STING,IRF7,MAVS,and NF-κB ***,the expression of pro-inflammatory cytokines(IL-12)and chemokines(CCL4 and CCL20)was upregulated in heat-stressed *** These results suggest that chronic heat exposure induces renal fibrosis and mitochondrial damage in laying *** damage by heat stress may activate the mtDNA-cGAS-STING signaling and cause subse-quent inflammation,which contributes to the progression of renal fibrosis and

Securing dilated lymphatic vessels with good function is challenging when performing lymphaticovenous anastomosis(LVA).To achieve this,we propose multi-point indocyanine green(ICG)lymphography and lymphatic ultrasound(D-CUPS;Doppler,Cross,Uncollapsible,Parallel,and Superficial fascia).With multi-point ICG lymphography,more lymphatic vessels can be found than with conventional ICG lymphography,which leads to better surgical *** ultrasound is more useful because it allows the observation of cross-sections of lymphatic *** is known that lymphatic degeneration occurs in the lymphatic vessels in lymphedematous limbs,and LVA is most effective when dilated lymphatic vessels are *** degree of lymphatic degeneration can be diagnosed with lymphatic ultrasound,and the proximity of dilated lymphatic vessels and veins suitable for anastomosis can be reliably identified and selected as the skin incision site for *** ultrasound is a safe,versatile and useful imaging technique that does not require a contrast agent and can be performed by *** mastering multi-point ICG lymphography and lymphatic ultrasound(D-CUPS),the operation time can be shortened,and more effective LVA can be *** this Technical Note article,we comprehensively describe lymphatic function examinations that we have developed so far.
